This microfluorometric assay clearly revealed that the relative fluorescence intensity (RFI) of K-DNA and N-DNA was greatly reduced by bleomycin, suggesting the occurrence of the single strand breaks in these DNAs. The present results have shown that bleomycin acts on N-DNA stronger than K-DNA at least in <em>Trypanosoma gambiense</em>. {"title":"Bacteriologic and epidemiologic studies on the occurrence of Listeria monocytogenes in healthy cattles","authors":"Ernesto Hofer","doi":"10.1016/S0174-3031(83)80067-5","DOIUrl":"10.1016/S0174-3031(83)80067-5","url":null,"abstract":"<div><p>The incidence of <em>Listeria monocytogenes</em> in 61 fresh stool specimens from normal beef cattle obtained from a slaughterhouse in Rio de Janeiro was studied. The animals were divided in two groups of similar number separated by age; animals of up to five years, and animals of 5 years and more. In the research of this microorganism 9 culture media were used: four enrichment media, one for transport and four selective media. From the results, 11 animals proved to be carriers, of which nine were five years of age or older and two were of the younger group. Two hundred and twenty-one samples of <em>Listeria</em> were obtained from these 11 animals. The combination of Tryptose broth containing 40 mcg/ml nalidixic acid and 10 mcg/ml trypaflavine, incubated for 5 days at ambient temperature (±28°C), with the second modification of Ralovich medium, showed the best results for detection of <em>Listeria</em> both quantitatively and qualitatively. Existence of intracellular lipase has not been yet established. In this investigation, cellular content liberated by lysostaphin digestion of cell wall revealed presence of lipolytic activity. This has been observed with living staphylococcal cells only. Presence of intracellular lipase was not observed in young bacterial cells, and the enzyme was appearing rapidly in the late phase of the culture. Highest level of enzyme was observed on the 3rd day of culture in described conditions. The isolates tested were highly susceptible to tetracycline, the aminoglycosides, chloramphenicol, nitrofurantoin, and pipemic acid. They were resistant to cephalosporins (except Cefotaxime) and penicillin and its derivatives. About 6% of the isolates were resistant to erythromycin (MIC 4 μg/ml) and about 15% to nalidixic acid (MIC 40 (μg/ml). Hahn","doi":"10.1016/S0174-3031(83)80062-6","DOIUrl":"10.1016/S0174-3031(83)80062-6","url":null,"abstract":"<div><p>Intoxications by <em>S. aureus</em> enterotoxins are predominant in our country, together with enteric infections by <em>Salmonella</em> spp. For lack of a specific therapy the possibility was examined to neutralize orally provided toxin in vivo. For this purpose before and after the intoxication, respectively, antitoxin and normal rabbit serum as a control were also applied per os. In extensive trials SPF-mice were used as a model in spite of their insensitivity to staphylococcal toxins even with 10000 times the emetic dose for monkeys. By serological means, the rate of free toxin in stomach, intestinal contents and fecal matter after and before application of antiserum was identified as an indicator for the neutralization of the toxin in vivo. As toxin, <em>S. aureus</em> enterotoxin B (SEB) was used which could be identified in the samples up to three hours after oral feeding. Antiserum was obtained by immunization of rabbits with toxoid. The application of antiserum one hour after intoxication resulted in an average reduction of free toxin in the intestine of 87,5 and 86,5%, (10 independent trials) one and two hours, respectively after serum administration. Effect of ionic strength and of protein structure modifying agents supports views suggesting the subunit structure of this enzyme. Chelating and reducing agents, thiol group inhibitors, and bile acid salts do not influence the enzyme activity. Clinically, a diagnosis of cirrhosis of the liver associated with atypical coma hepaticum had been suspected. Apart from damage due to alcohol abuse, no other underlying disease could be found. The fecal matter of a budgerigar (<em>Melopsittacus