A novel micromanipulator-aided technique was developed for selecting small exact numbers of specified fresh or old cysts/pseudocysts from a faecal homogenate. This technique can also be of value in manipulating protozoan parasites of any particular species.</p><p>Only fresh cysts/pseudocysts, which appeared bright under phase contrast optics, were infectious. Older cysts and pseudocysts which appeared dark in phase contrast were not infectious. The MID for three murine intestinal flagellates addressed above were: 10 (between 5 and 20), 1 and 5 cysts/pseudocysts respectively. Corresponding prepatent periods (PP) were 8, 5 and 10 days. A relation was evident between the infectious dose and the PP: The prepatent period shortened with an increased infectious dose.</p></div><div><p>Die minimale infektiöse Dosis (MID) von <em>Giardia muris</em>-Zysten, <em>Spironucleus muris</em>-Zysten und <em>Tritrichomonas muris</em>-Pseudozysten für thymus-aplastische Mäuse wurde bestimmt. Zur Isolierung kleiner, exakter Mengen von definierten (frischen oder alten) Zysten oder Pseudozysten aus dem Kothomogenat wurde ein Mikromanipulator eingesetzt. Diese neu entwickelte Methode kann beim Manipulieren mit protozoären Parasiten beliebiger Art nützlich sein.</p><p>Nur frische, im Phasenkontrast hell erscheinende Zysten (und Pseudozysten) sind infektiös. Ältere Zysten und Pseudozysten, die im Phasenkontrast dunkel erscheinen, sind nicht infektiös. Die MID für die drei oben genannten murinen Darmflagellaten sind: 10 (zwischen 5 und 20), 1 und 5 Zysten, bzw. Pseudozysten. Die entsprechenden Präpatenzperioden (PP) waren 8, 5 und 10 Tage. The bacteria released coagulase continuously in the medium containing serum albumin, but only at the early-mid log phase of growth in the medium not containing serum albumin. In each of the media, coagulase was considered to be a newly synthesized one encoded by a newly synthesized messenger RNA. By the experiments using bacteria pre-treated with acridine orange, it was suggested that the coagulase gene was located on the chromosome. The plasmid regulating coagulase production was considered to be present.</p></div><div><p>Die Erzeugung und Freisetzung von Koagulase wurden an <em>Staphylococcus aureus</em> Smith unter Verwendung von Rinderserumalbumin enthaltendem TC Medium 199 untersucht. In serumalbuminhaltigem Medium setzten die Bakterien ständig Koagulase frei, in dem albuminfreien Medium nur zu Beginn der mittleren logarithmischen Wachstumsphase. In beiden Medien wurde die Koagulase als neugebildet und durch neugebildete Boten-RNS kodiert angesehen. Die Versuche unter Verwendung der mit Acridinorange vorbehandelten Bakterien deuteten darauf hin, daß das Koagulasegen auf dem Chromosom lokalisiert war. {"title":"Induction of proteinases in legionella by growth on agar containing serum","authors":"Hans E. However, if the strains grow on agar plates containing serum the proteolysis is very strong. This effect may be due to an induction of the appropriate enzymes by their substrates. The alterations of 24 investigated proteins demonstrated by immunoelectrophoresis show anodic shifts of bands and their disappearance (Table 1, Fig. 1–15). The shifted proteins are not degraded significantly. Their altered electrophoretic mobility may be due to a change of electrical charge by loss of one or a few end standing amino acids. The disappearance of bands in the immunoelectrophoretic assay and the simultaneous diminution of the protein concentration measured by the method of immunodiffusion suggests a proteolysis (Table 2). No inhibitory effect of the proteinase inhibitor aprotinin could be observed. The results suggest that the extent of the enzyme induction may be decisive for the virulence of a strain.