Thaib, Mimi Takaria","doi":"10.52711/0974-360x.2024.00017","DOIUrl":"https://doi.org/10.52711/0974-360x.2024.00017","url":null,"abstract":"Renal cell carcinoma (RCC) is a malignant neoplasm originating from renal epithelium, with the clear cell renal cell carcinoma (ccRCC)being the most common type (80%) and the most common cause of death among other types of kidney cancer. Pathological stage is an important parameter that affects ccRCC survival, followed by nuclear grade. Pathological staging of RCC according to the AJCC (American Joint Committee on Cancer) TNM system 8th edition is based on local extension of the main tumor (T), involvement of lymph node (N), and metastasis (M). Ki-67 is a marker of proliferation used to assess tumor grade. High Ki-67 correlates with poor prognosis, advanced clinical and pathological features, thus Ki-67 can be used as a biomarker in the management of RCC.Ki-67 is routinely used to see the proliferation index in various cases of malignancy, but not in kidney malignancy. Programmed death ligand 1 (PD-L1) acts as a negative regulator of T cell-mediated anti-tumor immune responses. PD-L1 is expressed on T cells, B cells, macrophages, dendritic cells, endothelial cells and in various tumor cells including ccRCC. This study aims to determine the correlation between the expression of PD-L1 and Ki-67 in various T-stage clear cell renal cell carcinomas. Material and Method: This was an observational analytical study with cross-sectional approach toward 52 cases of ccRCC whose diagnosis was made histopathologically at the Anatomical Pathology Installation of Dr. Soetomo General Academic Hospital Surabaya from January 2014 to December 2020. Immunohistochemical stainingwas carried out using Ki-67 and PD-L1 antibodies, followed by an assessment using a scoring system. T-stage data were obtained from the patients’ medical recordswhich were then analyzed statistically with the Spearman test. Result: The study included 52 cases of ccRCC obtained from nephrectomy specimens at RSUD dr. Soetomo between 2014–2020. The age distribution of the subjects was 29–69 years and the mean and median age was 53 years. The ratio of male patients compared to female patients was 2.5:1. The majority was stage T2 (50%). Statistical test results showed no correlation between the expression of PD-L1 and Ki-67 in various T-stage clear cell renal cell carcinomas (p=0.965 and p=0.680). Conclusion: This study showed no correlation between the expression of PD-L1 and Ki-67 in various T-stage clear cell renal cell carcinomas. Heterospathe elata is a plant belonging to the Arecaceae family having numerous medicinal properties. The present study aimed to investigate the antidiabetic and anti-inflammatory activities of green solvent extracts of Heterospathe elata fruit. Method: The extraction was carried out with green solvents like isopropyl alcohol, hydro alcohol, and water with increasing order of polarity. In-vitro antidiabetic activity was evaluated by alpha-amylase and alpha-glucosidase inhibition assays. Anti-inflammatory activity was evaluated by the Bovin denaturation method and HRBC membrane stabilization. Result: Our results revealed that out of three green extracts hydro alcohol extract of Heterospathe elata fruit had the highest antidiabetic and anti-inflammatory activity. Gourami (Osphronemus goramy) fish scale comprises type 1 collagen and it has been used as scaffolding material in bone tissue engineering. As alternative bone graft material, the scales have a big potency to promote osteogenesis in periodontal bone defect when autografts are not feasible. This study will analise Runx2 and Osteocalcin expression in wistar rat alveolar bone induced by type 1 collagen derived from gourami fish scale. Methods: 32 male Wistar rats were divided into four groups; control group—7 days (C7), treatment group—7 days (P7), control group—14 days (C14), and treatment group—14 days (P14). The left mandibular incisivus was extracted and the tooth socket was treated with 10mg collagen. The rats were euthanized (at day 7th and 14th) and immunohistochemistry was performed using monoclonal antibodies anti-RUNX2 and anti-osteocalcin. Results: After seven days and 14days, the expression of RUNX2 and osteocalcin in the treatment group increased significantly (p<0.05) compared with the control group. Initially, the structural examination was conducted using various spectroscopic techniques, such as FTIR, and UV-visible spectroscopy, which are important in determining substituents, functional groups, and the presence of conjugated double bonds in CSO. From the FTIR spectra, a variety of sharp, strong, and weak peaks were specified relating to the main components of Linalol, and Decenolin CSO. The results of UV-visible spectroscopy confirmed the presence of Linalool as a major compound, and conjugated double bonds were also found. Furthermore, anti- microbial activity of CSO was studied against various types of bacteria. Strong bacterial inhibitory effects were observed, especially against S. aureus, with an average inhibition zone of 18 mm. It was also found from the structural characterization of CSO that the existence of Linalol is responsible for potential antibacterial activity. A C18 BEH analytical column was employed. A mobile phase of 0.2 percent formic acid in water and acetonitrile were used in a 20:80 v/v ratio. Samples are ionized using +ESI ionization and detected in MRM mode. The retention time of 0.80 min and 0.96 min were observed for Cefepime and Tazobactam