There have also been outbreaks in some non-European countries.</p><p>The latest meningococcal meningitis epidemic recorded in the CSR (Czech Socialist Republic) occured in 1953, with the morbidity reaching 14.8 per 100 000 population. After the mid 1950's there was a gradual decline in morbidity, the lowest value (0.3 per 100000 population) being reached in 1974. Since 1975 there has been a continual rise in morbidity, the value notified for 1982 was 1.2 per 100000 population.</p><p>An analysis of the epidemiological situation in meningococcal meningitis in CSR for the past 30 years was performed. Different indicators of the onset of epidemics were evaluated. Shift in the age distribution of patients was found to be the most valuable sign of changing epidemiological situtation. An attempt was made to prognosticate mathematically its further development.</p><p>The analysis of the epidemiological data suggests that CSR is in a preepidemic situation. This finding is all the more important because a change has been found in the serogroup and serotype patterns of <em>Neisseria meningitidis</em> strains isolated from cerebrospinal fluid.</p></div><div><p>In vielen europäischen Ländern ist die in den letzten 10–15 Jahren epidemisch aufgetretene Meningokokken-Meningitis zu einem Problem geworden. Auch in einigen nichteuropäischen Ländern ist es zu Ausbrüchen gekommen. In der ČSSR ereignete sich die letzte Meningokokken-Meningitis-Epidemie 1953 mit einer Morbidität von 14,8 Fällen pro 100 000 Einwohner. Ab Mitte der fünfziger Jahre nahm die Morbidität allmählich auf den bisher niedrigsten Wert von 0,3 im Jahre 1974 ab. Scit 1975 kam es zu einem fortgesetzten Anstieg. 1982 traten 1,2 Fälle pro 100 000 Einwohner auf. Es wurde die epidemische Situation der Meningokokken-Meningitis in der CSSR in den letzten 30 Jahren untersucht und verschiedene Indikatoren für das Auftreten von Epidemien bewertet. Veränderungen in der Altersverteilung der Patienten wurden als das brauchbarste Anzeichen einer veränderten</p><p>epidemischen Situation erkannt. Es wurde der Versuch einer mathematischen Vorhersage der weiteren Entwicklung unternommen. Die Analyse der epidemiologischen Daten deutet darauf hin, daß sich die CSSR in einer vorepidemischen Situation befindet. {"title":"Evaluation of a Typing Scheme for Group A Streptococci Based Upon Bacteriocin-Like Inhibitor Production","authors":"John Robert Tagg , Diana Rae Martin","doi":"10.1016/S0174-3031(84)80043-8","DOIUrl":"10.1016/S0174-3031(84)80043-8","url":null,"abstract":"<div><p>A method of typing (P-typing) streptococci according to their production of inhibitory activity against a set of 9 indicator strains has been evaluated. Modifications to a previously described P-typing procedure have been made which improve the reproducibility of the method and also reduce the proportion of untypable strains. The test conditions, particularly medium composition, incubation temperature and aeration were found to be critical determinants of inhibitor production and must be carefully controlled. The use of certain commercial sources of Blood Agar Base and the presence of human blood in the typing medium are requirements for detection of some inhibitors, particularly for P-type 777 strains. The previously “standardized” conditions for P-typing specify incubation of the test strains aerobically at 32°C. However, in order to increase the proportion of typable strains an additional set of cultures should be incubated anaerobically. Anaerobic incubation was found to be essential for the production of bacteriocin-like inhibitors by M-type 49 strains and markedly increased inhibitor production by M-type 4 and M-type 57 strains. Also associated with anaerobic incubation was an increase in the amount of non-specific, acid-mediated inhibition, but this could be eliminated by buffering the typing medium with 0.5 % (w/v) calcium carbonate. The epidemiological validity of P-typing was tested in a survey of group A streptococcus carriage in a school and in 9 family groupings. {"title":"Examinations on the Serodiagnosis of Sarcocystis Infections II. Class-specific Immunoglobulin Responses in Mice, Pigs and Sheep","authors":"Peter J. Enzyme-linked immunosorbent assays employing conjugates specific for the relevant host immunoglobulin classes