The growth cycle of the organism was investigated in different tissues and fluids derived from embryonated eggs following inoculation with a suspension of <em>L. monocytogenes</em> equivalent to the LD<sub>50</sub>. Eggs receiving doses of 100 and 1000 times the LD<sub>50</sub> were used to examine the effect of high doses on the pathogenesis and growth of <em>Listeria in ovo</em>.</p><p>The pattern of growth of the organism in embryonic blood showed two distinct peaks and correlated with these was the development of large and small pock lesions on the chorioallantoic membrane. Bacterial growth in the internal organs exhibited a single peak. Histological and electron microscopic evidence indicated that the primary cellular damage was due to a soluble haemolysin present prior to the establishment of the organism within the tissues.</p></div><div><p>Es wurde der LD<sub>50</sub>-Wert bei 15 Tage alten Hühnerembryonen bestimmt, die durch Injektion von <em>Listeria monocytogenes</em> in die Allantois infiziert worden waren. Der Wachstumszyklus des Erregers wurde nach Beimpfung mit einer dem LD<sub>50</sub>-Wert entsprechenden Suspension von <em>L. monocytogenes</em> an verschiedenen aus embryonierten Eiern gewonnenen Geweben und Flüssigkeiten untersucht. Eier, die Dosen von 100 bzw. 1000 LD<sub>50</sub> erhielten, dienten zur Untersuchung des Einflusses hoher Dosen auf die Pathogenese und auf das Wachstum von Listerien in <em>ovo</em>.</p><p>Das Wachstumsmuster des Erregers im Embryonalblut zeigte zwei deutliche Peaks, die</p><p>mit der Entwicklung großer und kleiner Pockenläsionen auf der Chorioallantoismembran korrelierten. Das Bakterienwachstum in den inneren Organen zeigte nur einen einzigen Peak. Talbot","doi":"10.1016/S0174-3031(84)80023-2","DOIUrl":"10.1016/S0174-3031(84)80023-2","url":null,"abstract":"<div><p>Conventional tests for the presumptive identification of <em>Streptococcus pneumoniae</em> from culturally similar non-pneumococcal alpha hemolytic streptococci include optochin sensitivity, bile solubility and inulin fermentation. These tests require up to 24 h for completion. In this paper we report the evaluation of a simple and rapid coagglutination procedure that requires less than 5 min. Of the 150 clinical isolates of pneumococci that were identified by conventional methods, all were correctly recognized by the Phadebact coagglutination technique within one minute. Forty strains of non-pneumococcal alpha hemolytic streptococci were also tested to determine the specificity of the method. Of these, 32 (80 %) did not react in either the typing or control reagents, whereas 8 (20%) showed coagglutination in the control. The number of attached bacterial cells was determined by measuring the light emission resulting from the reaction between firefly luciferase and ATP present in adhered staphylococcal cells. It was shown that staphylococci attach to synthetic polymers within a few minutes, although one hour incubation is required to reach a constant maximum value of attached cells. Ten different synthetic polymers and five <em>Staphyloccocus epidermidis</em> strains were investigated in our study. The relationship between surface properties of polymers and bacterial attachment was studied. Various physicochemical parameters of synthetic polymers and bacteria were determined (contact angle, surface tension). It was demonstrated that bacterial attachment decreases with decreasing contact angle and with increasing surface tension of synthetic materials. Modifications of surface charge and hydrophobicity of solid materials were also investigated. Leptospires elicited the production of both IgM and IgG classes of antibody, with IgG levels persisting for much longer than IgM. A comparison of MAT and EIA indicated that the detection of specific IgM by EIA was potentially useful in distinguishing between past and recent infection in pigs. Agglutinins were also detected in the urine of infected animals but these antibodies could not be detected by EIA.</p></div><div><p>Nach experimenteller Infektion von Schweinen mit Leptospiren der Serovar pomona wurde die Antikörperbildung mit dem Enzymimmunoassay (EIA) und dem Mikroagglutinationstest (MAT) untersucht.</p><p>Durch die Leptospiren wurde sowohl die Bildung von IgM- wie auch von IgG-Antikörpern hervorgerufen.</p><p>IgG-Titer blieben länger bestehen als IgM-Titer.</p><p>Ein Vergleich zwischen MAT und EIA zeigte an, daß der Nachweis spezifischer IgM-Antikörper mit dem EIA möglicherweise für die Untersuchung frischer und zurückliegender Infektionen bei Schweinen verwendet werden kann.</p><p>Agglutinine ließen sich im Urin der infizierten Tiere nachweisen. Out of 40 sarcoidosis lymph nodes examined, 31 nodes (77.5%) showed a positive culture of <em>P. acnes</em>, while 38 of 180 (21.1%) non-sarcoidosis tissues revealed positive results, the difference being significant (p < 0.001). Among non-sarcoidosis tissues, the intra-thoracic lymph nodes demonstrated a positive culture in low percentages, while intra-abdominal lymph nodes indicated no positive culture. The possible role of this microorganism in the etiology of sarcoidosis is discussed.