{"title":"Regulatory Effects of Cytokines on Spontaneous Pyroptosis in Neutrophils.","authors":"Tong Chen, Qian Ren, Feng-Xia Ma","doi":"10.3881/j.issn.1000-503X.16358","DOIUrl":"10.3881/j.issn.1000-503X.16358","url":null,"abstract":"<p><p>Objective To explore the regulatory effects of cytokines interleukin(IL)-1β,IL-6,tumor necrosis factor alpha(TNF-ɑ),gamma interferon(IFN-γ),granulocyte colony-stimulating factor(G-CSF),and granulocyte-macrophage colony-stimulating factor(GM-CSF)on spontaneous pyroptosis in neutrophils.Methods Neutrophils isolated from mouse bone marrow by density-gradient centrifugation were cultured <i>in vitro</i> for 20 h with or without 10,50 or 100 ng/mL IL-1β,IL-6,IFN-γ,G-CSF or GM-CSF,or for 12 h with or without 1,10 or 50 ng/mL TNF-α.After incubation,cells were stained with annexin Ⅴ(AV)/propidium iodide(PI),and the proportions and absolute number of neutrophils undergoing different forms of cell death were determined by fluorescence microscopy combined with manual counting.Pyroptotic neutrophils were identified by cell morphology in conjunction with AV/PI staining.Flow cytometry with counting beads was employed to measure the proportions and number of AV/PI-stained Ly6g<sup>+</sup>neutrophils in different forms of cell death.Western blotting was employed to assess the cleavage and activation levels of cysteinyl aspartate-specific proteinase-3(caspase-3)and gasdermin E(GSDME).Results Treatment with IL-1β or IL-6 had no significant effect on the proportion or number of neutrophils undergoing spontaneous pyroptosis.After 12 h of treatment with TNF-α at 1,10,and 50 ng/mL,the proportions of pyroptotic neutrophils were(14.79±0.45)%,(19.99±3.02)%,and(20.66±1.99)%,respectively,higher than that[(10.22±1.12)%]in the untreated control(<i>P</i>=0.024,<i>P</i><0.001,and <i>P</i><0.001,respectively).Treatment with 10,50,and 100 ng/mL IFN-γ for 20 h reduced the proportion of pyroptotic neutrophils from(17.43±1.88)%to 12.00%(all <i>P</i><0.001).G-CSF at 10,50,and 100 ng/mL reduced the proportion of pyroptotic cells to around 6.00%and greatly inhibited the cleavage of both caspase-3 and GSDME.After 20 h of treatment with 10,50,and 100 ng/mL GM-CSF,the proportions of pyroptotic neutrophils decreased to(7.52±0.53)%,(5.27±2.30)%,and(0.64±1.11)%,respectively.Conclusions Neither IL-1β nor IL-6 affects GSDME-mediated spontaneous pyroptosis in neutrophils.TNF-ɑ induces spontaneous pyroptosis in neutrophils,whereas IFN-γ,G-CSF,and GM-CSF demonstrate inhibitory effects.</p>","PeriodicalId":6919,"journal":{"name":"中国医学科学院学报","volume":"47 4","pages":"497-508"},"PeriodicalIF":0.0,"publicationDate":"2025-08-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"145063200","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

{"title":"Chinese Expert Consensus on the Clinical Application of Ultrasound Screening for Gastric Cancer(2025 Edition).","authors":"","doi":"10.3881/j.issn.1000-503X.16777","DOIUrl":"https://doi.org/10.3881/j.issn.1000-503X.16777","url":null,"abstract":"<p><p>The five-year survival rate of gastric cancer in China is close to that in European and American countries but far lower than that in the Republic of Korea and Japan,which have established national gastric cancer screening systems.It is of great significance to build a high-quality gastric cancer screening system adaptive to China's national conditions.Due to the large number of people at risk of gastric cancer and uneven distribution of medical resources,it is still difficult for China to carry out a nationwide gastroscopy screening plan for gastric cancer.Gastric ultrasound,with painlessness,no radiation,and easy acceptance and popularization,could be used as one of the alternative methods for initial screening of gastric cancer.Based on two gastric ultrasound-related consensuses published in 2020,this consensus elaborates on the necessity,feasibility,and existing problems of conducting preliminary gastric cancer ultrasound screening in China by analyzing the gastric cancer screening strategies and the difficulties faced by nationwide gastric cancer screening.Furthermore,this consensus introduces the indications and contraindications of gastric ultrasound examination,requirements for