{"title":"[Analysis of inflammation-related dietary patterns and sarcopenia in the elderly].","authors":"Tianran Jiao, Rongchang Pu, Bo Teng, Qingqing Man, Jian Zhang, Cheng Li, Pengkun Song","doi":"10.19813/j.cnki.weishengyanjiu.2025.06.004","DOIUrl":"https://doi.org/10.19813/j.cnki.weishengyanjiu.2025.06.004","url":null,"abstract":"<p><strong>Objective: </strong>To explore the association between inflammation-related dietary patterns and sarcopenia and its related muscle health indicators in the elderly.</p><p><strong>Methods: </strong>For this study on improving nutrition among the elderly in China, we selected Wuyuan County in Bayannur City, Inner Mongolia Autonomous Region; Taicang City in Jiangsu Province; and Yuexiu District in Guangzhou City, Guangdong Province.834 eligible subjects aged 65 and above were finally included. Dietary data were collected through the food frequency questionnaire(FFQ). Inflammation-related dietary patterns were extracted using the reduced-rank regression method, and sarcopenia was diagnosed using the Asian Working Group on Muscular Attenuation(AWGS) 2019 recommendation criteria. Logistic regression model was used to analyze the relationship between inflammation-related dietary patterns and sarcopenia and muscle health-related indicators in the elderly.</p><p><strong>Results: </strong>The prevalence of sarcopenia in the study population was 19.9%. The inflammatory markers hypersensitive C-reactive protein and interleukin-12 were significantly associated with the risk of sarcopenia(OR=1.12(95%CI 1.06-1.18), OR=1.08(95%CI 1.03-1.14)). Analysis of dietary patterns related to inflammation showed that poultry meat, legumes, flour, animal oil and other foods were more consumed, while vegetable oil, rice, soybeans, eggs, fruits, potatoes, vegetables and other foods were less consumed. In the adjusted model, there was no significant correlation between inflammation-related dietary patterns and the risk of sarcopenia. The compounds were separated by Waters BEH C_(18) column(2.1 mm×100 mm, 1.7 μ m) under the mobile phase consisted of 0.1% ammonia and methanol, and then detected by multi reaction monitoring scanning mode with ESI negative scan, and quantified by internal standard method.</p><p><strong>Results: </strong>The correlation coefficients(r) of the three compounds were more than 0.999 in the range of 0.05-50 ng/mL. The detection limit and quantification limit of the method were 0.0007-0.001 ng/m~3 and 0.003-0.005 ng/m~3, respectively. In 2023, a total of 651 manufacturing workers were selected from 8 manufacturing factories in two batches using the stratified targeted sampling. Sample 1(n=218) was used for the pre-test of the initial scale to form the formal scale, and Sample 2(n=433) was used for reliability and validity analysis, among which 37 participants were selected for retest reliability examinations. Descriptive statistics, item analysis, and exploratory factor analysis were conducted on the sample data, and the internal consistency reliability, criterion-related validity, and retest reliability of the data were tested. Confirmatory factor analysis was performed on the data from the formal test sample. Two scales, the effort-reward imbalance(ERI) and job content questionnaire(JCQ), are used as a criterion scale to test criterion validity. Item analysis, exploratory factor analysis and confirmatory factor analysis were used to test the reliability and validity of the scale.</p><p><strong>Results: </strong>The scale contains 33 items, which are divided into four dimensions: efforts-rewards, demand-ability, organizational climate and working environment, with 8, 8, 8 and 9 items respectively. The overall model had good fitness(χ~2/df=2.713, CFI=0.905, IFI=0.905, RMSEA=0.063), with each factor loading rangeing from 0.45 to 0.96. The correlation analysis between the scores of the formal scale and the criterion scale showed that the total scores of the two scales were positively correlated(r=0.758, P&lt;0.01). The internal consistency analysis showed that the Cronbach&apos;s α=0.960 for the whole scale, and each dimension&apos;s Cronbach&apos;s α ranged from 0.894 to 0.934. Baseline and follow-up surveys were conducted in November-December 2019 and November-December 2020, including a total of 3662 