{"title":"Expression and clinical significance of mTOR/p70S6K signaling pathway in breast carcinoma","authors":"Haili Lao, Xia Li, Haizhen Wang, Yanhai Li","doi":"10.3760/CMA.J.ISSN.1673-8799.2016.02.014","DOIUrl":"https://doi.org/10.3760/CMA.J.ISSN.1673-8799.2016.02.014","url":null,"abstract":"Objective \u0000To investigate the expression of TOR(mammalian target of rapamycinm)/p70S6K(70 kDa ribosomal protein S6 kinase) signaling pathway in human breast carcinoma. \u0000 \u0000 \u0000Methods \u0000From January 2013 to January 2016, 40 cases of breast carcinom tissues,40 cases of paracancerous tissues,32 cases of normal breast tissues were selected in Binzhou Central Hospital Affiliated to Binzhou Medical College.RT-PCR and Immunohistochemistry were employed to determine the expression of mTOR/p70S6K signaling pathway in cancerous and paracancerous tissuess of 40 cases and normal healthy tissues of 32 cases. The relationship between expression of the genes mRNA in breast carcinom tissues and related parameters was analyzed. \u0000 \u0000 \u0000Results \u0000The expression level of mTOR was markedly higher in the breast carcinoma tissues (0.611±0.098) than in the paracancerous tissues (0.419±0.101) or normal breast tissues (0.381±0.043) .The expression of p70S6K mRNA was also higher in the breast carcinoma tissues(0.625±0.109) than in the paracancerous tissues(0.471±0.129) or normal breast tissues(0.441±0.130), with a significant difference among groups (P<0.05). There was a positive correlation between mTOR mRNA and p70S6K mRNA expressions in breast carcinom tissues (γ=0.491, P=0.019). The expression of mTOR/p70S6K signaling pathway in the breast carcinom tissues was significantly correlated with the pathological grading, lymph node metastasis, but not with the diameter, and age. \u0000 \u0000 \u0000Conclusions \u0000The mTOR/P70S6K signaling pathway is specifically activated in breast carcinom tissues. All patients were scored by CS、CLIP、TNM staging and OKUDA. Evaluated this four staging systems on survival prediction value of 6 month advanced liver cancer. COX proportional hazard model was used to eliminate likelihood estimator in order to judge which staging criteria had better prognosis prediction. \u0000 \u0000 \u0000Results \u0000Follow-up period of 218 cases was 2-36 month, average follow-up time was (22.34±5.89) month, three-year survival rate was 33.94%, median survival time was 25.38 month. Of the four staging methods, degree of distinction was better TNM staging、OKUDA staging criteria. Randomly assign 24 patients to sorafenib group and 24 to sorafenib combine Chinese medicine group. To compare the change of their liver function and multilayer spiral MSCT image. \u0000 \u0000 \u0000Results \u0000There were no significant statistical difference between the two groups on size of the tumors, liver diffusion outside, cancer embolus of portal vein(P>0.05). There might be some factors that protect the cells from a proliferation inhibition by KAI1 in the solid tumors' microenvironment. Hypoxia and ischemia are the main characteristics of the microenvironment within solid tumors. Whether they affect the KAI1 inhibitory effects on cell proliferation is still unclear. \u0000 \u0000 \u0000Methods \u0000MiaPaCa-2 human pancreatic cancer cells do not express KAI1 protein. However, after being infected with Ad5-KAI1, they expressed KAI1 protein. We cultured them under hypoxic and serum-free conditions to simulate the solid tumor hypoxic-ischemic microenvironment. The cells were divided into the control, hypoxic, serum-free, and hypoxic with serum-free groups. The apoptosis were observed Annexin V-FITC/PI. The green fluorescent protein-labeled light chain 3 association with autophagosome membranes was detected by confocal microscopy. The ratio of LC3-Iito LC3-I expression level was detected by western blot. Pre-treatment of 3-MA was used to inhibit the autophagy.