M and G were then evaluated for use in serodiagnosing these infections. Soluble antigenic extracts were prepared from cystozoites harvested from macroscopic <em>S. muris</em> and <em>S. gigantea</em> (syn. <em>S. ovifelis</em>) cysts and from microscopic <em>S. miescheriana</em> cysts. Selected sera were firstly screened in the immunoassays for false positive and negative reactions which may have resulted from competition or interference occurring between the different antibody classes. No false reactions were detected as evidenced by the reactions of the sera before and after mercaptoethanol reduction of IgM and gel fractionation of IgM and IgG. The attempted absorption of IgG from the sera with Protein A proved ineffective in all 3 host animal species. Sera from the experimentally infected animals were then tested in the immunoassays against homologous and heterologous antigens and the kinetics of specific antibody development were determined. None of the antigens proved to be species-specific although heterologous reactions were usually weaker. Specific IgM-antibodies were detected early in the courses of all infections but they did not persist. These transitory responses occurred simultaneously with parasite merogonous proliferation in mice and sheep but later in pigs. Such responses may be of some value in the serodiagnosis of acute sarcocystosis. Specific IgG-antibodies were detected later in all infected animals and elevated levels were prominent after cyst formation by the parasite had occurred. The test on suckling mice did not prove the presence of thermostable enterotoxin. After separating the concentrated culture filtrates the biologic activity appeared in fraction 1 (relative molecular mass over 100000) concerning the delayed skin test and in the fraction 2 (relative molecular mass approx. 40 000) in the rapid skin test. The activity localized in the fraction 1 can be ascribed to a lipopolysaccharide. The fraction 2 with an expressed activity in the rapid skin test showed also cytotoxic and proteolytic activity. In both fractions the presence of active antigen substances with different mobility and different antigen specificity was demonstrated.</p></div><div><p>Suspensionen lebender Bakterien, und zwar ursprüngliche sowie konzentrierte Filtrate von <em>Proteus mirabilis</em>-Kulturen, erzeugten positive Reaktionen im Hautschnelltest (vaskuläre Permeabilitäts-Reaktion), im verzögerten Test (Kombination aus hämorrhagischer Reaktion, Gefäßerweiterung und Induration) und im Mäusefußödemtest. Im Test an Babymäusen ließ sich das Vorhandensein von thermostabilem Enterotoxin nicht nachweisen. Nach Auftrennung der konzentrierten Kulturfiltrate zeigte sich die biologische Aktivität in der Fraktion 1 (relatives Molekulargewicht über 100000) im verzögerten Hauttest und in der Fraktion 2 (relatives Molekulargewicht ca. 40 000) im Hautschnelltest. Die in der Fraktion 1 lokalisierte Aktivität kann einem Lipopolysaccharid zugeschrieben werden. Die Fraktion 2 mit einer deutlichen Aktivität im Hautschnelltest zeigte auch zytotoxische und proteolytische Aktivität. {"title":"Zur Frage eines Typstammes beziehungsweise Neotypstammes für die Leptospiren","authors":"Hanspeter Mochmann , Emil Kmety","doi":"10.1016/S0174-3031(84)80045-1","DOIUrl":"https://doi.org/10.1016/S0174-3031(84)80045-1","url":null,"abstract":"<div><p>A deep review of the scientific literature concerning the history of the two oldest icterohaemorrhagiae strains is given in order to promote a decision about the legitimate neotype strain on the genus <em>Leptospira</em>.</p><p>The strain RGA was found to meet completely the requirements for a Neotype culture given by the International Code of Nomenclature of Bacteria. Its origin from a patient with Weil's disease through guinea pig passages in 1915 is well documented and its culture is unequivocally described by <em>Ungermann</em> in 1916. The strain was maintained in pure culture. Since more than 60 years the strain is used for comparative investigations in classification studies in all laboratories performing such tests.