Mifepristone (4 mg kg−1 b.w.) was given daily to female rats for 8 days to develop PCOS, which was then detected by a protracted estrous cycle. The treatment regimen for the rats with PCOS consisted of 200 mg kg−1 b.w. of ELETC and nELETC administered orally every day for 28 days. Hormone tests showed that metformin acted as a favorable control. Nelson","doi":"10.1080/10496475.2022.2140739","DOIUrl":"https://doi.org/10.1080/10496475.2022.2140739","url":null,"abstract":"ABSTRACT This study was undertaken in Bidayuh communities in the Padawan, Bau, and Siburan districts in Malaysia to (1) compare the medicinal plant utilization, (2) compare the number of medicinal plants collected annually and total number of medicinal plant species collected, and (3) assess the influence of key socio-demographic factors on the pattern of annual collection and preferred species. Surveys were conducted on 226 households, based on stratified random sampling. Key informant in-depth investigation and group meetings were used to gather further information. A total of 44 medicinal plant species that were used to treat 25 ailments were recorded from all districts. Five primary ailments for which the plants were used were fever (11 plant species), wound (10 plant species), post-partum care (eight plant species), hypertension (seven plant species), and diseases of the skin (seven plant species). There was a difference in the number of medicinal plants collected annually (X 2(2) = 8.55, p = .01) with the highest mean difference between Kuching (Mean rank = 125.65) and Siburan (Mean rank = 97.78) districts. Antitumor effect was analyzed on mouse solid and ascites tumor models. The phytochemical profile of the extract was analyzed by liquid chromatography-mass spectrometry (LC-MS). Apoptosis and cell cycle were analyzed by fluorescence resonance energy transfer (FRET) and flow cytometry. PQ exhibited cytotoxicity on Dalton’s lymphoma ascites (DLA) and Ehrlich’s ascites carcinoma cells with IC50 of 9.4 and 9.2 μg mL−1 and anti-proliferative effect on breast (MCF7) and colorectal (HCT116) cancer cell lines with IC50 of 166.5 and 107.09 μg mL−1, respectively. The extract did not show toxicity in normal rat intestinal cells IEC6 and murine macrophage cells RAW264.7. In ascites tumor model, the mean survival days of the animals increased to 25 ± 4.30 d in the treated group compared to untreated (21.2 ± 2.68 d). In DLA-induced solid model, the tumor weight decreased to 1.6 ± 0.9 g from 4.5 ± 0.58 g by treatment with 400 mg kg−1 b.w. extract. LC-MS analysis revealed the presence of genistein, kaempferol, isovitexin, and quercetin that are known for anticancer potential. The acute toxicity in mice revealed that PQ is nontoxic. In a short-term in vitro study, ethyl acetate extract (VCEA) was found to be cytotoxic to DLA cells with an LC50 of 61.24 μg mL−1. GC-MS analysis of VCEA revealed presence of several volatile molecules including nerolidol. Molecular docking study using nerolidol with Ferroportin-1 predicted strong binding affinity, indicating ferroptosis induction which was later confirmed by in vitro assays. VCEA also demonstrated strong reduction in the proliferation of YAC-1, another lymphoma cell line with G2/M arrest leading to apoptosis. Escherichia coli and Listeria monocytogenes were sensitive to the extracts, while Bacillus subtilis and Klebsiella pneumoniae were resistant. Anti-inflammatory potential of methanolic extract was assessed in the LPS-induced RAW 264.7 macrophage cell lines. The GC-MS of methanolic extract showed the predominance of phenolic acids, fatty acids and phytosterols, while terpenoids and flavonoids were identified by LC-MS analysis. Species with the highest use values (UV) were selected for phytochemical screening. In total, 79 respondents were interviewed and 36 species distributed in 23 plant families were identified. Based on the UV, the most important species were Olax subscorpiodea (UV=0.13), Crinum glaucum (UV=0.12), Allium ascalonicum (UV=0.09), Allium sativum (UV=0.06), Zingiber officinale (UV=0.05), and Securidaca longipendiculata (UV=0.05). We found larger zones of inhibition of growth of C. albicans with the essential oil (EO) and methanolic extract (ME) than amphotericin B. Similarly, the MIC values for the EO and ME were lower than that of amphotericin B. The EO and ME caused greater reductions in biofilm formation than amphotericin-B. This study was designed to investigate the polyphenolic compound, antioxidant, and anti-inflammatory activities of the ethanolic extract of Viscum monoicum (EVM). The extract was subjected to HPLC-DAD analysis for the quantification of polyphenols. In addition, DPPH, ABTS free radical scavenging, reducing power, total antioxidant capacity, phenolic, and flavonoid content was measured for antioxidant activities of EVM. Besides, the anti-inflammatory activity of EVM was studied using carrageenan and histamine-induced rat paw edema at different doses. High levels of ellagic acid and (+)-catechin hydrate (410.15 and 340.88 mg/100 g extract, respectively) were determined for the first time through HPLC in EVM. IC50 of EVM was significant in DPPH and ABTS scavenging assay compared to that of ascorbic acid. The total antioxidant capacity, total phenolic, and total flavonoid contents were estimated and found significant amount equivalent to standard. At a dose of 400 mg kg−1, a significant anti-inflammatory activity (P<0.01) was observed in rats at both carrageenan and histamine-induced edema test models with a reduction in the paw volume; in comparison to indomethacin. The qualitative and quantitative profiles of the extracts were established by UHPLC-ESI/MSMS, and their capacity to quench DPPH∙ and ABTS∙+ radicals was determined. There were positive correlations between the phenolic content and antioxidant capacity for all species analyzed, highlighting to P. vitifolia and P. pittieri which exhibited the highest values for all the determined parameters.