undulatus</em>) has been suggested as the probable source of infection. Using Guizotia abyssinica creatinine agar, <em>Cryptococcus neoformans</em> could be isolated from dry fecal matter collected from the bird cage. In a pet shop in the neighbourhood of the patient's home, fecal matter from various pet bird species was collected. Using the same method, examination also revealed the presence of <em>Cr. neoformans</em>. Contrastingly, masses of pigeon manure found in the garret of the patient's house did not contain <em>Cr. neoformans</em>. The epidemiological significance of the fecal matter of pet birds as a habitat for <em>Cr. neoformans</em> is discussed.</p></div><div><p>Bei einer 69jährigen Patientin, die eine passionierte Vogelhalterin (für Wellensittiche) war, wurde bei der Autopsie als überraschungsdiagnose eine Cryptococcose mit vorwiegender Manifestation am Zentralnervensystem diagnostiziert. Die klinische Verdachtsdiagnose lautete: Leberzirrhose mit atypischem „Coma hepaticum”, wobei die Leberzirrhose autoptisch bestätigt wurde. Für eine maligne Erkrankung ergab die Sektion keinen Anhalt. Aus</p><p>den trockenen, mehr als ein Jahr alten Fäkalien aus dem Käfig eines Wellensittichs gelang mit Hilfe von <em>Guizotia abyssinica</em>-Kreatinin-Agar die Isolierung von <em>Cryptococcus neoformans</em>. Auch in der nächstgelegenen Vogelhandlung wurde mit derselben Methode in Vogelfäkalien (Papagei) <em>Cr. neoformans</em> nachgewiesen. Haufen von Taubenfäkalien auf dem Dachboden des Hauses der Verstorbenen enthielten keinen <em>Cr. neoformans</em>. Die epidemiologische Bedeutung der Fäkalien von Stubenvögeln als Standort und Streuherd von <em>Cr. neoformans</em> wird diskutiert. Assessment of the appropriate concentration of the two decontaminants and the optimum contact time was made. It was observed that 2.5% sodium hydroxide with a contact time of 20 min and 3.5% oxalic acid with a contact time of 30 min yielded better results as compared to other permutations tried. Sputum specimens from 368 cases admitted to Ojha Institute of Chest Diseases were cultured for <em>Mycobacterium tuberculosis</em> by the modified methods described here and compared with the standard method. Higher number of positive cultures from film positive, as well as film negative specimens and fewer negatives and contaminations were observed with pancreatin/3.5% oxalic acid method. Comparatively less positives and more negatives and contaminations were noted when pancreatin/2.5% sodium hydroxide combination was used. Standard method, as compared to the pancreatin/2.5% sodium hydroxide and pancreatin/3.5% oxalic acid methods, gave fewer positive cultures both from film positive specimens and film negative specimens and higher negatives and contaminations were observed. Merits and demerits of the three methods are discussed.</p></div><div><p>Pankreatin wurde in Kombination mit Natriumhydroxid und Oxalsäure zur Schleimlösung angewandt. Die geeigneten Konzentrationen der beiden Chemikalien, die die Begleitflora abtöten sollen und die günstigste Einwirkungszeit wurden ermittelt. Es stellte sich heraus, daß eine Konzentration von 2,5% Natriumhydroxid mit einer Einwirkungszeit von 20 Min. und 3,5% Oxalsäure mit einer Einwirkungszeit von 30 Min. bessere Ergebnisse erzielte als die anderen untersuchten Konzentrationen.</p><p>Sputumproben von 368 Patienten des Oija-Krankenhauses für Lungenkranke wurden mit den hier beschriebenen Methoden im Vergleich zur Standardmethode kulturell auf <em>Mycobacterium tuberculosis</em> untersucht. Eine höhere Zahl positiver Kulturen sowohl von mikroskopisch positiven als auch von mikroskopisch negativen Proben und weniger negative und verunreinigte Kulturen wurden mit der Methode Pankreatin +3,5% Oxalsäure erzielt. Bei der Anwendung von Pankreatin +2,5% Natriumhydroxid ergaben sich im Vergleich mehr negative und verunreinigte Proben. In the mother dogs, detectable IgG and IgM antibodies were found. The IgG antibodies gradually increased in the course of this observation. The most elevated levels of IgA antibodies were observed one month after birth. The same was true for the CTA levels.