students who completed both the baseline and the follow-up, among which there were 976 males and 2686 females. Childhood Trauma Questionnaire-28 Item Short Form, Self-rating Questionnaire for Adolescent Problematic Mobile Phone Use, Pittsburgh Sleep Quality Index, Sleep-Rating Depression Scale and Sleep-Rating Anxiety Scale were used to evaluate childhood abuse experiences, mobile phone dependence, sleep quality, depression and anxiety symptoms, respectively. The partial correlations analysis was used to analyze the association between variables. After adjusting for age, gender, urban and rural areas, single-child status, parental education level and family economic level, the chain mediating effect of mobile phone dependence and sleep quality on the association between childhood abuse experiences and depression and anxiety symptoms was examined by bootstrap program and PROCESS software.&lt;/p&gt;&lt;p&gt;&lt;strong&gt;Results: &lt;/strong&gt;The detection rates of depressive symptoms and anxiety symptoms are 8.49% and 9.48%, respectively. As the partial correlations analysis shows, after controlling for anxiety symptoms, childhood abuse experiences showed significant positive correlations with mobile phone dependence, sleep quality, and depression symptom(r=0.120, r=0.154, r=0.135, P&lt;0.01). Mobile phone dependence was significantly positively correlated with sleep quality and depression symptoms(r=0.333, r=0.135, P&lt;0.01), and sleep quality showed a significant positive correlation with depression symptoms(r=0.131, P&lt;0.01). After controlling for depression symptoms, childhood abuse experiences showed significant positive correlations with mobile phone dependence(r=0.108), sleep quality(r=0.147), and anxiety symptom(r=0.092, P&lt;0.01). Mobile phone dependence was significantly positively correlated with sleep quality and anxiety symptoms(r=0.323, r=0.047, P&lt;0.01), and sleep quality showed a significant positive correlation with anxiety symptoms(r=0.100, P&lt;0.01). {"title":"[Meta-analysis of the prevalence of malnutrition among the elderly in China in 2000-2023].","authors":"Yuying Wang, Fusheng Li, Jing Nan, Yu Zhou, Yuxiang Yang, Shuya Cai","doi":"10.19813/j.cnki.weishengyanjiu.2025.06.020","DOIUrl":"10.19813/j.cnki.weishengyanjiu.2025.06.020","url":null,"abstract":"<p><strong>Objective: </strong>Meta-analysis was conducted to estimate the prevalence of malnutrition among the elderly in China.</p><p><strong>Methods: </strong>According to the corresponding term in Chinese and English, such as malnutrition, underweight, elderly, China, etc. , literature searches were conducted on PubMed, Embase, Web of Science, Wanfang Digital Database and China National Knowledge Infrastructure Database from January 1, 2000 to December 31, 2023 for literature related to the nutritional status of the elderly(≥60 years old) in China. The design was cross-section study.</p><p><strong>Results: </strong>53 articles, 22 in English and 31 in Chinese, were finally included which covered 701 434 elderly people aged 60 years or older, and the prevalence of malnutrition varied widely(2.4%-52.5%). The Meta-analysis showed that the prevalence of malnutrition among the older was 13.0%(95%CI 10.7%-15.7%). There was significant heterogeneity among the studies(I~2=99.9%, P&lt;0.01). The prevalence of malnutrition among male and female group were 11.2% and 12.2%, respectively, with a statistically significant difference(χ~2=152.68, P&lt;0.01). The prevalence of malnutrition among the elderly was 11.5% in urban areas, which was lower than that in rural areas(13.6%), with a statistically significant difference(χ~2=613.29, P&lt;0.01). Both groups underwent group measurements before and after a 12-week basketball intervention program.