</p></div><div><p>Während nicht-proliferierende Zellen von <em>Legionella pneumophila</em> nur wenige menschliche Serumproteine proteolytisch verändern, greifen proliferierende Keime alle untersuchten Serumproteine an, verändern ihre immunelektrophoretische Beweglichkeit und bauen sie mehr oder weniger stark ab. Die proteolytische Aktivität ist bei <em>L. pneumophila</em> größer als bei anderen <em>Legionella</em>-Arten und innerhalb der Art <em>L. pneumophila</em> bei Serogruppe 1 am deutlichsten ausgeprägt. Eine Hemmwirkung durch den Protease-Hemmer Aprotinin konnte nicht beobachtet werden.</p><p>Das Phänomen kann als Enzyminduktion durch das Substrat interpretiert werden. {"title":"Cooperative effects in the process of elution of newcastle disease virus pre-attached on chick embryo cell monolayer culture","authors":"Michael Lipkind , Victor Urbakh","doi":"10.1016/S0174-3031(83)80061-4","DOIUrl":"10.1016/S0174-3031(83)80061-4","url":null,"abstract":"<div><p>After the virus attachment to the cell surface at 4°C followed by incubation of the virusloaded cells at 37°C two processes are going simultaneously: one portion of the attached virions penetrates inside the cells and another portion elutes outside the cells (a certain portion during a certain time period remains bound to the cell surface). Quantities of both penetrated and eluted viruses depended on the quantitiy of the attached virus. The dependence of the quantity of the penetrated virus on the quantity of the attached virus was linear, while there was an accelerating increase of the quantity of the eluted virus depending on the quantity of the attached virus. This non-linearity is described by a mathematical model based on cooperativity effects which are caused by a certain interaction between the attached virions. Since, according to the calculations, this interaction is effective at the distance equal to about 15-fold virion diameter, a kind of “at distance” interaction between the adjacent virions realized through spreading modifications of plasma membrane is suggested.</p></div><div><p>Nach Virusanheftung an die Zelloberfläche bei 4°C mit nachfolgender Inkubation der virusbeladenen Zellen bei 37°C laufen zwei Prozesse gleichzeitig ab: ein Teil der angehefteten Virionen dringt in die Zellen ein, und ein anderer wird außerhalb der Zellen eluiert (ein bestimmter Teil bleibt für eine gewisse Zeit an der Zelloberfläche gebunden). Strains of the phenotype Bac<sup>+</sup> Tox<sup>+</sup> had a positive result when the simultaneous method was used, and produced a wide inhibition of the indicator strains, especially of <em>C. renale</em>, if the deferred method was used. The strains of the phenotype Bac<sup>−</sup> Tox<sup>+</sup> were negative upon examination by the simultaneous method, but they caused narrow inhibition zones of the indicator strains to appear with the deferred method, especially with <em>C. renale. S. aureus</em> strains of diverse origin used as controls were in both methods negative with regard to inhibition of indicator strains. <em>C. renale</em> was found to be very suitable for the detection of bacteriocins of the exfoliatin-positive staphylococci, particularly when using the deferred method.</p></div><div><p>Exfoliatin-positive <em>S. aureus</em>-Stämme bekannter Phänotypen wurden unter Verwendung von S. aureus 209P, <em>C. pseudodiphtheriticum</em> oder <em>C. renale</em> als Indikatorstämmen auf Bakteriocinbildung untersucht. Die Stämme des Phänotyps Bac<sup>+</sup> Tox<sup>+</sup> boten ein positives Ergebnis unter Anwendung der simultanen Methode und führten bei der Methode der Vorbebrütung zu einer breiten Hemmzone des Indikatorstammes, insbesondere des <em>C. renale</em>. Die Stämme des Phänotyps Bac<sup>−</sup> Tox<sup>+</sup> waren bei Untersuchung mittels der simultanen Methode negativ. Sie bewirkten jedoch bei der Vorbebrütung schmale Hemmzonen der Indikatorstämme, insbesondere mit <em>C. renale</em>. For <em>Enterobacteriaceae</em>, the MIC's of temocillin are distinctly higher than that of cefotaxime and moxalactam. But