Analysis was performed using Sequenase and Klenow enzymes on the target sequence in an M13mp18/19 cloning system. The open reading frame consisted of 438 bp encoding 146 amino acids of the protein, with a calculated Mr of 15507. Specific insulin binding to receptors was found to be decreased in the elderly group as compared with the youngsters. The high affinity binding sites averaged 82 +/- 16/RBC in the elderly group, but 109 +/- 22/RBC in the youngsters, and the low affinity binding sites averaged 1497 +/- 371/RBC and 2177 +/- 461/RBC respectively. Affinity constants did not differ significantly between the two groups, and fasting blood glucose and insulin levels were the same statistically. Our results indicate that senility might lead to a decrease in the number of binding sites on cells. The results showed that chronic hypoxia (15 days) obviously increased cAMP content of blood plasma, lung and myocardiac tissues. Verapamil, a calcium agonist, attenuated the increase of cAMP content in blood plasma and lung but did not affect cAMP content of the myocardium. Exacerbation of intra-erythrocytic oxidative stress might provide a means to kill the parasites. Sodium artesunate (SA), an effective Chinese anti-malaria drug, markedly increased the levels of active oxygen species and production of malonyldialdehyde in normal red blood cells and, to a greater extent, in malaria infected red blood cells. SA caused a remarkable decrease of unsaturated fatty acids content in normal red blood cell membrane. These suggest that the anti-oxidative system in red blood cells infected with malaria is jeopardized. Certain active oxygen species generated and accumulated in such red blood cells might in turn kill the parasites. It was found that gossypol may be an inhibitor of outward (Na+, K+) cotransport in human red cells but has no significant effect on (Na+, K+)-ATPase. Sixteen cases were accompanied with infertility. All data were synchronized to the day of midcycle serum luteinizing hormone (LH) peak (D0) and analyzed accordingly. In the endometriosis group, mean serum P concentrations of early, mid luteal phases and the integrated P concentration of the whole luteal phase were significantly lower than in the control group. Five cases (26.3%) had low peak P values during mid-luteal phase and/or low integrated P levels. Delayed endometrial maturation was revealed in 10 of 16 cases (62.5%). Short luteal phase, as defined by BBT, was observed in 3 of 14 cases (21.4%). A total of 12 cases were screened for all three parameters, among them, ten (83.3%) had at least one abnormality and seven (58.3%) showed two aberrant results. In summary, the incidence of luteal phase defect was higher in the endometriosis group than in controls. The results showed that the taking rate of CC801 tumor cells was 70%. Two weeks after implantation, the tumor cells were surrounded by pseudocapsule and showed no invasiveness, but 4 weeks after transplantation, they began to proliferate and to penetrate the pseudocapsule. Thereafter, the tumor cells continued to invade the neighboring tissues, causing degeneration and atrophy of seminiferous tubules at later stages. After 12 weeks, the tumor tissues occupied the whole testis. The process of invasion could be divided into 5 stages: latency, proliferation, early invasion, intermediate invasion and late invasion. Ten min after i.v. administration of the drug, the highest level was found in the intestine. Levels in other tissues were in the following decreasing order: brain, liver, kidney, testicle, muscle, fat, heart, serum, eyeball, spleen, and lung. However, 1 h after administration, the drug levels dropped significantly in all tissues, but not in the same proportions. High levels remained in the brain, fat, intestine and serum. Drug levels in other tissues were very low. After oral administration of Lig (30 mg/kg body weight/day) for 10 days, the blood pressure of RHR was not significantly changed (before vs after experiment: 147.50 +/- 2.50 vs 150.00 +/- 13.42 mmHg). The active Ca2+ uptake rate of IOVs from RHR was 5.03 +/- 1.15 nmol/ng IOV protein/min, and this was lower than that of IOVs from normotensive Wistar rats (8.95 +/- 1.08 nmol/ng IOV protein/min, P less than 0.01). In RHR, no change in IOV Ca2+ uptake capacity was observed after administration of Lig in vivo. Experiments in vitro showed that Lig markedly reduced Ca2+ uptake by IOVs from both RHR and control rats.
