Lutsenko","doi":"10.15407/cryo32.03.196","DOIUrl":"https://doi.org/10.15407/cryo32.03.196","url":null,"abstract":"The state of the pituitary-thyroid system central and peripheral links (according to the levels of thyroid stimulating hormone (TSH) and thyroid hormones T4 and T3 in blood serum) were studied in 6- and 18-month-old rats under initiated circadian desynchronosis (CD) by the light regime inverting. The CD was simulated against the background of separate and combined eff ect of both the whole body cryostimulation (–120°C) (WBC) and injections of the nucleated cells (NCs) of human cord blood, applied preventively (for a day and week, respectively). In 6-month-old rats, the TSH level decreased under the CD and its initiation after separate NCs administration and combined with WBC (by 3.4, 3.9 and 6.3 times, respectively) and remained unchanged under CD against the WBC. In 18-month-old rats the TSH level was reduced only at CD by 1.8 times. The T4 level in rats of both age groups did not change under the CD as well as during its development against the background of a combined effect of WBC and NCs administration in 6-month-old animals and against the NCs injection in 18 months-old rats. The T4 level in rats of both age groups decreased at the background of separate effect of the WBC (by 1.6 and 1.4 times, respectively). In 18-month-old animals, the T4 increased under the CD development after a combined effect of WBC and NCs injection by 1.3 times. Stepaniuk, Ihor Grisha","doi":"10.15407/cryo32.02.121","DOIUrl":"https://doi.org/10.15407/cryo32.02.121","url":null,"abstract":"In this work, we experimentally substantiated the possibility of correcting the cytokine profile to reduce the inflammation in mice with adjuvant arthritis (AA) via combined use of lyophilized human cord blood leukoconcentrate (lHCBL) and cryopreserved human placental extract (cHPE). Adjuvant arthritis was modelled in CBA/H mice. To day 7 after pathology induction, the animals were injected with lHCBL and cHPE singly or in combination. To days 14 and 28 of observation, all the introduced products showed a therapeutic effect. The bone marrow mitotic indices were compared within an immediate examination of the thigh removed from the body and during a short-term hypothermia. As a result, no statistical differences were found between the mitotic levels during immediate examination and a short-term hypothermia in the intact animals and those after medicinal leech application, that evidenced to a hypothermal fixation of initial level of this index, preservation of biomembrane physiological state prior to hypotonic stress and cell morphology. Babijchuk","doi":"10.15407/cryo32.02.092","DOIUrl":"https://doi.org/10.15407/cryo32.02.092","url":null,"abstract":"Production of reactive oxygen species (ROS) and protein state of membrane-cytoskeleton complex (MCC) in the erythrocytes, subjected to freeze-thawing with polyethylene glycol (PEG) and subsequently transferred into physiological conditions in vitro after cryoprotective agent removal were examined. Results of SDS-PAGE with β-mercaptoethanol showed a decrease in spectrin content in the MCC profile of cryopreserved erythrocytes. The using of diamide revealed an increase in the accessibility of –SH groups of spectrin for the reagent and an enhanced level of high molecular weight polypeptide complexes relative to the control. A decrease in the content of protein band 8 (peroxiredoxin) in the MCC of cryopreserved erythrocytes revealed by SDS-PAGE with β-mercaptoethanol is apparently associated with a change in its localization caused by an increased ROS production at the previous stages of cryopreservation. Kalashnykova, Irina Shchetinska","doi":"10.15407/cryo32.02.144","DOIUrl":"https://doi.org/10.15407/cryo32.02.144","url":null,"abstract":"Here we have studied the effect of low-temperature storage of the placenta at different temperatures on anti-inflammatory activity of its extracts in cell models of erythrocyte thermohemolysis and adenosine-5’-diphosphate-induced platelet aggregation. The wound-healing effect of cryopreserved placental extracts was also evaluated in rat thermal burn model. When preserving placenta for up to a month, the temperature of storage was shown to cause no significant impact on the percentage of inhibition of erythrocyte thermohemolysis and platelet aggregation by the extracts. Placental storage for three months at –20°C decreased anti-inflammatory activity of its extracts, and led to its complete loss during 6 months. Placental cryopreservation at –196°C enabled to preserve the anti-inflammatory and wound-healing activity. Seed samples with a moisture content of 5–7% were stored in an airtight container for up to 42 months at 4 and –20°C. Control of plant germination and assessment of yield-related traits were carried out for three years according to standard methods. The research findings demonstrated the variation of seed germination depending on the sample genotype. No differences were noted in the longevity of seeds after 30 and 42 months of storage under various temperature regimens. The results of the analysis of yield-related traits of plants for two or three consecutive years did not show any significant advantages when influenced by 4 or –20°C on the spike length, number of productive stems and the mass of grain derived from the spike. The dynamics of blood pressure, heart rate, rheographic index and cardiorhythmography before and after the cold pressor test were evaluated. A significantly higher increase in systolic and diastolic BP in response to the cold pressor test was found in the persons with a normal high level of initial blood pressure (130/85–139/89 mm Hg) (p < 0.05). During the examination, a higher percentage of those with a hyperreactive blood pressure response was found (25%). In the experiment participants with initial blood pressure > 130/85 mm Hg a significant influence of the sympathetic nervous system on the cardiovascular system central and peripheral links in response to cold stress was observed. A decreased glycerol concentration in a cryopreservative and an increased content of impermeable protective component sorbitol were established to affect the erythrocyte characteristics at different stages of freezing and hypothermic storage. Reduction of 1,2-propanediol and NaCl concentrations when augmenting the sucrose content in cryopreservative agent enabled changing a two-stage mode of erythrocyte freeze-thawing to the single-stage one.
