Protease production in Candida strains isolated from patients with otomycosis is, according to our knowledge, not investigated. The present study was aimed at determining in vitro protease activity in 28 strains of Candida spp. (C. parapsilosis, C. famata, C. guilliermondii, C. albicans and C. kefyr) isolated from patients with otomycosis. The majority of isolated strains 25/28 (89.28%) were protease positive. The protease Pz ranged from 0.691 to 0.851. The animals received ICV three 1.0 mg doses of octreotide dissolved in 10 mL saline every second day. The controls were treated with equivalent volume of physiological saline by the same schedule. FSH- and LH-producing cells were examined using peroxidase-antiperoxidase immunocytochemical procedure. Morphometric and stereologic examinations were performed to evaluate changes in the number, volume and volume densities of gonadotrophic cells. In females treated with octreotide, the gonadotrophic cells were smaller and often pycnotic, while the number of FSH- and LH-immunopositive cells per unit area (mm2) was significantly reduced. Octreotide also induced a significant reduction of the FSH- and LH-immunoreactive cells volume, as well as of their volume densities. Five days after the lesions the animals were checked for specific rotational behavior induced by middle dose of amphetamine and the results confirmed the effectiveness of the lesions. The RNAs from the striatum were isolated at different time points after the lesion, and the RT-PCR analyse were performed for the D1 and D2 receptor mRNA. The results show a decline in the D2 receptor mRNA level (40%) at 6 h and 24 h points while this change was not observed seven days after the lesion. Endothelial cells express a variety of adhesive receptors that regulate their adhesion to leukocytes and also to the extracellular matrix. These interactions are complex phenomena that require multiple recognition mechanisms, and include the first rolling and later the stationary adhesion and transmigration of leukocytes. It is known that cytokines have regulatory effects on cell adhesion molecules expression. In the present study we investigated the influence of cytokines (IL-1b, IL-2, IL-4, IL-6, IL-8 IL-10, TNF-a, IFN-g) and the combined use of IL-2, IL-4, IL-6, IL-8 and IL-10 with IL-1b, TNF-b or IFN-g on the expression of adhesion molecules of cultured human umbilical vein endothelial cells (HUVECs) after stimulation for 16 hours. Likewise, in vitro model described herein is designed to mimic the activation of endothelial cells by cytokines as seen during inflammatory processes. This process is mediated by specific cell adhesion molecules being crucial for the generation of immune and inflammatory responses. Therefore, HUVECs are treated with two different cytokine combinations consisting of either IL-2, IL-6, IL-8 IFN-g and TNF-a or IL-1b, IL-2, IL-4, IL-6, IL-10, IFN-g and TNF-a. Endothelial cells were collected from hunam umbilical vain using collagense type II, and cell cultures in complete medium were kept in the incubator (37.4°C, 5% CO2). After stimulation cells were prepared for analysis using tripsinisation procedure. The surface expression of the following adhesion molecules was determined in cultured human umbilical vein endothelial cells (HUVECs) by means of flow-cytometric analysis: CD 62P (P-selectin), CD 62E (E-selectin, ELAM-1) CD 106CD 34 (L-selectin ligand). The highest CD 62E expression on the surface of HUVECs was found when endothelial cells were stimulated with TNF-a alone. Also they were increased after stimulation with IL-1b, while IL-4 led to down-regulation of CD 62E. Incubation of HUVEC monolayers with IL-1b, IL-4 as well as TNF-a and IFN-g, statistically significant, reduced the surface expression of CD 34 while other cytokines did not affect CD 34 expression. Incubation of HUVECs with a single cytokine caused no statistically significant changes in CD 62P expression compared to controls. The most potent effect on CD 54 expression was found under TNF-a stimulation; IL-1b and IFN-g had also amplifying effects, while all other tested cytokines caused no significant changes in surface molecule expression. Surface expression of CD 106 was amplified during incubation with IL-1b, IL-4, TNF-a and IFN-g. Single stimulation of tested cytokines did not significantly alter the cell surface expression of CD 31. Based on the improvements in understanding the biology of these cancers, treatment and ongoing development of new diagnostic tools aiming towards »personalized medicine« are under clinical evaluation. This article describes the improved value of one of the immunodetectable forms of the