temocillin activity was not affected by resistance against ureidopenicillins and/or cefacedone. No resistant strains were encountered among <em>E. coli, K. pneumoniae, Proteus</em> spp. except one strain of <em>P. vulgaris</em>. In <em>E. cloacae</em>- strains with resistance against cefotaxime, temocillin was more active than moxalactam. <em>S. marcescens</em>- strains with a high MIC of temocillin were sensitive to cefotaxime and moxalactam. Temocillin is of the same order of activity as ampicillin against ampicillin-sensitive strains of <em>H. influenzae</em> and <em>H. parainfluenzae</em>. The enzyme-linked immunosorbent assay was applied to the sera of dogs for the detection of the specific IgA antibodies and at the same time, autopsy and stool examination were performed on these dogs.</p><p>The most elevated IgA antibody levels were observed in the one-month age group of puppies. Thereafter, the levels gradually decreased until six and 10 months of age. These low levels were maintained in adult dogs. However, the IgA antibody activities were not related to the age and the number of worms when the correlation coefficients were calculated. Traub , Marlene Spohr, Dierk Bauer","doi":"10.1016/S0174-3031(83)80068-7","DOIUrl":"10.1016/S0174-3031(83)80068-7","url":null,"abstract":"<div><p>The multiple-drug-resistant <em>Serratia marcescens</em> isolate SH 186 (serotype 06/014:H12, bacteriocin type 18) carried a 44 megadalton, nonconjugative resistance (R-) plasmid as demonstrated with ‘curing’ experiments and the DNA agarose gel electrophoresis technique. ‘Cured’ variants, which had lost part of or the entire R-plasmid, proved as virulent for outbred NMRI mice (intraperitoneal route) as the wild-type parent strain. Therefore, the virulence of this 5. marcescens strain was plasmid-independent. Protease-deficient variants of this strain as well as protease-negative variants of two additional <em>S. marcescens</em> strains displayed comparable murine virulence. None of 19 representative <em>S. marcescens</em> strains, including isolate SH 186, gave rise to guinea pig keratoconjunctivitis (negative Anton-Sereny tests), i. e., were non-invasive; NMRI mice pretreated with either cyclophosphamide (leukopenia), type II carrageenan (blockade of macrophages) or with zymosan (depletion of complement) revealed essentially unaltered susceptibility to <em>S. marcescens</em>. However, mice pretreated with cyclophosphamide followed by zymosan were significantly more susceptible for 4 tests strains of <em>S. marcescens</em>, including isolate SH 186. Postupa","doi":"10.1016/S0174-3031(83)80057-2","DOIUrl":"10.1016/S0174-3031(83)80057-2","url":null,"abstract":"<div><p>Seventy-one of 73 <em>E. sakazakii</em> strains isolated in Czechoslovakia produced Tween 80 esterase; its production was slower and less intensive than in <em>Serratia</em>, vibrios and aeromonads. The <em>E. sakazakii</em> strains did not produce lecithinase (yolk reaction was negative for 8 days). The following set of tests was recommended for verification of <em>E. sakazakii</em> identification: pigment (was produced by 100% of strains), Tween 80 esterase (was positive in 97.3%), mucate (negative in 100%) and sorbitol (negative in 100%).</p></div><div><p>71 von 73 in der Tschechoslowakei isolierten <em>Enterobacter sakazakii</em>-Stämmen bildeten Tween 80-Esterase. Die Bildung verlief langsamer und war weniger intensiv als bei <em>Serratia</em>, Vibrionen und Aeromonaden. Die <em>E. sakazakii</em>-Stämme bildeten keine Lecithinase (8 Tage lang negative Eidotterreaktion). Zur Bestätigung von <em>E. sakazakii</em>-Isolierungen werden folgende Testungen empfohlen: Pigmentbildung (positiv bei 100% aller Stämme), Tween 80-Esterase (positiv bei 97,3%), Mucat (negativ bei 100%) und Sorbit (negativ bei 100%).</p></div>","PeriodicalId":79282,"journal":{"name":"Zentralblatt fur Bakteriologie, Mikrobiologie und Hygiene. 1.
