{"title":"[Infective pathogens and drug resistance in burned patients].","authors":"Xi Zhang, Bo-cheng Zhao","doi":"","DOIUrl":"","url":null,"abstract":"<p><strong>Objective: </strong>To find out the changes in the infective pathogens and their drug resistance in burned patients.</p><p><strong>Methods: </strong>The patients were divided into two groups. The first group was from July 1991 to June 1996, and the second group was from July 1996 to June 2001. The bacteria of burned body surface and blood were cultured, and the bacteria and their drug sensitivity were analyzed.</p><p><strong>Results: </strong>Gram-negative bacteria were the major bacteria in burn infection, among which Pseudomonas aeruginosa ranked the top. Staphylococcus aureous ranked the first among the Gram-positive bacteria, and the isolation rate of methicillin resistant Staphylococcus aureus increased; The isolation rate of Enterobacter cloacae (10.4%), Escherichia coli (8.3%), Klebsiella pneumoniae (7.3%), and fungus (4.2%) all rose. {"title":"[Competitive RT-PCR assay for quantification of GPI-PLD gene expression].","authors":"Xiao-jie Zhang, Chun-ping Lu, Jian-hua Tang","doi":"","DOIUrl":"","url":null,"abstract":"<p><strong>Objective: </strong>To establish a competitive reverse transcription-polymerase chain reaction (RT-PCR) assay to quantify the expression of glycosylphosphatidylinositol specific phospholipase D (GPI-PLD) gene.</p><p><strong>Methods: </strong>A competitive GPI-PLD cDNA fragment was constructed by site-directed mutagenesis by PCR method. The competitive cDNA fragment had the same primer binding sites as the target cDNA, but was 20 bp shorter in length. The competitive cDNA was transcripted into the competitive RNA in vitro as the internal standard. Then the competitive RNA was reversely transcripted and amplified together with total RNA extracted from K562 cells. The two amplified cDNA fragments could be distinguished by 10% polyacrylamide gel electrophoresis. The concentration of cellular GPI-PLD mRNA was derived from the ratio between the intensities of the bands corresponding to the amplified products.</p><p><strong>Results: </strong>A 198 bp GPI-PLD competitive RNA was constructed and prepared. The competitive RNA could compete well with the target RNA in the RT-PCR reaction. The expression of GPI-PLD gene in K562 cells was (440 +/- 20) copies/ng.</p><p><strong>Conclusion: </strong>A competitive RT-PCR assay for the quantification of GPI-PLD gene expression may be established successfully. {"title":"[Expression of S100 in oral submucose fibrosis].","authors":"Xiao-li Xie, Zhan-gui Tang, Zhi-gang Yao","doi":"","DOIUrl":"","url":null,"abstract":"<p><strong>Objective: </strong>To study the expression of S100 in oral submucose fibrosis(OSF).</p><p><strong>Methods: </strong>Immunohistochemistry assay was performed on 10 cases of OSF, 10 cases of oral epithelial dysplasia (OED) and 8 cases of normal oral epithelium using monoclonal antibody against S100 protein.</p><p><strong>Results: </strong>S100 did not express in the normal oral muscosal epithelium. The S100 positive nuclear staining was found in 50% of the OSF and OED cases, which was significantly higher than that in the normal oral epithelium (P < 0.05). But the positive staining in the OSF and OED cases had no difference (P > 0.05).</p><p><strong>Conclusion: </strong>S100 expression is closely related to the development of OSF and OED. {"title":"[Surgical treatment of 92 cases of thoracic aortic aneurysms].","authors":"Guo-bao Song, Xin-min Zhou, Jian-guo Hu","doi":"","DOIUrl":"","url":null,"abstract":"<p><strong>Objective: </strong>To explore the timing of surgical treatment of thoracic aortic aneurysms.</p><p><strong>Methods: </strong>The clinical data of 92 patients with thoracic aortic aneurysm who underwent surgical treatment in our department during the latest 10 years were retrospectively reviewed.</p><p><strong>Results: </strong>The overall operative mortality was 13.04% (12/92) 30 days after the operation. Risk factors for the early mortality were the preoperative aneurysm breach, hypertension, important organ dysfunction and circulatory arrest during the operation, as well as the serious postoperative complications, such as arrhythmia, dysfunction or failure and infection of important organ, and so on (P < 0.05-0.01).