{"title":"[Protective effect of Polygonum multiflorum thunb on the cerebral cholinergic neurofibers in rats].","authors":"Min Li, Xiao-ping Du, Hui Ye","doi":"","DOIUrl":"","url":null,"abstract":"<p><strong>Objective: </strong>To observe the number and morphological changes of cholinergic neurofibers in rats under the destruction of kainic acid, and to investigate the protective effect and the mechanism of polygonum multiflorum thunb (PMT) on acetyl-chorine esterase (AChE) neurofibers.</p><p><strong>Methods: </strong>Excitative neurotoxin kainic acid was injected into the basal forebrain, Meynert neucleus, medial septal neucleus and the Broca neucleus to establish the destruction model. {"title":"[Dry chemistry method for measuring serum urea and creatinine].","authors":"Zheng-yong Zhong, Li-na Yang, Guo-feng Luo","doi":"","DOIUrl":"","url":null,"abstract":"<p><strong>Objective: </strong>To evaluate the dry chemistry method for measuring serum urea and creatinine.</p><p><strong>Methods: </strong>Urea and Creatinine were measured by dry and wet chemistry methods respectively. The results and the relativity between the two methods were compared.</p><p><strong>Results: </strong>The correlative factors were 0.996 and 0.995, respectively, implying fairly good comparability. The precision of the dry chemistry method was good, with the coefficient variation (CV) below 5%. {"title":"[Nuclear factor-kappa B activity at the early stage of brain ischemia and reperfusion in mice].","authors":"Jin-xiu Li, Jun-wei Deng, Guang-sen Zhang","doi":"","DOIUrl":"","url":null,"abstract":"<p><strong>Objective: </strong>To observe the activity of nuclear factor-kappa B(NF-kappa B) during reperfusion after temporary brain ischemia and to evaluate its effect.</p><p><strong>Methods: </strong>The brain ischemia-reperfusion(I/R) model of mice was established with ligating bilateral common carotid arteries and bleeding for 0.3 ml in the tail. Abnormal nervous symptoms (ANS) were recorded. Immunohistochemical technique was used to detect the activity of NF-kappa B subunit P65 and the inhibitory factor-kappa B alpha(I-kappa B alpha) in cerebral cortex during different periods of ischemia and reperfusion. Their mRNA expressions were also measured by RT-PCR.</p><p><strong>Results: </strong>NF-kappa B P65 activity significantly increased 30 minutes after the reperfusion, peaking at the 2nd hour, and remaining high within 24 hours (all P < 0.05). But the mRNA expression didn't change much; I-kappa B alpha and its mRNA expression began to decrease at 30 minutes after the reperfusion, peaking at the 2nd hour after the reperfusion (P < 0.05) and then gradually restored. Positive correlation was found between NF-kappa B P65 activity and ANS (P < 0.05), while negative correlation was shown among I-kappa B alpha, its mRNA, and ANS (P < 0.05, respectively).</p><p><strong>Conclusion: </strong>Both activated NF-kappa B and decreased I-kappa B alpha exist in the neural tissues of mice at the early stage of brain ischemia-reperfusion. {"title":"[Amplification of beta-catenin gene in the paraffin-embedded epidermal tissue].","authors":"Xiang Chen, Hong-fu Xie, Fu-wen Chen","doi":"","DOIUrl":"","url":null,"abstract":"<p><strong>Objective: </strong>To establish a method of extracting DNA from the paraffin-embedded epidermal tissue and examine the amplification of beta-catenin gene by polymerase chain reaction (PCR) analysis.</p><p><strong>Methods: </strong>Exon 3 of the beta-catenin gene was amplified by PCR in 14 paraffin-embedded Bowen's disease (BD) of various weight and years.</p><p><strong>Results: </strong>The amplification of exon 3 of beta-catenin gene was successful in all the 10 mg and 20 mg samples, but not in the 5 mg samples. {"title":"[Detection of single-nucleotide polymorphisms in insulin receptor substrate-2 gene 3'-untranslated region by denaturing high-performance liquid chromatography].","authors":"Wei-min Zeng, Shu-hua Chen, Ping Xie","doi":"","DOIUrl":"","url":null,"abstract":"<p><strong>Objective: </strong>To explore the use of denaturing high-performance liquid chromatography (DHPLC) in detecting single-nucleotide polymorphisms(SNPs) of insulin receptor substrate-2(IRS-2) gene 3'-untranslated region (3'-UTR).</p><p><strong>Methods: </strong>We detected the SNPs and mutation of IRS-2 gene 3'-UTR sequence, with polymerase chain reaction (PCR), DHPLC, single-strand conformation polymorphism (SSCP), and DNA sequence analysis respectively in 30 Type 2 diabetic subjects and 30 healthy controls.</p><p><strong>Results: </strong>The SNPs of IRS-2 gene 3'-UTR in 4 patients with Type 2 diabetes mellitus were detected by PCR-DHPLC and were identified by DNA sequencing.</p><p><strong>Conclusion: </strong>DHPLC is a rapid and automated technology, which is simpler and more accurate for detecting longer fragment of DNA than SSCP.