prostate-specific antigen (PSA), the so-called »complexed PSA« (cPSA), in detecting prostate cancer, and also presents an overview of publications on the clinical value of a new immunoassay detecting the extracellular part of the HER-2 molecule in serum of breast cancer patients. In lectin blot, TBG reacted positively with the following lectins: Sambucus nigra agglutinin (SNA I), Ricinus communis agglutinin (RCA I), wheat germ lectin (WGA), phytoheamagglutinin (PHA) and pea lectin (PSA). The obtained results indicate that purified TBG contains N-linked oligosaccharide chains consisting of mannose, galactose, N-acetylglucosamine and sialic acid. Isoelectric focusing of TBG at pI 4.2’ 4.6 revealed three bands, which confirmed that isolated TBG had retained its structure without desialylation. Our study is aimed at determining correlation between IL-4 and IL-5 serum concentrations and severity of the disease in patients with bronchial asthma. The total of 65 patients with asthma is classified into three groups: 1. patients with mild, 2. moderate and 3. severe asthma. The control group comprised 10 healthy individuals. Measurement of IL-4 and IL-5 serum concentrations was performed according to ELISA method. Minimal detectable concentrations of IL-4 and IL-5 were 0.27 pg/mL and 4 pg/mL, respectively. IL-4 (Z = 2.245, p = 0.025) and IL-5 (Z = 2.139, p = 0.032) serum concentrations are significantly lower in healthy individuals in comparison to the individuals with asthma. Comparison of the study results between the groups (mild, moderate and severe asthma) evidenced that IL-4 serum concentrations in the group with moderate (Z = 2.604, p = 0.009) and severe asthma (Z= 2.658, p = 0.008) were statistically significantly higher in comparison to the group with mild asthma. IL-5 serum concentrations in the group with moderate (Z = 3.910, p = 0.000) and severe asthma (Z = 4.037, p = 0.000) were statistically significantly higher in comparison to the group with mild asthma. Osteocalcin (OC) is a non-collagen peptide involved in the bone mineralization process. It is used in early diagnosis of the primary osteoporosis. The aim of our work was to determine if there was a difference in both the catalytic activity of the TRAP and the serum concentration of OC in patients affected by osteoporosis and osteopenia. The bone mineral density T-score (BMD) of the first group of patients (n=60) was not higher than -2.3, defined by the WHO as osteoporosis. The T-score of the second group (n=60) was found to be between -0.4 and -1.7, defined by the WHO as osteopenia. The catalytic activity of TRAP was determined by the kinetic method proposed by the BIOMERIEUX Company (Germany), using a Mira Cobas plus analyzer (Germany). The BMD was determined by an ultrasound technique. The ROCHE ECL technique and the Elecsys 2010 analyzer (Germany) were used to measure the OC concentration. The results show a significant increase in the catalytic activity of TRAP in the osteoporosis patients sera when compared with the group with osteopenia (p<0.001). Statistically significant difference was found between the groups in the case of OC (p<0.01). Biochemical markers play a pivotal role in the diagnosis and management of patients with acute coronary syndrome (ACS), as witnessed by the incorporation of cardiac troponins into new international guidelines for patients with ACS and in the redefinition of myocardial infarction. Despite the success of cardiac troponins, there is still a need for development of early markers that can reliably rule out ACS from the emergency room at presentation and detect myocardial ischemia also in the absence of irreversible myocyte injury. Under investigation are two classes of indicators: markers of early injury/ischemia and markers of coronary plaque instability and disruption. From that aspect, an adequate clinical evaluation in stone forming patients is of paramount importance. An effective and rational attitude in detecting possible causes of stone formation demands the adequate analytic program in biochemical investigations. Presented program is adjusted to the guidelines on urolithiasis, recommended by Board EAU (European Association of Urologists) Healthcare Office, for year 2003. Regarding to known chemical stone composition, category of stone former and presence of an specific risk factor, analytic program could be minimal (fasting morning spot urine sample analysis, urineculture and blood analysis (calcium, albumine, creatinine and urate and stone analysis) and extended (quantification of all parameters in serum and in two consequtive 24h urine collections, relevant for stone formation and evaluation of renal function).