</p><p><strong>Conclusion: </strong>The surgical treatment is an ideal way of treatment of thoracic aortic aneurysm. {"title":"[Clinical observation of recombinant growth hormone therapy for liver cirrhosis with hypoproteinemia].","authors":"Shao-jun Liu, Wu-liang Tang, Shou-rong Shen","doi":"","DOIUrl":"","url":null,"abstract":"<p><strong>Objective: </strong>To assess the effect of recombinant growth hormone (rhGH) therapy for liver cirrhosis patients with hypoproteinemia.</p><p><strong>Methods: </strong>Fifteen patients with liver cirrhosis were treated with rhGH (4 U per day) for 7 days and 15 controls were given human albumin (10 g) for every other day (altogether 3 times). {"title":"[Effect of Coptis Chinensis compound on the gene expression in transplanted tumor tissue in nasopharyngeal carcinoma cell line of CNE1 by cDNA microarray].","authors":"Guang-ping Wang, Fa-qing Tang, Jin-ping Zhou","doi":"","DOIUrl":"","url":null,"abstract":"<p><strong>Objective: </strong>To investigate the effect of Coptis Chinensis compound on the gene expression of transplanted tumor in nasopharyngeal carcinoma cell line of CNE1.</p><p><strong>Methods: </strong>The cells were injected into Balb/C nude mice, and after the transplanted tumor was established, the nude mice with tumors were treated with Coptis chinensis compound. The nude mice were then killed, tumor tissues were isolated and the total RNA was extracted. Hybridization with a cDNA microarray was conducted using a probe of fluorescent-labeled cDNA mixture that was reversely transcribed from the RNA. The gene expression level was analyzed after scanning the fluorescent intensity by a laser scanner, and further confirmed by RT-PCR.</p><p><strong>Results: </strong>After using Coptis Chinensis compound for 30 days, the size of transplanted tumors were remarkably reduced, and the inhibition rate of tumor growth was 29.5%. In the meantime, the expression of 147 genes in the tissue of implanted tumor treated with the compound altered: the expression of 102 genes was down-regulated, and that of 45 genes was up-regulated. {"title":"[Expression of ET-3 in hippocampal CA1 of ovariectomied mice after cerebral ischemia/reperfusion].","authors":"Zhi-jun Zhang, Zheng-qing Liu, Zhi-jian Ma","doi":"","DOIUrl":"","url":null,"abstract":"<p><strong>Objective: </strong>To detect the expression of endothelins-3 (ET-3) immunoactivity in hippocampal CA1 of ovariectomied (OVX) mice after ischemia/reperfusion (IR), and to further explore the relationship between estrogen and ET-3.</p><p><strong>Methods: </strong>Fifty-four adult female mice were randomly divided into 3 groups. 1. IR group: the bilateral common carotid occlusion (BCCO) was carried out for 7 min on the 1st, 3rd, 5th and 14th day of reperfusion. 2. OVX/IR group: firstly the ovariectomy was performed, and after a week, BCCO was carried out for 7 min on the 1st, 3rd, 5th and 14th day of reperfusion (each time spot, n = 6). 3. sham-operated group (n = 6).</p><p><strong>Results: </strong>Few ET-3 positive cells were diffusely distributed in the hippocampal CA1 region in the control group, whereas the expression of ET-3 increased in the experimental groups. The number of ET-3 positive cells significantly increased in the OVX/IR group compared with that of the IR group 1 d after the reperfusion (P < 0.01). The diagnostic standard of rare chromosomal aberation was: 1-2 abnormal kary types were found in every 30, 60, 200 mitotic phass.</p><p><strong>Results: </strong>The main clinical characteristics of 52 cases of individual chromosomal aberation in peripheral blood included: 1. spontaneous abortion; 2. abnormal sex development; 3. reproduction diseases; and 4. morbid and deformity.</p><p><strong>Conclusion: </strong>Rare chromosomal aberation should be regarded as chromosomal diseases. {"title":"[Ambulatory pulse pressure and left ventricular hypertrophy in patients with essential hypertension].","authors":"Yang Cao, Shui-ping Zhao, Lin Luo","doi":"","DOIUrl":"","url":null,"abstract":"<p><strong>Objective: </strong>To evaluate the relationship between the left ventricular hypertrophy (LVH) and the ambulatory pulse pressure and aortic root dimension (AOD) in essential hypertensive patients.</p><p><strong>Methods: </strong>We monitored the 24-hour ambulatory blood pressure and applied echocardiography in 107 essential hypertensive patients. Using the left ventricular mass index (LVMI) as an index in evaluating LVH, the patients were divided into 2 groups: 29 cases in the LVH group and 78 in the non-LVH group.</p><p><strong>Results: </strong>The average levels of 24-hour pulse pressure, daytime pulse pressure, nighttime pulse pressure and AOD were significantly different between patients with LVH and without LVH (P < 0.05).
