The immune response of the vaccinated animals was controlled by challenge infection via aerosol technique and by examination of HAI antibodies in the serum and in the washings of lungs.</p><p>After a single aerosol vaccination a weak protection was observed only, if the vaccines contained BPE. But a second immunization with a vaccine containing BPE induced a high degree of immunity, even if a reduced amount of antigen was used for booster vaccination. After three aerosol vaccine doses in two-weeks intervals, however, we were unable to infect the immunized animals even with largest amounts of challenge virus (approximately 50000 LD<sub>50</sub>).</p><p>The addition of BPE as an adjuvant induced a significantly better protection and resulted in much higher titres of HAI antibodies in the serum and in the respiratory tract compared to mice vaccinated by the same procedure but without BPE.</p></div>","PeriodicalId":101293,"journal":{"name":"Zentralblatt für Bakteriologie. 1. Enzyme production was studied in yeast extract bouillon of different pH containing various amounts of human milk as neuraminidase inductor.</p><p>Enzyme activity was found in the bacterial sediments as well as in the culture filtrates. Since neither ultrasonic treatment nor lysozyme incubation of bacterial sediments did release reasonable amounts of enzyme, culture filtrates were used for enzyme preparation.</p><p>Neuraminidase was isolated by 40% ammonium sulfate precipitation, dialysis, concentration and repeated gel chromatography on Sephadex G-100. The enzyme possesses a molecular weight of about 33000 and a pH-optimum around 5.0. The Michaelis constants are 1.8 × 10<sup>-3</sup> M for α2 → 3 linked N-acetylneuraminic acid (NeuAc) in II<sup>3</sup>NeuAc-Lac, 3.7 × 10<sup>-3</sup> M for the α2 → 6 linkage in II<sup>6</sup>NeuAc-Lac, and 2.1 × 10<sup>-3</sup> M for the α2 → 8 linkage of II<sup>3</sup> (← 2αNeuAc8)<sub>2</sub>-Lac, respectively.</p><p>Among the different groups of naturally occurring NeuAc-containing substrates, i.e. oligosaccharides, glycolipids and glycoproteins, the enzyme exhibits its highest activity towards low molecular weight oligosaccharides. Activity is considerably lower on glycoproteins. Glycolipids (gangliosides) are only little attacked under conditions used in the test. However, there is no remarkable specificity towards one of the different linkage types of N-acetylneuraminic acid. In general, the enzyme reveals a specificity pattern similar to that found in other bacteria of low pathogenicity towards man.</p></div><div><p>Neuraminidase konnte in 32 von 38 verschiedenen Stämmen von <em>Propionibacterium acnes</em> nachgewiesen werden. Die Syntheseraten wurden in Yeast-Extrakt-Bouillon in Abhängigkeit vom pH-Wert und von als Neuraminidase-Induktor hinzugesetzter Frauenmilch bestimmt.</p><p>Enzymaktivitäten fanden sich sowohl in den Bakteriensedimenten, als auch in den Kulturfiltraten. Das Enzym aus dem Bakteriensediment konnte weder durch Ultraschall noch durch Lysozymbehandlung in nennenswerten Mengen freigesetzt werden. Dies gelang jedoch teilweise durch wiederholtes Einfrieren und Auftauen. Die Reinigung erfolgte mittels Ammoniumsulfat-Fällung (40% Sättigung), Dialyse, Konzentrierung und mehrmaliger Gel-Chromatographie an Sephadex. Das Enzym besitzt ein Molekulargewicht von ungefähr 33000 und ein pH-Optimum um 5,0. A distinct crossreaction was observed when anti-<em>icterohaemorrhagiae</em> — and anti-<em>bratislava</em>-immunsera reacted with several serotypes out of the “<em>interrogans complex</em>”. But only a weak crossreaction was found when both sera reacted with the serotyp patoc from the “<em>biflexa complex</em>”. The evaluation of the indirect immunofluorescence test was complicated by a streaky background of fluorescence. The alpha toxin is a necrotizing toxin produced in lethal amounts by strains of type A. This lethal toxin induces a characteristic subcutaneous colorless gelatineous edema when guinea pigs are infected with <em>C. novyi</em> type A.</p><p>In this paper we report further evidence on the relationship of bacteriophages to the production of alpha toxin by <em>C. novyi</em> type A. The results showed that five phage-sensitive bacterial strains that did not produce alpha toxin could be isolated from alpha toxin producing strains. These nontoxigenic strains could be infected with specific bacteriophages isolated from the different toxigenic strains of <em>C. novyi</em> type A and converted to alpha toxin production. The toxigenicity of these converted strains depended upon the continued participation of specific bacteriophages. Fifteen multiple and 3 single (OT, S, S<sub>3</sub>) antibiotic resistance patterns were obtained among 333 isolates. Of the 285 resistant strains, 133 transfered their resistant determinant to sensitive <em>E. coli</em> K12 recipients in part or as a block. None of the donors had sex pili and they were not lysed by Ms2 phage. It required about 2 h for <em>E. coli</em> K12 to acquire a resistance determinant from <em>E. coli</em> strains Nos. 295, 62 and 67 habouring resistance to oxytetracycline, streptomycin and compound of sulphonamides (S<sub>3</sub>). In microbiological and serological (O-agglutination) characterization — as well as in stability tests (agglutination in auramin and saline and heating at 100 °C) — the behaviour of the two strains was the same as that of the wild type. The two strains were found to be indistinguishable from the wild type strain also with respect to the chemical composition of their lipopolysaccharides. Thus the amount and proportion of fatty acids and sugar residues as well as the number of repeating units in the O-chain were all identical. In contrast, the isolated revertants were similar to the Rd<sub>1</sub> mutant with respect to their auxotrophic markers methionine and tryptophane, to the absence of flagella as well as to the reduced content of cyclopropane fatty acids (C<sub>17</sub>, C<sub>19</sub>). Protein analysis revealed no significant qualitative or quantitative differences between the wild type strain and the two revertants with respect to the major proteins of their outer membranes.