Methods The Cancer Genome Atlas was employed to analyze the mRNA levels of PSMB8 in ccRCC and normal tissue,and the expression levels of PSMB8 in ccRCC tissue and cells were determined by real-time quantitative PCR,Western blotting,and immunohistochemistry.Furthermore,the cell lines with stable overexpression and knockdown of PSMB8 were constructed.The CCK-8 assay and colony formation assay were employed to examine the cell proliferation,and the wound healing assay and Transwell assay were employed to examine the invasion and migration of cells.Kyoto Encyclopedia of Genes and Genomes pathway enrichment was performed to analyze the co-expressed genes of PSMB8.Western blotting was used to measure the phosphorylation levels of the proteins in the MEK/ERK signaling pathway.Finally,the rescue experiment was carried out with the ERK agonist C16-PAF. Results Compared with the normal tissue,the ccRCC tissue showed up-regulated mRNA and protein levels of PSMB8 (both <i>P</i><0.001),which were associated with the TNM stage of patients with ccRCC (<i>P</i><0.001).Compared with the negative control group,overexpression of PSMB8 promoted the proliferation (<i>P</i>=0.021,<i>P</i>=0.039),migration and invasion (all <i>P</i><0.001) of 786-O and ACHN cells,and the knockdown of PSMB8 inhibited the proliferation (<i>P</i>=0.022,<i>P</i>=0.005),migration and invasion (all <i>P</i><0.001) of 786-O and ACHN cells.The pathway enrichment analysis of co-expressed genes of PSMB8 predicted the mitogen-activated protein kinase signaling pathway (<i>P</i><0.001).After the knockdown of PSMB8,786-O and ACHN cells showed lowered phosphorylation levels of MEK1/2 (<i>P</i>=0.017,<i>P</i>=0.016) and ERK1/2 (<i>P</i>=0.010,<i>P</i>=0.040) and down-regulated transcription levels of ERK downstream factors c-Myc (<i>P</i>=0.043,<i>P</i>=0.038),c-Fos (<i>P</i>=0.025,<i>P</i>=0.008),and CyclinD1 (<i>P</i>=0.006,<i>P</i>=0.047).Compared with the ERK agonist C16-PAF group,the PSMB8 knockdown + C16-PAF group showed inhibited proliferation (<i>P</i>=0.003,<i>P</i>=0.002),migration and invasion (all <i>P</i><0.001) of 786-O and ACHN cells. Methods Wild-type C57 adult mice with normal cognitive function were selected,and DEACMP was modeled by static inhalation of carbon monoxide.The mice were randomized into three groups:DEACMP,control,and NBP.The NBP group was administrated with NBP suspension at 6 mg/kg by gavage for 21 days,and the DEACMP and control groups were administrated with the same amount of vegetable oil by gavage.The hippocampal injury was observed by HE staining.The protein level of ionized calcium-binding adapter molecule 1 (IBA1) was determined by Western blotting,and the levels of downstream inflammatory cytokines were measured by ELISA. Results Compared with the control group,the DEACMP and NBP groups showed prolonged escape latency (<i>P</i>=0.001,<i>P</i>=0.029),reduced nerve cells (<i>P</i>=0.001,<i>P</i>=0.035),up-regulated expression of IBA1 (<i>P</i>=0.001,<i>P</i>=0.042),increased mean fluorescence intensity of IBA1 (<i>P</i>=0.001,<i>P</i>=0.021),and elevated levels of tumor necrosis factor-α (TNF-α) (<i>P</i>=0.002,<i>P</i>=0.024),interleukin (IL)-6 (<i>P</i>=0.001,<i>P</i>=0.015),and IL-1β (<i>P</i>=0.001,<i>P</i>=0.023).Compared with the DEACMP group,the NBP group showed shortened escape latency (<i>P</i>=0.025),increased nerve cells (<i>P</i>=0.039),down-regulated expression of IBA1 (<i>P</i>=0.035),decreased average fluorescence intensity of IBA1 (<i>P</i>=0.031),and lowered levels of TNF-α (<i>P</i>=0.028),IL-6 (<i>P</i>=0.037),and IL-1β (<i>P</i>=0.034). {"title":"[Effect of Sakuranetin on Microglia-Mediated Neuroinflammation After Spinal Cord Injury].","authors":"Lin-Yu Xiao, Yue Chen, Ting Duan, Yang Sun, Yi-Bo Xu, Ya-Jing Zhao, Xue Song, Xing-Zhou Yan, Jian-Guo Hu","doi":"10.3881/j.issn.1000-503X.16087","DOIUrl":"https://doi.org/10.3881/j.issn.1000-503X.16087","url":null,"abstract":"<p><p>Objective