respectively. The drugs have linear responses in the investigated range, with r2 = 0.9955 for Cefepime and r2 = 0.9816 for Tazobactam with increased precision and accuracy. The LOD for Cefepime and Tazobactam was 66ng/ml and 8.3ng/ml respectively. The LOQ for Cefepime and Tazobactam was 200ng/ml and 25ng/ml respectively. Many factors come into the picture, which reflects the ultimate elution of chemical compounds. Factors like the physicochemical properties of chemical compounds, the chemical nature of the mobile phase, instrumental factors, and experimental conditions play crucial roles in RP-HPLC method development. This research article discusses the challenges faced while developing a method of L-Arg along with polyphenolic compounds. Both the compounds have contrasting solubility profiles as L-Arg is soluble in water but PIC is soluble in organic solvents. Strategies were used to develop RP-HPLC of this combination with mobile phases like acetonitrile, orthophosphoric acid, methanol, and potassium dihydrogen phosphate buffer of pH 2.6. Siponimod (SPM) a selective modulator of sphingosine 1-phosphate receptor subtype 1 and 5(S1P1,S1P5 receptors), was found to have neuroprotective effect in neurological disorders. The present study was conducted to evaluate its beneficial effects in AD. Materials and Methods: In-silico molecular docking and molecular dynamic simulation studies were carried to know its potential interactions with selected target proteins. In-vivo study was conducted inthirty rats divided randomly into five groups with six rats per group: Control group received Carboxy methyl cellulose; disease group were administered aluminium chloride (AlCl3);standard group received rivastigmine (RVST) with AlCl3; and the test groups received SPM (0.05mg/kg and 0.2mg/kg) with AlCl3. Morris water maze test and elevated plus maze was used to evaluate learning and memory. Behavioural changes and biochemical parameters estimation were performed at the end of experiment. Results: The molecular docking study using selected protein and ligands showed higher docking score and stable interactions at acetylcholinesterase (AChE) protein with SPM.Behavioural studies showed: decrease in transfer latency time in elevated plus maze; decrease in time to reach target platform and increase in time spent in target quadrant in Morris water maze test in SPM treated rats. Biochemical evaluation showed marked decrease in malondialdehyde (MDA), nitrite, myeloperoxidase (MPO)levels and increased antioxidant levels in SPM treated groups. SPM exhibited significant inhibitory activity onAChE. Conclusion: SPM was found to be effective in ameliorating AlCl3 induced AD. A method of using a potent third-generation cephalosporin, ceftizoxime, was suggested to synthesize new dipeptide derivatives. These derivatives were successfully synthesized by linking a dipeptide moiety, which includes L-Tryptophan-L-valine, L-Tryptophan-L-alanine, L-Tryptophan-L-histidine, and L-Tryptophan-L-phenylalanine as dipeptides to the aminothiazole moiety of ceftizoxime by an amide bond. Their chemical structures were confirmed by spectral analysis, including 1H-NMR,13CNMR, and FT-IR spectroscopy. Molecular docking for these new derivatives was performed on penicillin-binding proteins (PBPs) type 2a (3ZG0) of Methicillin-resistant Staphylococcus aureus, type 2X (5OJ0) of Streptococcus pneumonia and type 1b (5HLA) of E. coli, and has recorded higher affinity binding represented as PLP fitness on target enzymes. The binding scores were significant and may indicate better antimicrobial activities when compared with ceftizoxime. This improvement in affinity binding can be explained by the presence of indole and/or imidazole moieties in those derivatives. The indole and imidazole moieties are actual pharmacophores with various biological activities and may contribute to affinity binding, and the derivatives are considered molecular hybrids. Furthermore, a preliminary evaluation of the antibacterial activity of the synthesized derivatives was performed against two significant bacterial species (MRSA and E. coli), which showed better activity in compression to ceftizoxime. Moreover, the derivatives were tested on the protein oligopeptide (POT) family system and have recorded very interesting results for possible oral absorption when compared with Ceftizoxime, Val-acyclovir, and Val-Val-Acyclovir, as reference drugs. The papers were analyzed using R and SciMAT software. A total of 1820 articles were analyzed and 4191 keywords plus, and 3242 author keywords were reported in the journals. Moreover, 7482 authors contributed in conducting these studies, of which 111 articles were written by single authors, and the collaborative coefficient was 4.36. Neuropsychopharmacology, International Clinical Psychopharmacology, and Journal of Clinical Psychopharmacology are the most significant journals publishing OCD articles. This study uncovered new insights into the evolution trend of the leading research stream on OCD over the past years. There is also no tabulated monograph on any such topic. Methods: Search the scholastic domain from various perspectives and set the citations in chronological manner. Results: There are more than 175 published scientific documents dated between 1957-2023. Less known. Conclusion: An unique histographic monograph comprised of 178 citations is presented with Punica and EA as the candidate pair. Never before like ground breaking. A fresh new way of distilate packed ‘review presentation’.