</p><p>The sensitivity of the revertants to crystal violet, erythromycin and rifamycin SV was intermediate between the wild type and the Rd<sub>1</sub> mutant. Their temperature maximum in nutrient broth was 43 °C, the retardation in growth at this temperature corresponding to that of the Rd<sub>1</sub> mutant. At 37 °C, however, the growth rate of the revertants was identical to that of the wild-type, while that of the Rd<sub>1</sub> mutant was slower. Addition of sodium chloride to the growth medium rendered the temperature dependent behaviour of the mutants and revertants similar to that of the wild type. Studies in NMRI mice revealed that the revertants, also with regard to their virulence, occupy an intermediate position between the mutant and the wild type. Nevertheless their ability to afford protection to <em>Salmonella typhimurium</em> infection following active immunization with acetone killed cells was as high as that of the wild type.</p><p>The results show that the biologic behaviour of <em>S. typhimurium</em> is determined by the type of lipopolysaccharide it contains but also to a large extent by other cell-wall constituents.</p></div>","PeriodicalId":101293,"journal":{"name":"Zentralblatt für Bakteriologie. 1. A suspension washed three times in succession was intravenously injected into albino mice. At certain intervals liver, spleen and kidney were examinated by routine autoradiography. Intact labelled bacteria were found as early as 5 min after administration in the blood capillaries, around cells of the RES and in some nuclei of hepatocytes. Later they are detected in the cytoplasm of micro- and macrophages, around megacaryocytes and in some of their nuclei. After 6 h the labelled population sharply decreases due to a very active reproduction and mortality of the initial population.</p><p>The localisation of bacteria in liver nuclei and the spreading of S-forms of <em>E. rhusiopathiae</em> in comparison with the avirulent L-forms therefrom are discussed.</p></div><div><p>S-Formen von <em>E. rhusiopathiae</em> wurden in einem <sup>3</sup>H-Thymidin enthaltenden Medium kultiviert. Eine Bakteriensuspension wurde nach dreimaligem Waschen intravenös in Albinomäuse injiziert. Nach verschiedenen Intervallen wurden Leber, Milz und Nieren autoradiografiert. Intakte markierte Bakterien wurden schon 5 Min. nach der Injektion in den Blutkapillaren, im Bereich des RES und in einigen Kernen von Hepatozyten gefunden. Später konnten sie in dem Zytoplasma der Mikro- und Makrophagen, im Bereich von Megakariozyten und in einigen ihrer Kerne nachgewiesen werden. Nach 6 Std. fiel die Zahl markierter Zellen scharf ab als Folge einer sehr aktiven Reproduktion und des Absterbens der Zellpopulation. Cervical swabs of 74 prostitutes and 100 unselected gynecological outpatients were examined for <em>Chlamydia trachomatis</em>. The isolation of Chlamydia was carried out by <em>McCoy</em> cell culture; the inclusions were identified by <em>Giemsa</em> and <em>Lugol</em>'s stain procedure.</p><p>The isolation rate of <em>Chlamydia trachomatis</em> was 17% in the group of the prostitutes, whereas in the group of unselected women no Chlamydia were detected.</p><p>The correspondence of our findings with previous results indicates the applicability of the method used.</p></div>","PeriodicalId":101293,"journal":{"name":"Zentralblatt für Bakteriologie. 1. Standard method, <span><em>Marks'</em> method (1959)</span>, and <span><em>Becker's</em> method (1961)</span>. Out of the 231 specimens, 132 were film positive while 99 were film negative. 141 cases were found positive after cultivation on LJ medium. The number of cases found to be positive with Standard method, <em>Marks'</em> method and <em>Becker's</em> method were 91, 84, 105 respectively. Contaminations encountered with Standard method were 43, with <em>Marks'</em> method 41, and with <em>Becker's</em> method 20. 97 negatives were observed with Standard method and 106 each with <em>Marks'</em> and <em>Beckers'</em> methods. The merits and demerits of the three methods are discussed.</p></div><div><p>231 Sputumproben aus dem Ojha-Institut für Brustkrankheiten in Karachi wurden untersucht, um die Wirksamkeit und Verläßlichkeit dreier Vorbehandlungsmethoden, nämlich der Standardmethode, der <span><em>Marek's</em> Methode (1959)</span> und der Methode von <span><em>Becker</em> (1961)</span>, zu vergleichen. Bei den 231 Proben erwiesen sich 132 als film-positiv und 99 als film-negativ. 141 Proben zeigten sich nach Züchtung auf LJ-Medium als positiv. Die Anzahl der positiven Proben war folgende: Standardmethode: 91, <em>Marek's</em> Methode: 84 und die Methode von <em>Becker:</em> 105. Die Verunreinigungen bei der Standardmethode betrugen 43, 41 bei der <em>Marek's</em> Methode und 20 bei der Methode von <em>Becker</em>. 92 Proben erwiesen sich bei der Standardmethode als negativ, bei den beiden anderen Methoden betrug die Anzahl jeweils 106.