</p><p><strong>Results: </strong>Significant time × group interactions were observed for aggressive behavior(F=33.27, P&lt;0.01, η_p~2=0.23), self-efficacy(F=23.08, P&lt;0.01, η_p~2=0.17), and self-control(F=21.20, P&lt;0.01, η_p~2=0.16) following the basketball intervention. The intervention had a direct effect on aggressive behavior(β=-0.59, P&lt;0.01), accounting for 60.82% of the variance. Self-efficacy(β=-0.26, P&lt;0.01) mediated 19.59% of the effect(indirect effect=-0.19), while self-control(β=-0.23, P&lt;0.01) mediated 12.37%(indirect effect=-0.12). Demographic information, dietary intake during pregnancy and weight before pregnancy were collected through questionnaires. Serum samples in the third trimester and umbilical cord serum samples at birth were collected. The levels of L-arginine in serum were detected by liquid chromatography-mass spectrometry, the levels of cytokines interferon-γ(INF-γ) and interleukin-6(IL-6) were detected by enzyme-linked immunosorbent assay, the levels of immunoglobulins(Ig)A, IgG and IgM were detected by immunoturbidimetry. The correlation between L-arginine level and some immune indexes was analyzed by univariate analysis and multifactorial analysis.</p><p><strong>Results: </strong>The serum L-arginine level of pregnant women was(266.74±128.46) μmol/L and the level of L-arginine in umbilical cord serum was 160.00(106.50) μmol/L. The levels of IgA, IgM and IgG in umbilical cord blood were 0.068(0.013), (0.877±0.135) and(10.637±2.407) g/L respectively. The levels of IL-6 and IFN-γ in umbilical cord blood and maternal blood were 3.361(4.686), 52.531(61.387), 3.924(9.154) and 29.492(31.780) pg/mL respectively. L-arginine level in umbilical cord serum was negatively correlated with IL-6(r=-0.219, P=0.04), and positively correlated with IFN-γ(r=0.229, P=0.03) in pregnancy serum. After adjusting for factors such as the energy intake in the pregnant woman&apos;s diet, the age of the pregnant woman, the gender of the newborn, the gestational age at birth, and the birth weight and other factors, L-arginine level in umbilical cord serum were positively correlated with IgA(β=0.191, P=0.02) and IgG(β=0.191, P=0.02) levels in umbilical cord serum and IFN-γ(β=0.322, P&lt;0.01) levels in pregnant serum.</p><p><strong>Conclusion: </strong>The level of L-arginine in umbilical cord blood is positively correlated with the levels of IgA and IgG in umbilical cord blood and the level of IFN-γ in maternal blood. {"title":"[Construction and mechanisms of D-galactose-induced blood-brain barrier organoid aging model].","authors":"Jing Luan, Qin Zhuo, Jingbo Wang, Liyuan Wang, Wen Qin, Zhixuan Fan, Xueting Mei, Zhuo Yang","doi":"10.19813/j.cnki.weishengyanjiu.2025.06.016","DOIUrl":"https://doi.org/10.19813/j.cnki.weishengyanjiu.2025.06.016","url":null,"abstract":"<p><strong>Objective: </strong>To establish a D-galactose-induced aging model of the blood-brain barrier(BBB) with organoid technology and explore the pathophysiological mechanisms of BBB aging.</p><p><strong>Methods: </strong>A BBB model was constructed on an organ-on-a-chip using human cerebral microvascular endothelial cells(hCMEC/D3), followed by D-galactose exposure to induce aging. The model was evaluated by measuring transendothelial electrical resistance(TEER), apparent permeability coefficient(Papp) of fluorescein, β-galactosidase staining, live/dead cell staining, lactate dehydrogenase(LDH) activity, and reactive oxygen species(ROS) levels. The expression of tight junction proteins(ZO-1, Occludin), senescence-associated gene(p53), and anti-inflammatory genes(HO-1, Nrf2) was assessed using RT-qPCR to characterize aging features.</p><p><strong>Results: </strong>The optimal concentration for inducing BBB organoid aging was 50 mg/mL D-galactose. The model group significantly reduced TEER((19.46±0.98)Ω·cm~2 vs. (22.07±0.21)Ω·cm~2, P&lt;0.05), increased Papp of fluorescein((1.08±0.02)×10~(-4 )cm/s vs. (4.39±0.01)×10~(-5 )cm/s, P&lt;0.05), elevated β-galactosidase-positive cells, decreased live cells, increased dead cells, and increased LDH release and ROS levels. RT-qPCR analysis showed downregulation of ZO-1 and Occludin, along with upregulation of p53, HO-1, and Nrf2.