</p></div><div><p>Aus Lymphknoten von Patienten mit Sarcoidose und Lymphknoten oder anderen Geweben von Patienten mit anderen Erkrankungen wurde <em>Propionibacterium acnes</em> isoliert. Dabei wurde darauf geachtet, eine Kontamination mit <em>P. acnes</em>, einem normalerweise auf der Haut vorkommenden Keim, zu vermeiden. Von 40 Lymphknoten von Sarcoidose-Patienten zeigten 31 Proben (77.5%) <em>P. acnes</em>-positive Kulturen, während aus den Gewebeproben von Patienten mit anderen Erkrankungen in 38 von 180 Fällen (21.1%) <em>P. acnes</em> isoliert werden konnte. Diese Differenz ist signifikant (p < 0.001). Von den Proben der Sarcoidosefreien Patienten zeigten die intrathorakalen Lymphknoten positive Kulturen, während aus den intraabdominalen Lymphknoten <em>P. acnes</em> nicht isoliert werden konnte. On the whole, 45 fish proved positive, from which 71 strains were taken. Among the culture media analyzed the best results were obtained with the new enrichment media represented by two of the nutrient broth (Difco) pH 8.3; one supplemented with 2 g% of NaCl and 500 mg% eosin Y, and another supplemented with 125 mg% metachrome yellow II RD. {"title":"Bacteriuria screening and antimicrobial susceptibility testing of aerobic bacteria by an electrochemical method","authors":"Jürgen Strassburger , Friedrich-Wilhelm Tiller","doi":"10.1016/S0174-3031(84)80022-0","DOIUrl":"10.1016/S0174-3031(84)80022-0","url":null,"abstract":"<div><p>A method is described for detecting significant bacteriuria and determination of minimal inhibition concentrations (MIC's) of aerobically growing bacteria by using electrochemical electrodes to measure changes of oxygen tensions in liquid nutrient media resulting from bacterial growth.</p><p>Urine specimens (n = 577) were screened electrochemically, parallel investigations were performed by standard culture methods and by photometrical measurements.</p><p>All the specimens showing significant bacteriuria in standard culture were selected within 3.5 h by the electrochemical technique.</p><p>An oxygen index OI was introduced which quantitatively reflects changes in oxygen tension of nutrient media during growth. OI shows good agreement with extinction and light scattering indices, respectively.</p><p>On the basis of OI as a parameter of inhibited and uninhibited growth a correlation between OI and MIC's of aerobically growing bacteria was found.</p><p>The electrochemical method provides an useful aid for rapid, preliminary antimicrobial susceptibility testing and definite bacteriuria screening.</p><p>The application of this method in bacteriological urine diagnostics significantly reduces laboratory work and costs, and can be recommended for the screening of urine specimens to exclude negative specimens from further processing.</p></div><div><p>Eine Methode zur Ermittlung signifikanter Bakteriurien sowie zur Bestimmung minimaler Hemmkonzentrationen (MHK) aerob wachsender Bakterien wird beschrieben, bei der durch bakterielles Wachstum hervorgerufene Änderungen des Sauerstoffpartialdruckes in flüssigen Nährmedien mit Hilfe elektrochemischer Elektroden gemessen werden.</p><p>Es wurden 577 Urinproben elektrochemisch untersucht, vergleichende Untersuchungen wurden mit herkömmlichen Kulturverfahren sowie photometrischen Messungen durchge- führt. Alle Urinproben, die im Kulturverfahren eine signifikante Bakteriurie zeigten, konnten mit der elektrochemischen Technik innerhalb von 3.5 h bestimmt werden.</p><p>Ein Sauerstoff-Index OI wurde eingeführt; er widerspiegelt quantitativ die Änderungen des Sauerstoffpartialdruckes in den Nährmedien während des bakteriellen Wachstums.</p><p>OI korreliert gut sowohl mit Extinktions- als auch mit Streulicht-Indices.</p><p>Auf der Grundlage von OI als einem Parameter für gehemmtes und nicht gehemmtes bakterielles Wachstum wurde eine Korrelation zwischen OI und MHK aerob wachsender Bakterien gefunden. Die elektrochemische Methode stellt sich als ein geeignetes Verfahren für eine schnelle, vorläufige Erreger-Resistenzbestimmung sowie für ein definitives Bakteriurie-Screening dar.</p><p>Die Anwendung dieser Methode in der bakteriologischen Harnwegsdiagnostik vermindert deutlich Laborarbeit und Kosten. Augmentin is a new formulation of the broad-spectrum β-lactam-antibiotic amoxicillin together with the β-lactamase-inhibitor clavulanic acid.</p><p>It was demonstrated that 88 % of all isolates tested were sensitive to Augmentin, 9 % were resistant. 88 % of all <em>Pseudomonas aeruginosa</em> strains fell in the “resistant” category. Only 1/71 anaerobes and 15/286 staphylococci were classified as resistant to Augmentin.</p></div><div><p>1417 Bakterienstämme aus klinischem Untersuchungsmaterial wurden auf ihre Empfindlichkeit gegenüber Augmentin getestet. Augmentin ist eine neue Kombination aus dem Breitspektrumantibiotikum Amoxicillin und dem β-Laktamase-Inhibitor Clavulansäure.</p><p>88 % aller getesteten Stämme erwiesen sich als Augmentin-sensibel. 9% waren resistent, darunter finden sich 88 % aller <em>Pseudomonas aeruginosa</em>-Stämme.
