Intact control and Ovx rats were i.p. treated with sterile olive oil for 4 weeks. Peroxidase-antiperoxidase (PAP) immunohistochemical procedure was applied to localize calcitonin (CT) in thyroid C cells while its serum content was determined by RIA method. Morphometric analyses of the C cells volume, that of their nuclei and relative volume density included stereological method with the multipurpose test system M42. Also, the average number of C cells number per mm2 was calculated. Ovx led to a significant increase in body weight (21%; p<0.005). At the same time the C cells of Ovx rats had a significantly decreased cell volume (13%; p<0.005) and their number per mm2 was increased by 59% (p<0.001) in comparison with the controls. Ovx resulted in reduction of serum CT level by 45% comparing to the corresponding controls. Hyperglycemia induces a large number of alterations in vascular tissue that potentially accelerate the atherosclerotic process. Nonenzymatic glycosylation of proteins and lipids which can interfere with their normal function by disrupting molecular conformation, alter enzymatic activity, reduce degradative capacity and interfere with receptor recognition, oxidative stress and protein kinase C activations are the major processes that encompass most of the pathological alterations in the diabetic vasculature. Diabetes-associated oxidative stress is a consequence of both increased production of free radicals and reduced capacity for antioxidative defense. The aim of this study was to test the parameters of antioxidative defense: superoxide dismutase (SOD), glutathione peroxidase (GSH-Px), glutathione reductase (GR) and total antioxidant status (TAS), in type 2 diabetic patients with and without cardiovascular complications in order to determine the effect of hyperglycemia on the extent of disorders of antioxidative parameters. Sixty nine type 2 diabetic patients with cardiovascular complications, along with 48 diabetics without complications and 42 healthy subjects were examined. Determination of antioxidant parameters was performed using commercial tests manufactured by Randox Laboratories, UK, based on spectrophotometer determination methods. Structurally they are peptides (often glycosylated) used by cells for intercellular communication and control the inner environment of the cells in which they operate. Cytokines are produced by the cells involved in the immune response, inflammation, hemopoiesis, healing and systemic response to injury. Immunity, inflammatory reactions and haemostasis involve close interactions between immunocompetent cells and vascular endothelium. Vascular cells are both a target for cytokines and their source. The spectrum of endothelial cell responses challenged by cytokines is wide and varied, with different cytokines activating distinct, but overlapping, sets of functions. Under normal resting conditions endothelial cells constitutively express certain protective genes with the purpose to maintain the endothelial cells in their quiescent phenotype by inhibiting NF-kB activation and exerting antiapoptotic functions. In this status endothelial cells can exhibit their barrier and anticoagulant functions even in the presence of low levels of stimulants. When the endothelial cells are exposed to numerous stimuli such as TNF, IL-1, endotoxin or xenoreactive antibodies and complement, which are usually associated with infections, graft rejection or autoimmune diseases such as, vasculitis, NF-kB induces the expression of adhesion molecules such as E-selectin, chemokines such as IL-8 and procoagulant molecules such as TF. Besides the induction of expression of a functional programme related to thrombosis and inflammation, IL-1 and TNF also induce production of autocoids including nitric oxide (NO). Both the inducible form of NO synthase (iNOS) type II and the constitutive (type III) isoform of NOS are present in endothelial cells catalyzing the conversion of arginine into citruline and NO. The formation of NO is an ubiquitous biochemical pathway involved in the regulation of neurotransmission, vasodilatation, immunity and cytotoxicity. During inflammatory reaction NO produced by endothelial cells exerts its autocrine function through the inhibition of cytokine-induced expression of adhesion molecules and cytokine production by endothelial cells. Also, it has a protective role in inflammation through the inactivation of NADPH oxidase and the consequent impairment of superoxide production for cell mediated injury. On the other hand, there is considerable evidence that NO contributes to tissue destruction in inflammatory and immune diseases being a key component of the cytostatic/cytotoxic function of the immune system. Milosevic","doi":"10.2298/JMH0404393K","DOIUrl":"https://doi.org/10.2298/JMH0404393K","url":null,"abstract":"Serum levels of somatostatin were determined by RIA method in 50 patients with liver cirrhosis, 15 with pernicious anemia and atrophic gastritis, 31 with inflamatory bowel disease, 32 with colorectal tumors and in 40 control persons. In patients with liver cirrhosis somatostatin levels were significantly higher than in control group (p<0.01). Patients with pernicious anemia and ulcerative colitis in acute phase of the disease had significantly lower levels of somatostatin (p<0.01) (p<0.005). In patients with colorectal tumors somatostatin were significantly lower then in control group (p<0.01). Our results show that the liver is involved in somatostatin metabolism. Atrophic gastritis, ulcerative colitis and