Four volunteers received a carbohydrate-reduced, the other 4 persons a carbohydrate-enriched diet. Clinical investigations on the development of plaque and gingivitis and bacteriological examinations of the supragingival microflora were performed simultaneously. The change in the composition of the supragingival plaque-microflora were nearly independent of the amount of food-carbohydrate. In the beginning of the experiment all volunteers had an almost pure streptococcal microflora but during the course of the experiment, the supragingival flora developed to a complex microflora with 50% anaerobes. The reduction of the number of streptococci was accompanied by an increase in the number of Gram-negative rods and actinomycetes. The development of gingivitis in persons with carbohydrate-enriched food was more rapid than in persons with carbohydrate-reduced food. It was striking that the occurence of clinical symptoms correlated with the increase of the number of <em>Actinomyces viscosus</em> in the plaque material.</p></div>","PeriodicalId":79282,"journal":{"name":"Zentralblatt fur Bakteriologie, Mikrobiologie und Hygiene. 1. In comparison to the controls the growth of bacteria was reduced but not stopped. If human blood is added to the culture medium, staphylococci multiply in the presence of all colchicine concentrations tested. In contrast <em>E. coli</em> grows only at a concentration of colchicine of 0.02–0.03%.</p><p>Indomethacin dissolved in ethanol inhibited clearly the phagocytosis of <em>S. aureus</em> and <em>E. coli</em>. This inhibition was significant for concentrations of 2 × 10<sup>-4</sup> and 1 × 10<sup>-4</sup> mol/l. The behavior of the tested bacteria with respect to growth and multiplications was not affected. Indomethacin, however, retarded the bacterial growth in concentrations from 2 × 10<sup>-4</sup> up to 3.75 × 10<sup>-5</sup> mol/l. The multiplication of <em>E. coli</em> was stopped by indomethacin at a concentration of 2 × 10<sup>-4</sup> mol/l. If human blood is added to the phagocytosis test system with <em>S. aureus</em> and indomethacin, the bacteria will show a little reduction in the rate of the multiplication in concentrations of the compound from 2 × 10<sup>-4</sup> up to 3.75 × 10<sup>-5</sup> mol/l. At a concentration of 2 × 10<sup>-4</sup> mol/l, the growth of <em>E. coli</em> was inhibited.</p><p>It is concluded that for the inhibition of phagocytosis — f. e. in blood culture systems — the concentration of colchicin must be 0.03–0.02%; for indomethacin an optimal concentration of 1 × 10<sup>-4</sup> mol/l is recommended.</p></div>","PeriodicalId":79282,"journal":{"name":"Zentralblatt fur Bakteriologie, Mikrobiologie und Hygiene. 1. Three doses of vaccine induced anti-HBs antibodies in 94 to 100% of the vaccinees. Different batches and doses of vaccine did not significantly influence the seroconversion rates at the end of the vaccine trial. Higher doses were followed by more rapidly and more vigorously occurring anti-HBs-levels. Significant differences were noticed between the 40 μg- und the 10- or 8 μg-doses. Variation of schedules (0-1-6 or 0-1-5) did not result in significant differences. Batch-no. I/B 070881 induced significant lower anti-HBs levels than batch-no. II/310781. Females developed significant higher anti-HBs values than males, younger vaccinees higher than older vaccinees.</p><p>Rate zonal ultracentrifugation analysis of anti-HBs-positive sera collected after vaccination regularly revealed anti-HBs of the IgM class in case of early and high immune response to the vaccine. Individuals characterized by a late onset of anti-HBs production developed anti-HBs antibodies of the IgG class only.</p><p>The vaccine induced an anti-HBs response in 64 to 75% of hemodialysis patients. The conversion rates and the achieved anti-HBs levels were significantly higher in young and/or female patients than in old and/or male patients.</p></div><div><p>Die experimentelle deutsche Hepatitis B Vakzine wurde bei 343 gesunden Impflingen hinsichtlich ihrer Immunogenität überprüft.</p><p>Die dreimalige Gabe des Impfstoffes induzierte bei 94 bis 100% der Impflinge anti-HBs-Antikörper. Die Verwendung unterschiedlicher Impfstoffchargen und -dosen ließ nach Abschluß der Impfserie keine signifikant unterschiedlichen Serokonversionsraten erkennen.