To investigate the effects of sakuranetin (SK) on motor functions in the mouse model of spinal cord injury (SCI) and decipher the mechanism.Methods Fifty-four C57BL/6J mice were randomized into sham,SCI,and SK groups.The mice in the sham group underwent only laminectomy at T9,while those in the SCI and SK groups were subjected to spinal cord contusion injury at T9.Behavioral tests were conducted at different time points after surgery to evaluate the motor functions of mice in each group.The pathological changes in the tissue were observed to assess the extent of SCI in each group.The role and mechanism of SK in SCI were predicted by gene ontology (GO) and Kyoto encyclopedia of genes and genomes (KEGG) enrichment analyses.Reverse transcription real-time fluorescence quantitative PCR,ELISA,and immunofluorescence were employed to evaluate the inflammation and activation of microglia in SCI mice.BV2 cells in vitro were classified into control (Con),lipopolysaccharide (LPS),and LPS+SK groups.The effects of SK intervention on the release of inflammatory cytokines and the activation of BV2 cells were evaluated.Furthermore,the phosphatidylinositol-3-kinase(PI3K)/protein kinase B (AKT) signaling pathway activator insulin-like growth factor-1 (IGF-1) was used to treat the SK-induced BV2 cells in vitro (SK+IGF-1 group),and SK was used to treat the IGF-1-induced BV2 cells in vitro (IGF-1+SK group).Western blotting was conducted for molecular mechanism validation.Results Behavioral tests and histological staining results showed that compared with the SCI group,the SK group exhibited improved motor abilities and reduced area of damage in the spinal cord tissue (all P<0.001).The GO enrichment analysis predicted that SK may be involved in the inflammation following SCI.The KEGG enrichment analysis predicted that SK regulated the PI3K/Akt pathway to exert the neuroprotective effect.The results from in vitro and in vivo experiments showed that SK lowered the levels of tumor necrosis factor-α,interleukin-6,and interleukin-1β and inhibited the activation of microglia (all P<0.05).The results of Western blotting showed that SK down-regulated the phosphorylation levels of PI3K and Akt (all P<0.001) and inhibited the IGF-1-induced elevation of PI3K and Akt phosphorylation levels (all P<0.001).Conversely,IGF-1 had the opposite effects (P=0.001,P<0.001).The results of reverse transcription real-time fluorescence quantitative PCR,ELISA,and immunofluorescence showed that the SK+IGF-1 group had higher levels of inflammatory cytokines and more activated microglia than the SK group(all P<0.05).Conclusion SK may suppress the activation of the PI3K/Akt pathway to inhibit the inflammation mediated by SCI-induced activation of microglia,ameliorate the pathological damage of the spinal cord tissue,and promote the recovery of motor functions in SCI mice.</p>","PeriodicalId":6919,"journal":{"name":"中国医学科学院学报","volume":null,"pages":null},"PeriodicalIF":0.0,"publicationDate":"2024-08-30","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"142103277","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Methods Thirty patients undergoing cardiac transcatheter closure under elective intravenous anesthesia were included in this study.Paired <i>t</i>-tests were performed to compare the mRNA levels of the genes encoding circadian locomotor output cycles kaput(CLOCK),brain and muscle ARNT-1 like protein-1(BMAL1),cryptochrome 1(CRY1),and period circadian clock 2(PER2),the Munich Chronotype Questionnaire(MCTQ)score,and the Pittsburgh Sleep Quality Index(PSQI)score before and after anesthesia.Multiple stepwise regression analysis was performed to screen the factors influencing sleep chronotype and PSQI total score one week after surgery. Methods According to the random number table method,SD male rats were assigned into control,sleep