</p><p><strong>Conclusion: </strong>D-galactose induced aging-associated damage in the BBB organoid model, providing a reliable in vitro biomimetic platform for studying BBB aging mechanisms and potential therapeutic interventions for neurodegenerative diseases. The control group received oral gavage of corn oil and intratracheal instillation of sterile saline. The β-carotene group was administered β-carotene orally and saline intratracheally. The PM_(2.5) group received corn oil orally and PM_(2.5) intratracheally. The β-carotene+PM_(2.5) group was treated with β-carotene orally and PM_(2.5) intratracheally. Oral doses of corn oil and β-carotene were administered daily at 50 mg/kg. PM_(2.5) was instilled intratracheally at 5 mg/kg every 3 days. After 25 cycles, 3 mice from the control and PM_(2.5) groups were randomly euthanized for lung tissue collection and Masson&apos;s trichrome staining to validate fibrosis model establishment. After the successful establishment of the model, the mice were euthanized. Lung sections were prepared for hematoxylin-eosin(HE) and Masson&apos;s staining under light microscopy, collagen volume fraction(CVF) in Masson-stained sections was analyzed using ImageJ. Serum, bronchoalveolar lavage fluid(BALF), and lung tissues were collected to measure serum levels of hydroxyproline(HYP), glutathione(GSH), superoxide dismutase(SOD), malondialdehyde(MDA), and BALF total protein using commercial kits. Western blotting was performed to assess protein expression of collagen type I(COL-I), epithelial cadherin(E-cadherin), inositol 1, 4, 5-trisphosphate receptor(IP_3R), voltage-dependent anion channel(VDAC), and mitochondrial calcium uniporter(MCU)in lung tissues, normalized to β-actin.&lt;/p&gt;&lt;p&gt;&lt;strong&gt;Results: &lt;/strong&gt;After 30 cycles, Masson staining showed that the model was successfully established. Compared to the control group, PM_(2.5)-exposed mice exhibited significantly reduced weight and increased lung coefficient(1.54±1.97)g and 1.13%±0.26%, P&lt;0.01). No significant abnormalities were observed in the lung tissue of control and β-carotene group mice, CVF showed no statistically significant difference. Compared to the control group, PM_(2.5)-exposed mice pathological analysis revealed substantial inflammatory cell infiltration and erythrocyte exudation in alveolar spaces and pulmonary interstitium, along with interstitial thickening, alveolar structural deformation(including alveolar rupture or fusion), and compromised alveolar integrity. Pulmonary tissue exhibited significant blue collagen deposition, The CVF in the PM_(2.5) group(18.34%±1.96%) was significantly higher than that in controls(P&lt;0.01). Serum levels of GSH((40.56±5.77)μmol/L, P&lt;0.01) and SOD((9.24±1.46)U/mL, P&lt;0.01) were markedly reduced, while MDA increased((16.33±1.40)nmol/mL, P&lt;0.01). The control group was inoculated with gut microbiota from normotriglyceridemic humans and maintained on a standard sterile basal diet, whereas the other two groups received microbiota from hypertriglyceridemic humans and were fed either the same basal diet(NDG group)or a high-fat diet(HFDG group), respectively. A fecal microbial suspension was administered by gavage at 1 mL per 100 g body weight on alternate days for a total of three inoculations, followed by a 2-week period to allow microbiota engraftment and adaptation. Subsequently, the rats were maintained on their respective basal or high-fat diets for 9 weeks. Body-weight changes, serum lipid profiles, hepatic lipid accumulation, liver function indices, and gut microbiota composition were assessed.</p><p><strong>Results: </strong>After intestinal microbiota colonization followed by 9 weeks of continuous feeding on their respective diets, the NDG group exhibited higher body weight, liver index, epididymal fat index, and subcutaneous fat index than the control group, although none of these differences reached statistical significance. In contrast, the HFDG group was significantly elevated for all these parameters versus the control group, while its brown adipose tissue index was markedly lower. Serum lipid and hepatic function analyses revealed no significant differences between the control and NDG groups. Relative to the control group, the HFDG group displayed a 1.47-fold increase in serum total cholesterol(TC) and a 2.06-fold increase in LDL-C, both statistically significant. Serum ALT in the HFDG group was significantly higher than in controls(P&lt;0.05), whereas AST showed an upward trend but lacked statistical significance. Histologically, the HFDG livers contained markedly more red-stained lipid droplets, and the droplet area fraction was significantly greater than in controls. Concurrently, microbial diversity in the HFDG group was lower than in the control group, and both phylum-and genus-level community structures diverged substantially from those of the NDG group.