</p><p>Höhere Impfstoffdosen führten aber zu einer schnelleren und stärkeren anti-HBs-Bildung. Die Immunantwort war nach 40 μg-Dosen signifikant stärker als nach 10 μg- oder 8 μg-Dosen.</p><p>Eine Veränderung des Impfschemas (3. Impfung nach 5 statt 6 Monaten) hatte keine signifikanten Unterschiede zur Folge. Die Impfstoffcharge I/B 070881 induzierte signifikant niedrigere anti-HBs-Spiegel als die Charge II/310781.</p><p>Bei Frauen wurden signifikant höhere anti-HBs-Werte beobachtet als bei Männern. Auch das Lebensalter der Impflinge spielte eine signifikante Rolle bei der Immunantwort.</p><p>Die Analyse anti-HBs-haltiger Seren nach ratenzonaler Ultrazentrifugation ergab bei früher und starker Immunantwort regelmäßig auch anti-HBs-Antikörper vom IgM-Typ. Bei spät einsetzender anti-HBs-Bildung wurden dagegen nur anti-HBs-Antikörper der IgG-Klasse entdeckt.</p><p>Der Hepatitis B-Impfstoff führte nur bei 64 bis 75% der 166 Dialysepatienten zu einer meßbaren anti-HBs-Antwort. A crude enzyme preparation was obtained by 20–80% ammonium sulfate precipitation of the broth. The β-glucuronidase was purified using DEAE cellulose column chromatography, gel filtration on Sephadex G-200, and affinity chromatography on Sepharose 4B-bound glucuronolactone. We obtained two kinds of β-glucuronidase. Properties of the purified β-glucuronidase I were an optimum pH of 7.2, a pH stability range below 7.0, and a molecular weight of 115,000. The purified β-glucuronidase II had an optimum pH of 6.0, pH stability at around 6.0, and a molecular weight of 195,000.</p><p>Cu<sup>++</sup> and Hg<sup>++</sup> were strong inhibitors, which inhibition was restored by cysteine. EDTA did not influence enzyme activity. The Michaelis constants of β-glucuronidase I and β-glucuronidase II for p-nitrophenyl glucuronide were 1.25 × 10<sup>-3</sup> M and 4.17 × 10<sup>-4</sup> M, for naphthol AS-BI glucuronide 1.35 × 10<sup>-4</sup> M and 1.14 × 10<sup>-4</sup> M, and for phenolphthalein glucuronide 7.46 × 10<sup>-5</sup> M and 2.50 × 10<sup>-4</sup> M.</p></div><div><p>Glatte Kolonien von <em>Clostridium perfringens</em> (Hobbs' Typ 4), das aus dem Stuhl eines Patienten isoliert worden war, wurden in GAM-Bouillon 5 Tage bei 37°C gezüchtet, um maximale Ausbeuten an extrazellulären β-Glucuronidase zu erhalten. Ein Rohenzympräparat wurde durch Fällung mit 20–80% Ammoniumsulfat aus der Bouillon gewonnen.</p><p>Die β-Glucuronidase wurde gereinigt mittels DEAE-Cellulose-Säulenchromatographie, Gelfiltration über Sephadex G 200 und Affinitätschromatographie an Sepharose-4B-gebundenem Glucuronolacton.</p><p>Wir erhielten 2 Arten von β-Glucuronidase. The bacteria were inoculated on a 1,5% agarose gel in Petri plates of 4 cm ∅ or gel discs of 7 mm ∅, containing 0,1 % of K-hyaluronate as well as nutritient medium, and were incubated for 2–20 h at 37°C in a moist chamber. Subsequently some ml of a 10% solution of cetylpyridiniumchloride were poured on the gel to precipitate the polymere hyaluronate. If the hyaluronate was depolymerized by hyaluronidase, a translucent area was visible around the colonies. We found out, that a gel layer of 1 mm was sufficient to detect the small amounts of hyaluronidase, which were produced by bacteria within an incubation time of 2 h. These results were confirmed by incubation for 20 h and in some cases 36 h.</p><p>The hyaluronidase production by different anaerobic <em>Clostridium</em> strains was always proved after a 20 h growth period.</p><p>The bacteria were inoculated with the whole loop of a self made platin sowing wire loop. By this method quantitative differences of hyaluronidase activities between different strains of bacteria could be detected.</p></div><div><p>Zum Nachweis der Hyaluronidase-Aktivität wurden 339 aerobe und anaerobe Bakterienstämme untersucht.</p><p>Die Bakterien wurden hierzu auf ein in Petrischälchen abgefülltes bzw. aus Scheibchen von 7 mm ∅ bestehendes l,5%iges Agarose-Gel geimpft, das 0,1 % K-Hyaluronat als Enzymsubstrat und Nährmedium enthielt. Bei einer nur 1 mm dicken Gelschicht genügte für alle untersuchten Bakterienstämme ein zweistündiges Wachstum, die während dieser Zeit gebildete Hyaluronidase mittels 10%iger Cetylpyridiniumchlorid-Lösung nachweisen zu können. Nur bei den anaerob wachsenden Clostridien wurde das Hyaluronidase-Bildungsvermögen erst nach 20stündiger Wachstumsdauer überprüft.