In order to make the most of the therapeutic potential of EVs, various loading strategies have been devised. Despite the widespread application of exogenous methods, endogenous approaches are becoming increasingly preferred instead. Medicinal macro molecules such as proteins and nucleic acids can be loaded with relative ease using this technology that manipulates parental cells through genetic engineering. We examine the most effective ways for EV loading and demonstrate why endogenous is superior. Extracellular vesicles (EVs), which are able to transport pharmaceuticals, have the potential to tackle the problems of biologic therapeutic stability and off-target undesirable effects. In addition, we investigate the most recent findings and applications of this innovative method in order to shed light on the numerous potential therapeutic possibilities in the field of EV-based treatments. The possibility of endogenous loading of EVs with big biological medicines has been brought to light by recent research. In order to optimize the loading of biologic drugs in extracellular vesicles (EVs), researchers have been able to apply cargo selection and loading pathway molecules thanks to advancements in EV biogenesis. It has been established that endogenous EV loading is successful in preclinical in vivo trials, which demonstrates its promise in a variety of therapeutic situations.","PeriodicalId":14182,"journal":{"name":"International Journal of Pharmaceutical Chemistry and Analysis","volume":" 44","pages":""},"PeriodicalIF":0.0,"publicationDate":"2024-07-15","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"141832912","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

The AML patients consisted of sixty patients (30 male, 30 female). The average age of patients was (36.44±8.44) years (range 25-45). Thirty healthy subjects were considered as controls (15 male and 15 female), and the average age of control group was (32.22±5.13) years. All subjects had to go through clinical examination to determine existence of other diseases in addition to evaluate some tests as complete blood count [hemoglobin (Hb), platelet (PLT), and white blood cells (WBCs)], ferritin, FBS (fasting blood sugar), TG (triglycerides), TC (total cholesterol), and HDL (high density lipoprotein cholesterol) (HDL-Ch), urea, and creatinine. Vitamin D3 was measured by minividase Biomerux/French. The VitD3 was highly significantly lower in AML patients than in controls (p=0.001). While, VitD3 was lower in male than in female AML patients but the difference was not statistically significant. There were statistically significant positive correlations between VitD3 and Hb, PLT, Wbc, ferritin, FBS, and TG in both male and female AML patients. These findings suggest that lower serum concentrations of vitamin D3 may possibly be considered as a potential factor for early assessment of acute myeloid leukemia.","PeriodicalId":14182,"journal":{"name":"International Journal of Pharmaceutical Chemistry and Analysis","volume":" 72","pages":""},"PeriodicalIF":0.0,"publicationDate":"2024-03-15","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"140392048","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Formulation excipients, including emulsifiers, thickeners, and preservatives, commonly used in skincare products, can significantly influence skin barrier function and permeability. This review evaluates the impact of these Excipients on skin barrier function using biophysical measurements such as Trans-epidermal Water Loss (TEWL), skin hydration, and trans-epidermal water content. Furthermore, it discusses the potential implications of excipient-induced alterations in skin barrier function for skin health and tolerability.","PeriodicalId":14182,"journal":{"name":"International Journal of Pharmaceutical Chemistry and Analysis","volume":" 13","pages":""},"PeriodicalIF":0.0,"publicationDate":"2024-03-15","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"140391441","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

It is believed that these plaques impair cognitive and functional capacities and induce neurodegeneration. Research on uncommon genetic variants that either increase or decrease the quantity of Aβ deposited offers proof that amyloid plaques are involved in the disease's progression. Early-stage amyloid plaque buildup increases the likelihood that moderate cognitive impairment may progress to dementia.Donanemab, a humanized IgG1 antibody, targets an N-terminal pyroglutamate Aβ epitope that is unique to plaques that have already begun to develop. It is specific to this epitope and has no known clinical effect. Furthermore, it doesn't show any off-target binding to neurotransmitters, their receptors, or other Aβ species. In this systematic review, we first collect the necessary information after carefully reviewing four different study articles. After the data is collected, we evaluate it and create a graph to compare those study articles.","PeriodicalId":14182,"journal":{"name":"International Journal of Pharmaceutical Chemistry and Analysis","volume":" 43","pages":""},"PeriodicalIF":0.0,"publicationDate":"2024-03-15","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"140391760","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

The main reason for its life-threatening nature is its high tendency to metastasis. The outlook for melanoma patients with distal metastases is grim, as they have a median survival of only six months, despite the utilization of the most advanced treatments now available. The Extracellular signal-regulated kinase (ERK) pathway is the most frequently altered oncogene in melanoma. Phytochemicals are receiving significant recognition due to their minimal toxicity, affordable price, and widespread acceptance as dietary supplements. Preclinical investigations have revealed many cellular and molecular processes via which phytochemicals function in the prevention and treatment of metastatic melanoma.: In this study, we performed virtual screening of 464 phytoconstituents were obtained from the Naturally Occurring Plant-based Anti-cancer Compound-Activity-Target (NPACT) database to identify novel ERK inhibitors.: Virtual screening carried out prediction of drug-likeness and molecular docking studies with 2OJG, protein target related to ERK pathway.: Nine compounds achieved better docking score as compared to co-crystallized ligand. The accuracy of the docking method was checked using re-docking. Picetannol and sulfurentin were identified as potential inhibitors with docking score of -9.2 and -8.2, respectively. These two phytoconstituents also found to be non-carcinogenic which was predicted using a free webtool, Swiss ADME.: Our finding suggests that Piceatannol has promising potential to be further explored as ERK-pathway inhibitor in treatment of melanoma","PeriodicalId":14182,"journal":{"name":"International Journal of Pharmaceutical Chemistry and Analysis","volume":" 9","pages":""},"PeriodicalIF":0.0,"publicationDate":"2024-03-15","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"140392041","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}