M. Patients were divided into three groups: patients presented with alcohol induced acute pancreatitis (AAP) (n=30); patients suffering from biliary acute pancreatitis (ABP) (n=58); and patients having an idiopathic form of acute pancreatitis (n=28). Till the 24th hour after admission to the clinic, we evaluated serum amylase, aspartate aminotransferase (AST), alanin aminotransferase (ALT), alkaline phosphatase (ALP), gamma-glutamyl transferase (GGT), total bilirubin, and the cytometric indicator - mean corpuscular volume (MCV). These laboratory findings were analyzed with the help of routine clinical laboratory methods, and the results were elaborated with the software package SPSS. In patients suffering from ABP, we evaluated higher median levels of serum amylase (P=0.047), AST (P=0.019), and ALT (P=0.001), in comparison to the group suffering from AAP who presented with higher levels of GGT (P=0.128) and particularly MCV (P=0.015). As a whole, serum amylase, AST and particularly ALT continue to play a major role in diagnostic and differential-diagnostic assessment of AP. The role of the hematological parameter MCV is similar. Stanković","doi":"10.2298/JMB0602153C","DOIUrl":"https://doi.org/10.2298/JMB0602153C","url":null,"abstract":"Summary: Since nowadays morbidity and mortality of bronchial asthma are increasing more and more, this issue presents a global challenge despite the significant development of knowledge about the pathogenetic mechanism of asthma, role of inflammatory cells, regulation of inflammatory and immune responses, about many mediators and their co-regulation. Cytokines and their modulators are becoming increasingly important because they are crucial intercellular transmitters which contribute to the development of airways remodeling. The aim of this study is to evaluate Th1/Th2 ratio and to estimate the inter-dependence between cytokines and pro/antioxidant parameters in patients with newly diagnosed bronchial asthma, which could be the basis for some new possibilities in the prevention and therapeutic application of this chronic disease. The parameters were determined in the patients’ plasma, erythrocytes and cultures of lymphocytes treated with Concanavaline A (Con A) and Phorbol 12-myristate 13-acetate (PMA). Contrary to Th1 cytokines, Th2 profile of cytokines shows a significant increase (p<0.01) vs. control group, with higher level in extrinsic (atopic asthma) vs. intrinsic asthma. The results of oxidant stress markers (increased levels of lipid peroxides, xanthine oxidase activity in blood of patients with BA) and antioxidant parameters (decreased superoxide dismutase (SOD) activity, increased glutathion peroxidase (GPx) and catalase activities, and glutathione (GSH) concentration) suggest that asthma is associated with disturbances in pro/antioxidants indicating that oxidant stress occurres in patients with newly diagnosed BA. Investigation of cytokines in lymphocyte cultures of patients with BA showed a significant increase in basal TNF-a concentration (p<0.05), IL-13 and IL-4 (p<0.05) after stimulation with Con A, and TNF-a after stimulation with PMA (p<0.01). The aim od this study was to investigate the effect of angiotensin II receptor blockers-valsartan, on plasma lipid profiles in diabetic patients with persistent microalbuminuria (McA). Among the group of 67 type 1 diabetic patients, we choose 20 normotensive patients with incipient DN, who were divided into two groups (10 patients each): valsartan group, treated with 80 mg valsartan daily during 6 months, and the group treated with placebo during the same period. After 6 months therapy, the reduced values of total cholesterol (5.35 “ 1.19 vs 5.20 “ 1.94 mmol/L), triglyceride (1.42 “ 0.79 vs 1.21 “ 0.69 mmol/L) and LDL-cholesterol fraction (3.41 “ 1.21 vs 3.31 “ 0.84 mmol/L) were registrated in valsartan group. While compared with placebo gruop, reduction in serum triglyceride values reached statistical singificancy (-15.5%; p<0.05). Patients with persistent McA have a higher risk of cardiovascular morbidity and mortality than diabetics without this complication. In physiological conditions only plasma proteins of low molecule weight are completely filtered through basal membrane. Due to damages of basal glomerular membrane there is increase in filtration of plasma protein of middle and high molecular weight. Depending on etiology of proteinuria it can be prerenal, renal and postrenal. By analyzing albumin a1-microglobulin, immunoglobulin G and a2-macroglobulin, together with total protein in urine, it is possible to detect and differentiate causes of prerenal, glomerular, tubular and postrenal proteinuria. Majkić-Singh","doi":"10.2298/JMH0401037S","DOIUrl":"https://doi.org/10.2298/JMH0401037S","url":null,"abstract":"Summary: Hemostatic system, as an integral part of biological homeostatic mechanism in humans, is subject to changes due to effect of hormonal therapy used in preparation for in vitro fertilization. The objective of the study was to analyze the impact of hormonal changes on homeostatic parameters during four cycles of preparation, according to long-term protocol for in vitro fertilization. The following parameters were determined in the study: prothrombin time, activated partial thromboplastin time, fibrinogen, antithrombin III, plasminogen, a2-antiplasmin and plasminogen activator inhibitor. In the period of ovarian hyperstimulation and in postovulatory period, significantly lower PT, APTT, AT III, PAI-1 and a2-APL values were obtained (p 0.404, for p<0.05).
