{"title":"Antimicrobial activity of obioactin","authors":"Humio Osaki ,&nbsp;Masato Furuya ,&nbsp;Hideyuki Nagasawa ,&nbsp;Naoyoshi Suzuki","doi":"10.1016/S0174-3031(84)80010-4","DOIUrl":"10.1016/S0174-3031(84)80010-4","url":null,"abstract":"<div><p>A newly developed enzyme-hydrolyzed toxoplasma-hyperimmune bovine serum preparation, Obioactin, was examined for its anti-microbial and related actions against heterologous protozoa, bacteria, and viruses both <em>in-vivo</em> and <em>in-vitro</em>.</p><p>An appreciable efficacy of Obioactin in a combination with nifurtimox, Lampit<sup>®</sup> was observed in experimental <em>Trypanosoma cruzi</em> infections in mice. But prophylactic applications of the agent alone or joint with Lampit were in vain in mice.</p><p><em>In-vitro</em> digestion of certain bacteria in mouse peritoneal macrophages was accelerated in the presence of the agent being three to ten times stronger than that in untreated macrophages.</p><p>Inhibitory effect of Obioactin on <em>in-vitro</em> infectivity of viruses was suggested.</p></div>","PeriodicalId":79282,"journal":{"name":"Zentralblatt fur Bakteriologie, Mikrobiologie und Hygiene. 1. When the peripheral blood of chickens containing mature gametocytes was withdrawn or ingested by <em>Culicoides arakawae</em>, differentiation of the microgametocytes began immediately. The cytoplasm of host cell and the outer membrane of the microgametocyte disappeared. Axoneme formation began in the cytoplasm and the condensed chromatin masses were observed at the periphery of the nucleus of microgametocyte after disappearance of the nuclear membrane. The axonemes and chromatin masses migrated to the surface of the microgametocyte and participated in the formation of flagellar buds. The flagellar buds passed through interruptions of the inner membrane and were surrounded by the outer membrane of the microgametocyte. The microgametocyte formed eight slender microgametes. The microgamete was a long undulated thread-like structure and composed of a single axoneme and a nucleus. The nucleus was located at the mid to anterior region of the microgamete. The axoneme consisted of nine peripheral doublets arranged around two central microtubules. Penetration of the macrogamete by the microgamete occurred rapidly after ingestion. While <em>S. singaporensis</em> and <em>S. villivillosi</em> developed only in murids closely related to the genus <em>Rattus</em>, a third <em>Sarcocystis</em> species (natural definitive host: <em>Bitis nasicornis</em> — syn. <em>Isospora dirumpens?</em>) showed a broad intermediate host range. Two agglutination types can be distinguished: Typ 1 includes stocks which react with <em>Triticum vulgaris</em> and <em>Aaptos papillota</em> II (WGA-type). Type 2 includes stocks agglutinated by <em>Arachis hypogaea</em> and <em>Aaptos papillata</em> II (PNA-type). The reactions with the lectins from <em>Triticum vulgaris</em> and <em>Aaptos papillata</em> II can be correlated with the presence of N-Acetylneuraminic acid on the cell surface of these flagellates. Raether","doi":"10.1016/S0174-3031(84)80007-4","DOIUrl":"10.1016/S0174-3031(84)80007-4","url":null,"abstract":"<div><p><em>In vitro</em> exposure of <em>Plasmodium falciparum</em> and <em>P. berghei</em> to salinomycin-Na showed that 10 min incubation in RPMI 1640 medium containing 100μg/ml of the polyether antibiotic led to complete destruction of most parasites; in media containing 10 or 1 μg/ml salinomycin-Na some young developmental stages seemed to survive, apparently due to the protection of the mostly intact host cell. <em>In vitro</em> treatment of rats infected with <em>P. berghei</em> revealed that a single subcutaneous (oral) dose of 20 mg/kg (80 mg/kg) salinomycin-Na caused complete destruction of parasites. Incipient degeneration of the parasites could be already observed 1 h after treatment. After 22 h parasites had disappeared from blood smears. Repeated subcutaneous doses of the polyether as low as 5 mg/kg (×3) completely destroyed the asexual stages of <em>P. berghei</em>. During the ultrastructural investigation of the action of salinomycin-Na it was found that initially the inner lacunes (such as endoplasmic reticulum and perinuclear space) and the mitochondrion were markedly swollen. This was followed by mitochondrial disruption with rupture of the parasites’ pellicle. Since the infected host cells also ruptured, mainly extracellular parasites were seen in blood smears beginning 6 h after treatment. The dosis curativa minima/dosis tolerata maxima values against the drug-sensitive <em>P. berghei</em> strain ascertained in the ‘28-day test’ in mice, were 1.56 (× 5) /400(× 1) mg/kg after the oral route and 3.12(× 5)/400(× 1) mg/kg after the subcutaneous (sc) route. Morphological changes in erythrocytic stages of <em>P. berghei</em> following a single oral/sc dose of 25 and 50 mg/kg respectively in rats were at first swollen lacunes of the endoplasmic reticulum and extremely enlarged mitochondrion (6 h after treatment), then an apparent vacuolisation of the cytoplasm and pyknosis of the nucleus, as well as distinctly enlarged perinuclear space and later marked fissuring of the cytoplasm. After 23 h most of the parasites were destroyed by disruption of their pellicle. The majority of the degenerate parasites were situated outside of the apparently ruptured host cell. No organism was found being adhere to type II cells which have numerous microvilli on their surface. By adding tannic acid to the fixative we could preserve the alveolar lining layer, in which the organisms were found all the time. Therefore <em>P. carinii</em> is not exposed to alveolar air but is living in the liquid layer. Those findings are important in considering nutrition of the organism as well as pathogenicity to the host.</p><p>The trophozoites and type I cells were in close contact mostly with their smooth surface rather than with tubular expansions of the organism. The cytoplasm of type I cell was often found protruded along the pellicle of the organism as if it surrounded the organism. Phagocytosis of <em>P. carinii</em> by alveolar macrophage and neutrophil was often seen in the alveolar lumen when the infection advanced.</p><p>Sometimes the trophozoites were found sub- and intra-epithelially. Those trophozoites seemed alive from their ultrastructures.