deprivation,low-,medium-,and high-dose Schisandrae Chinensis Fructus lignans,and atomoxetine hydrochloride groups,with 8 rats in each group.The rats in other groups except the control group were subjected to sleep deprivation by treadmill exercise for 3 d.During the deprivation period,each administration group was administrated with the corresponding drug by gavage,and a 5-9 hole tester was used to test the alertness performance of rats in each group. Furthermore,other SD male rats were selected and randomized into control,sleep deprivation,Schisandrae Chinensis Fructus lignans (67.2 mg/kg) and atomoxetine hydrochloride groups,with 10 rats in each group.The rats were modeled with the sleep deprivation method the same as that above and administrated with corresponding agents.ELISA was employed to measure the serum level of orexin A in each group of rats.The protein levels of c-Fos,orexin receptor 1,and orexin receptor 2 in the prefrontal cortex of rats in each group were observed by immunofluorescence and Western blotting. Results Compared with the control group,sleep deprivation reduced the choice accuracy (<i>P</i><0.001) and increased the omission responses,omission percent,and mean correct response latency (<i>P</i>=0.002,<i>P</i>=0.003,<i>P</i>=0.020).Compared with the sleep deprivation group,medium- and high-dose Schisandrae Chinensis Fructus lignans and atomoxetine hydrochloride improved the alertness of rats,as demonstrated by the increased choice accuracy (<i>P</i>=0.001,<i>P</i>=0.006,<i>P</i><0.001) and reduced omission responses (<i>P</i>=0.001,<i>P</i>=0.001,<i>P</i><0.001),omission percent (<i>P</i>=0.001,<i>P</i>=0.002,<i>P</i><0.001),and mean correct response latency (<i>P</i>=0.018,<i>P</i>=0.003,<i>P</i>=0.014).Compared with the control group,the sleep deprivation group showed elevated level of orexin A in the serum (<i>P</i><0.001),up-regulated expression of c-Fos (<i>P</i><0.001),and down-regulated expression of orexin receptor 1 (<i>P</i>=0.037) in the prefrontal cortex.Compared with the sleep deprivation group,Schisandrae Chinensis Fructus lignans (67.2 mg/kg) and atomoxetine hydrochloride lowered the orexin A level in the serum (<i>P</i>=0.005,<i>P</i>=0.029),down-regulated the expression of c-Fos (<i>P</i>=0.028,<i>P</i>=0.036),and up-regulated the expression of orexin receptor 1 (<i>P</i>=0.043,<i>P</i>=0.013) in the prefrontal cortex. Methods The clinical data of 10 patients undergoing surgical treatment of extracranial supra-aortic aneurysms from May 2019 to November 2023 in the Department of Vascular Surgery of Beijing Tiantan Hospital affiliated to Capital Medical University were collected.The 10 patients included 5 patients with internal carotid artery aneurysm,2 patients with subclavian artery aneurysm,2 patients with vertebral artery aneurysm,and 1 patient with internal carotid artery aneurysm combined with ipsilateral subclavian artery aneurysm.The surgical indications,surgical regimens,clinical efficacy,and complications were retrospectively analyzed. Results All the 10 patients underwent surgery successfully,with the surgery duration range of 60-420 min and the median surgery duration of 180.0 (121.5,307.5) min.Intraoperative bleeding volume varied within 30-400 mL,with a median of 90 (50,125) mL.The time of carotid artery blocking and vertebral artery blocking varied within the ranges of 10-20 min and 20-30 min,with the medians of 15.0 (11.5,16.3) min and 25.0 (15.0,22.5) min,respectively.No cardiac accident,cerebral infarction,or cerebral hemorrhage occurred during the perioperative period.The 10 patients were followed up for 3-58 months,with the median follow-up time of 8.5 (5.3,17.0) months.One patient with subclavian artery aneurysm developed artificial vessel occlusion 20 months after surgery.One patient with internal carotid artery aneurysm developed distal carotid artery stenosis 6 months after surgery.