the operator and the contrast agent,ultrasound standard section,essentials of scanning operations,and stomach ultrasound report and data system (Su-RADS) and proposes the relevant consensus opinions accordingly.After multiple rounds of discussions and voting by experts from multiple societies,a total of 17 consensus opinions have been formed on gastric ultrasound as a preliminary screening technique for gastric cancer,with the aim of standardizing the popularization of gastric ultrasound.In addition,the consensus calls for conducting nationwide multicenter prospective studies to improve the level of evidence and provide data support for the construction of a preliminary gastric cancer ultrasound screening system that is in line with China's national conditions.</p>","PeriodicalId":6919,"journal":{"name":"中国医学科学院学报","volume":" ","pages":""},"PeriodicalIF":0.0,"publicationDate":"2025-08-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"144774463","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Methods Patients with clinically suspected endometrial cancer were prospectively enrolled and underwent MRI before the operation,which mainly included APTw and T2 mapping.Two radiologists independently performed regions of interest and measured the quantitative parameters.The intraclass correlation coefficients were used to assess the interobserver agreement,the independent samples t-test and Mann-Whitney U test were applied to compare quantitative variables between the MSI and microsatellite stable(MSS)groups,the receiver operator characteristic(ROC)curve was conducted to evaluate the predictive performances of different parameters,the Delong test was implemented to compare the area under the curve(AUC)between single parameters and their combination. Results Twenty-one patients in the MSI group and 37 patients in the MSS group were finally enrolled in this study.The results showed excellent observer agreements between two observers in assessing MRI parameters.APTw values[(2.72±0.52)% vs.(2.08±0.39)%,P<0.001]in the MSI group were significantly higher than those in the MSS group, T2 values [92.25(90.82, 99.91) ms vs.101.80(96.15, 112.13)ms, P<0.001]were significantly lower in the MSI group than those in the MSS group.The ROC curves showed that the AUCs of APTw and T2 values were 0.821 and 0.793,respectively,and no significant difference was observed between the two sequences(P>0.05).The combination of APTw and T2 mapping yielded the highest AUC of 0.897,which was significantly higher than APTw and T2 mapping alone(both P<0.05). {"title":"Association Between Gastroesophageal Reflux Disease and the Risk of Incident Chronic Obstructive Pulmonary Disease.","authors":"Ye Liao, Yun-Feng Zhou, Xiao-Rui Zhou, Xin Hu, Juan Liao, Lu Long","doi":"10.3881/j.issn.1000-503X.16279","DOIUrl":"https://doi.org/10.3881/j.issn.1000-503X.16279","url":null,"abstract":"<p><p>Objective To investigate the association between gastroesophageal reflux disease(GERD)and the risk of incident chronic obstructive pulmonary disease(COPD)and explore potential effect modifiers influencing this association.Methods Clinical data from 476 175 participants in the UK Biobank(2006-2010)were collected.A Cox proportional hazards model was used to assess the relationship between GERD and the risk of incident COPD.Subgroup analyses were conducted to examine potential modifiers of the primary findings.Results A total of 11 587(2.43%)new COPD cases were diagnosed.The Cox proportional hazards model revealed that GERD was associated with an increased risk of incident COPD(<i>HR</i>=1.59,95%<i>CI</i>=1.46-1.74,<i>P</i><0.001).GERD was linked to a higher risk of incident COPD in individuals aged<60 years(<i>P</i><0.001)and non-smokers(<i>P</i>=0.011).No association was observed between GERD and the risk of incident COPD in current smokers with a daily cigarette consumption<10 cigarettes(<i>P</i>=0.261).Conclusion GERD may increase the risk of incident COPD.</p>","PeriodicalId":6919,"journal":{"name":"中国医学科学院学报","volume":"47 3","pages":"402-407"},"PeriodicalIF":0.0,"publicationDate":"2025-06-30","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"144697258","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}