</p><p>Mit einer selbstgefertigten Impföse gelang es, gleich breite Impfstriche zu ziehen, denen zufolge Unterschiede in der Hyaluronidase-Produktion zwischen den einzelnen Stämmen erkennbar wurden.</p></div>","PeriodicalId":79282,"journal":{"name":"Zentralblatt fur Bakteriologie, Mikrobiologie und Hygiene. 1. Hexon was found in 44% of about 100 patients with adenovirus conjunctivitis, proven by virus isolation, antihexon in 53%, either one or both in 69% of the conjunctival swabs from the acute phase. Hexon was found in the acute phase more frequently, antihexon in the convalescent phase. Thre results did not contradict the assumption of animals being a source of human infections. In general, these strains of the serovars 0:3 and 0:9 possessed a virulence plasmid with a molecular weight of 46 Mdal (± 3 Mdal), irrespective of their origin from humans or domestic animals with latent infections. There was a consistent correlation of the presence of the virulence plasmid and of positive reactions in <em>Laird's</em> and <em>Cavanaugh's</em> autoagglutination test (1980) (Tab. 1): The strains harbouring this plasmid were calcium dependent on MOX agar (18) as well. These strains were capable of colonizing the intestine of mice in a modified mouse diarrhea model (Fig. 1). In these latently infected mice low titers of species-specific antibodies were detected. It was possible to select plasmid harbouring substrains out of heterogenic strains, which had undergone conversion to plasmidless isogenic variants when applying the modified mouse diarrhea model. In a newly developed “macrophage-virulence-test”, strains harbouring the virulence plasmid were able to multiply within the macrophage of mice (Fig. 4, 5) with cytopathogenic effects. In a modified <em>Sereny</em>-test European strains of the serovars 0:3 and 0:9 were shown to be invasive in the conjunctival epithelium of guinea pigs' eyes as an additional plasmid-associated property (Fig. 3). European strains of atypical biochemical or serological patterns (among them <em>Y. intermedia, Y. frederiksenii</em>) as well as some strains of the serovars 0:3 and 0:9 did not harbour the virulence plasmid. These plasmidless strains were altogether negative regarding the pathogenic properties mentioned above. On average 41% of cefmenoxime-moderately susceptible (MIC between 2 and 32 μg/ml) or cefmenoxime-resistant (MIC &gt; 32 μg/ml) strains were inhibited by synergistic, and 23,5% by partial synergistic cefmenoxime-aminoglycosides combinations. Against cefmenoxime-susceptible strains, these rates of synergy or partial synergy were twofold lower, but only one isolate was not susceptible to these combinations. The distribution patterns of their susceptibility to Hg, Cd and As clearly revealed two peaks, but only a single peak of resistance to Pb. The frequencies of resistance to Hg, Cd, As, SM, TC., CP and KM were 31.8, 94.2, 57.8, 51.3, 44.8, 35.7 and 28.9%, respectively. And we selected 88 mercury resistant strains, and they were tested for their ability to transfer the resistance to the mercury and drug sensitive recipients of <em>E. coli</em> K12-ML1410-Nx and <em>E. coli</em> JE17-Rif. Among 88 strains of Hg resistant <em>Citrobacter</em>, 80 R plasmids with Hg resistance could be demonstrated. It should be noted that transferable R plasmids with Hg resistance were demonstrated in 91% of the Hg-resistant isolates. Furthermore, we tested the volatilization of mercury by strains containing these mercury resistance plasmids, by using radioactive <sup>203</sup>Hg<sup>2+</sup>. All of these isolates of <em>Citrobacter</em> have volatilization activity of Hg<sup>2+</sup>. The subjects of this research include: Soil of potted indoor plants as a habitat for aspergilli; exposure of sensitive patients sufferring from different basic diseases to <em>Aspergillus</em> spores; polyene-producing streptomycetes beside aspergilli in the rhizosphere of potted plants and their role; and ecological and clinical significance of atypical <em>Aspergillus</em> strains isolated from clinical specimens.