</p><p>At contrary the history of strain <em>Ictero I</em> is very incomplete. The strain was claimed by <em>Yamamoto</em> to be one of the strain isolated by <em>Inada</em> and <em>Ido</em> in 1915 originally designated as <em>Yamasaki</em>. However, the statement the maintenance of these strains because of loss of virulence was discontinued, is repeatedly mentioned in some old publications of the authors. Moreover it is reported that the strain <em>Ictero I</em> because of contamination with a fungus was recultivated after a passage through a splenectomized mouse, i. e. the strain was not maintained in a pure culture. Beside this it is hardly understandable why has the strain not been submitted to other laboratories before 1965.</p><p>The strain <em>Ictero I</em> was found to contain an additional thermolabile antigen not present in RGA. At present it is impossible to decide whether this property was already present in the original culture or developed only later, eventually after its mouse passage.</p><p>Summarizing all these facts, it must be stated that the strain <em>Ictero I</em> cannot be considered to meet all the necessary requirements of its recognition as neotype culture of the Genus Leptospira.</p></div>","PeriodicalId":79282,"journal":{"name":"Zentralblatt fur Bakteriologie, Mikrobiologie und Hygiene. 1. All the penicillinase producing strains of gonococci (23) and <em>Haemophilus influenzae</em> (36) gave positive results with PAD AC, but the 361 isolates of <em>Staphylococcus aureus</em> and <em>S. epidermidis</em> remained negative. In control mice 22 871 ± 9913 larvae were found in the whole body, whereas in mice treated with <em>P. granulosum</em> KP 45 on the day of inoculation 9450 larvae ± 8244 larvae were counted (p < 0.01). Immunomodulation 7 days before or 7 days after <em>T. spiralis</em> inoculation gave less pronounced effects but the decreasing number of larvae was still significant. It is suggested that the above effects are due to an activation of macrophages.</p></div><div><p>Im Verlauf einer experimentellen Trichinose wurden verschiedene Immunsuppressionseffekte beobachtet. In vorliegender Arbeit wurde daher versucht, diese parasitäre Erkrankung mit Hilfe eines potenten bakteriellen Immunodulators (<em>Propionibacterium granulosum</em> — Stamm KP 45) positiv zu beeinflussen. In der Kontrollgruppe wurden nach oraler Gabe von 200 lebenden <em>Trichinella spiralis</em>-Lawen im Durchschnitt 22 871 ± 9913 Larven je Maus gezählt. Bei gleichzeitiger i. p.-Gabe von 0.5 oder 2.0 mg <em>P. granulosum</em> KP 45 verminderte sich diese Larvenzahl signifikant auf durchschnittlich 9450 ± 8244 je Maus (p < 0,01). Eine Immunomodulation 7 Tage vor oder nach der <em>T. spiralis</em>-Inokulation zeigte ebenfalls deutliche, jedoch schwächere Effekte. The activity of the RNA polymerase of this virus preparation was determined by incorporating <sup>3</sup>H-UMP in acid insoluble material following preincubation of the virus with the nonionic detergens Nonidet P-40 for 15 min at 32°C.</p><p>The influence of different concentrations was investigated of dinucleotid, NaCl, MgCl<sub>2</sub>, Nonidet P-40 and different incubation temperatures.</p><p>Optimal incorporation rates were found at following conditions: 0.2 mM dinucleotid ApG, 150 mM sodium chloride and 8 mM magnesium chloride by concentration of ions, 0.25–0.5% detergens Nonidet P-40 as well as a temperature of incubation of 32°C. Für die experimentelle Pyelonephritis weiblicher Wistarrarten nach transurethraler Infektion werden Standardisierungsmöglichkeiten in der akuten Infektionsphase gezeigt.</p><p></p><ul><li><span>1.</span><span><p>Die Höhe der renalen Keimzahlen und Infektionsraten steht in positiver Korrelation zum Instillationsvolumen und dessen Bakteriendichte.</p></span></li><li><span>2.</span><span><p>Wistarratten-Stämme verschiedener Versuchstierzuchtanstalten weisen eine unterschiedliche Infektionsresistenz auf. So geht die <em>E. coli</em>-Pyelonephritis bei weiblichen Wistarratten vom Stamm Han: WIST leichter an als bei dem Stamm Bor: WIST.</p></span></li><li><span>3.</span><span><p>Im Anschluß an die Sektion der Tiere können infizierte Nieren für mindestens 4 Wochen bei −30°C oder in flüssigem N2 gelagert werden, da sich die renalen Keimkonzentrationen nicht ändern. In Nierenhomogenaten fällt die Keimzahl jedoch rasch ab.</p></span></li><li><span>4.</span><span><p>Während der ersten 4 Tage post infectionem ist der Bakteriengehalt der Nieren relativ konstant.
