Kovalenko","doi":"10.15407/cryo32.04.256","DOIUrl":"https://doi.org/10.15407/cryo32.04.256","url":null,"abstract":"In this work, we have studied the impact of various stages of cryopreservation by vitrification in ethylene glycolsucrose medium on plasma membranes adhesive properties of 8-cell murine embryos and their specific electrical conductivity. Embryos were divided into the following experimental groups: the control, group 1 (incubation in vitrification medium) and group 2 (complete cycle of cryopreservation). The embryo exposure to vitrification medium did not affect their ability to cell-to-cell adhesion. After a complete cycle of cryopreservation, no embryo adhesion was observed. Electrical conductivity of embryo membranes was determined using the pulsed conductometry. After incubation in vitrification medium, their resistance to electric pulse was shown to decrease, that was manifested in the phenomenon of irreversible electric breakdown. The average values of electrical conductivity varied within the following ranges: ((12.1 ± 1.5)...(55.5 ± 2.6)), ((28.7 ± 5.7)...(44.± 8.9)), ((31.0 ± 9.3)...(87.9 ± 26.1)) μS/cm in the control, groups 1 and 2, respectively. These findings may be explained by appearance of first structural disorders in lipid bilayer of embryo membranes even at the stage of their incubation in vitrification medium. Gulevsky","doi":"10.15407/cryo32.03.206","DOIUrl":"https://doi.org/10.15407/cryo32.03.206","url":null,"abstract":"In this report, we have studied a cardioprotective effect of low molecular weight (below 5 kDa) fraction derived from cattle cord blood in a rat model of infarction-like cardiovasopathy. The myocardial infarction development was initiated in the animals, maintained for two months on a diet with excessive intake of fat, calcium and sodium salts, by making hypoxia-hypercapnia in them. The use of this model allowed inducing the infarction-like cardiovasopathy, comparable by many respects with the clinical picture of myocardial infarction, which was confirmed by electrocardiography, enzyme-linked immunosorbent assay and histological analysis. Preliminary administration of low molecular weight (below 5 kDa) fraction of cattle cord blood, as well as Actovegin did not prevent the heart attack, but increased the endurance of rats to hypoxia-hypercapnia and reduced the damage rate to myocardium. The assessment of pain intensity on visual-analogue scale upon completing the rehabilitation therapy showed the pain disappearance in 68.4% of patients, and its significant reduction in 31.6%; increased daily motor activity, and decreased restrictions during walking in 90.8% of cases; improved well-being in 35.7%; enhanced activity in 43.9%, and mood change in 38.8%. Each WBEACT session increased the muscle strength and endurance for both genders of patients, thus testifying to a stimulating effect of cryotherapy to human body. The optimal moisture, ozone dose within the ozone-air mixture, and the exposure time, not reducing the germination rate, were determined. The ozonation, freezing, and their combined effect affected neither the seed germination in laboratory, nor biometric indices in a field. The seed freezing down to liquid nitrogen temperature was found to significantly increase its crop yield. Biochemical analysis of root crops showed all the ways of presowing treatment to reduce the content of total sugars and betanine. After freezing and ozonation, the amount of dry matter decreased, but after combined exposure it augmented. Ozonation and subsequent freezing lowered the content of ascorbic acid. Significantly reduced studied parameters did not reach the minimum values specific to this crop. The current methods to identify the DNA fragmentation rate have been shown to be quite efficient, but the choice for the optimal way is stipulated by animal species. The oxidative stress caused by an increased content of reactive oxygen species was recognized as the main mechanism in DNA cryodamage. To prevent this negative effect on germ genetic material, many protective media and rehabilitation solutions supplemented with antioxidants (non-enzymatic, enzymatic compounds and nanoparticles) have been developed. The patterns of morphogenesis in the soft winter wheat in vitro culture during exposure to positive low temperature of various durations (15, 30 and 45 days) have been established. The callus culture of 2–3 passages of cv. Doridna, Statna and Astet was used. The results of experiments showed the preliminary vernalization of calluses for 30 and 45 days to strongly stimulate the chlorophylogenesis and hemogenesis, thereby increasing the frequency of obtaining the regenerant plants during further cultivation. The efficiency of these processes depended on the original cultivar genotype and cold treatment duration.
