Objective: The aim of this study was to analyze quantitative and qualitative methods of intrathecal immunoproduction evaluation. Methods: We retrospectively assessed data from cerebrospinal fluid (CSF) and serum samples sent to Senne Liquor Diagnóstico from 2001 to 2017. Cytological, biochemical, and immunological data were compared between cases with and without oligoclonal bands (OCBs). Comparisons between samples with OCBs (OCB+) and without them (OCB-) were carried out, and the ability to predict the presence of OCBs was assessed with ROC analysis. Results: We included 8,947 samples (2,599 OCB+ samples and 6,348 OCBsamples). CSF lymphocytes and monocytes were significantly associated with the presence of OCB (p < 0.001). All the inflammatory parameters were significantly associated with OCB, and the methods with the highest predictive ability for OCB were IgG index and Reiber diagram [area under the receiver operating characteristic (AUROC) curve = 0.881 and 0.863, respectively]. A small percentage of cases without OCB had high IgG index (9.56%) or Reiber diagram, showing increased IgG production (4.6%). Conclusion: We showed a strong association between OCB and other CSF inflammatory parameters. The presence of cases with increased quantitative CSF IgG synthesis without OCB suggests that both quantitative and qualitative methods should be performed in the evaluation of neuroinflammatory processes.","PeriodicalId":35397,"journal":{"name":"Jornal Brasileiro de Patologia e Medicina Laboratorial","volume":null,"pages":null},"PeriodicalIF":0.0,"publicationDate":"2020-01-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"71115918","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Methods: Fascin expression in nasal tissues of 11 CRSwNP patients and 10 CRSsNP patients was immunohistochemically evaluated and compared with control subjects. Results: Fascin was found to be strongly expressed in epithelial cells in polyps in CRSwNP and nasal tissue in CRSsNP. Its strong expression was observed both in lamina propria and nasal epithelial cells in CRSsNP. Fascin overexpression in nasal mucosa in CRSwNP was more pronounced compared with CRSsNP. In addition, proliferating epithelial cells in polyp tissue were weakly immunostained, whereas mature cells expressed much more fascin. Conclusion: CRSwNP and CRSsNP are associated with fascin overexpression, which makes fascin a promising target for therapeutic interventions.","PeriodicalId":35397,"journal":{"name":"Jornal Brasileiro de Patologia e Medicina Laboratorial","volume":null,"pages":null},"PeriodicalIF":0.0,"publicationDate":"2020-01-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"71116297","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Material and method: This is an observational, cross-sectional study using the database of a national clinical laboratory on PSA requests from October 1, 1997 until December 31, 2016. Descriptive statistics were used. The work was approved by the Ethics Committee of our institution. Results: 2,514,375 PSA requests were evaluated, 158,399 (6.3%) in men younger than 40 years old. These percentages did not vary significantly when observed over time (1998-2016). The prevalence of requests for patients under 40 years of age, was 18.2% among general practitioners; 16.5% among cardiologists, 8.4% among geriatricians and 6.8% among urologists. Conclusion: There is a very high frequency of PSA requests in men with low probability of benefiting from the test. It is necessary to invest in the dissemination of best practices related to prostate cancer screening, especially among clinicians and cardiologists.","PeriodicalId":35397,"journal":{"name":"Jornal Brasileiro de Patologia e Medicina Laboratorial","volume":null,"pages":null},"PeriodicalIF":0.0,"publicationDate":"2020-01-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"71116438","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Objectives: Determine the prevalence of unnecessary OGTT requests and analyze patients’ suitability criteria for glucose load. Method: Cross-sectional, descriptive and analytical study conducted with 554 patients who had OGTT requested from January to April 2018. Data from laboratory tests were collected through Complab Advanced version 6.9.6 system, organized into a Microsoft Excel table and analyzed using Epi Info, version 7.2.1.0. The accuracy of the glucometer paired analysis was performed by determinations of Student’s t test, using SPSS version of IBM 21. Patients with a previous diagnosis of DM and/or who showed OGTT request as a screening test along with blood glucose or glycated hemoglobin were classified as unnecessary requests. Results: Among the studied patients, 17% (94) had unnecessary OGTT requests, 53 (53.4%) patients had blood glucose ≥ 140 mg/dl and previous DM and/or OGTT used as screening; 41 (43.6 %) patients with capillary blood glucose < 140 mg/dl, but with a diagnosis of DM. The glucometer proved to be accurate with a high correlation (r = 0.97, p < 0.0001) with serum glucose. Approaches during screening and capillary blood glucose prevented unnecessary exposure to glucose overload in 67% (63) of the patients. Conclusion: The high prevalence of unnecessary OGTT requests underscores the need of criteria for OGTT requesting and the standardization of procedures for screening in the exam.","PeriodicalId":35397,"journal":{"name":"Jornal Brasileiro de Patologia e Medicina Laboratorial","volume":null,"pages":null},"PeriodicalIF":0.0,"publicationDate":"2020-01-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"71116529","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Objective: To evaluate the performance of five rapid test kits for troponin I detection in serum. Materials and methods: Samples from 100 patients with suspected acute myocardial infarction (AMI) were selected from a hospital in Barbacena, MG, Brazil. They were tested in the five troponin I commercial rapid test kits and the results were compared with the study reference method (quantitative chemiluminescent immunoassay). Sensitivity, specificity, positive predictive value (PPV), negative predictive value (NPV) and correlation coefficients of the rapid tests against the reference method were defined. Results: The kits from Abon ® , Interteck ® , Medtest ® and Wama ® presented the same performance, reaching equal levels of sensitivity (98.08%), specificity (100%), PPV (100%), NPV (97.96%), and correlation coefficient (0.99). The Eco Diagnostica ® kit presented lower sensitivity (82.69%), low correlation coefficient (0.91) and NPV (84.21%), not reaching ideal levels even though lying within the confidence interval. Discussion: The results obtained with the Abon ® , Interteck ® , Medtest ® and Wama ® test kits corroborate pre-existing literature data on the diagnostic accuracy of rapid tests for troponin detection. These tests are allowed for immediate decision making by physicians and can be used to reduce unnecessary hospitalization time and costs associated with a suspected AMI. Conclusion: The kits from Abon ® , Interteck ® , Medtest ® and Wama ® showed excellent diagnostic performance, what makes them an important diagnostic tool in smaller laboratories without automated equipment.","PeriodicalId":35397,"journal":{"name":"Jornal Brasileiro de Patologia e Medicina Laboratorial","volume":null,"pages":null},"PeriodicalIF":0.0,"publicationDate":"2020-01-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"71116549","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Estrogen and progesterone are the main regulators of endometrial tissue, and the expression of receptors of these hormones in the ectopic tissue seems to be related to the pathophysiology of the disease. Ki-67 is a marker of tissue proliferation and an important marker of epithelial kinetics. Endometriosis can be classified as superficial, in the peritoneum, and deep, when it extends into ligaments and other organs. Objective: Our objective was to analyze the expression of estrogen and progesterone receptors and Ki-67, through immunohistochemistry, in different sites of endometriosis tissues (superficial peritoneal/ovarian endometriosis and deep infiltrating endometriosis). Casuistic and methods: We studied nine patients; five with superficial and four with deep endometriosis. Statistical correlation was performed with the Shapiro-Wilk test (significance level of 5%) and linear correlation analysis using Spearman’s non-parametric test (significance of 1%). There was a correlation of Spearman between the estrogen receptor variable and Ki-67 in patients with superficial endometriosis. There was also a correlation between the variables estrogen receptor and progesterone receptor in patients with deep endometriosis. Results: Contrary to what was found for superficial endometriosis, there is linear increase of the variables, with a strong and positive correlation coefficient. This demonstrates that the variation of estrogen receptors can be explained in 99.1% by the same variation of progesterone receptors in deep endometriosis. Conclusion: It is possible to infer that other factors are involved in the response to hormonal variations for superficial and deep endometriosi s.","PeriodicalId":35397,"journal":{"name":"Jornal Brasileiro de Patologia e Medicina Laboratorial","volume":null,"pages":null},"PeriodicalIF":0.0,"publicationDate":"2020-01-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"71116173","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

HLA-G has been associated with the process of tumorigenesis and tumor escape. In this study, we aim to evaluate the HLA-G expression in melanocytic lesions and in melanoma for determining when melanocytic lesions start its expression. Twenty-two skin biopsies samples were submitted to immunohistochemistry; HLA-G expression was detected in 63.6% of the samples. This expression in melanocytic cells was significantly higher in melanoma than in benign melanocytic lesions ( p < 0.002).
