After the slaughter of the cows, the ovaries in physiological state of follicular growth were selected and the size of the follicles was evaluated. Cells of the granulosa layer were aspirated from small (< 4 mm), medium (4–7 mm) and large (> 7 mm) follicles and determined: intensity of oxygen absorption – polarographically, redox capacity – potentiometrically. K3[Fe(CN)6] was used as an extracellular electron acceptor. The share of oxygen consumed by aerobic glycolysis was determined by using sodium fluoride inhibitor, the NAD-dependent link of the mitochondrial electron transport chain - amytal, and the terminal (cytochrome oxidase) – sodium azide. The share of free-radical oxidation of unsaturated fatty acids in the realization of oxygen was detected by Na2EDTA. \u0000Granulosa cells from the ovary of follicular growth are characterized by respiratory activity and regenerative capacity, the intensity of which depends on the size of the follicles from which they are extracted. The highest values of the investigated indicators are characteristic of cells extracted from large follicles, lower – from small follicles, and the lowest values are from medium follicles. The high intensity of oxidative processes in granulosa from small follicles ensures the growth of oocytes, and from large follicles – maturation of the germ cell in the dominant follicle. At the same time, the increased intensity of oxygen consumption by the cells of small and dominant follicles is manifested against the background of limitations in electron acceptors in the extracellular environment. \u0000The use of inhibitors of ATP resynthesis of both glycolysis and individual links of the mitochondrial respiratory chain testify the activity of the indicated metabolic pathways in granulosa and the possibility of using alternative substrates to ensure the existence of cells. The main way of utilization of oxygen in the granulosa layer cells of small and large follicles is aerobic glycolysis, and from the medium there are alternative ways of supplying and using substrates. Applikus-vet is a new domestic combined veterinary agent for external use, which has adsorption and bactericidal action. The preparation is designed on the basis of hydrogel of methylcremic acid with immobilized Cuprum ions and is intended for the treatment of wounds and ulcerative lesions of the skin. Hydrogel of methylcremic acid has an adsorption capacity, Cuprum ions have an antimicrobial effect and cause the denaturation of microorganism’s proteins. \u0000The article outlines the clinical study of a new veterinary drug Applikus-vet, paste for external use, production of PJSC \"EOF\" Creom-Farm \"(Ukraine), in the treatment of cats with clinical signs of aseptic skin necrosis (brittle skin syndrome). \u0000The drug was used in the complex treatment of two cats of long-haired breeds – female, aged 7 years, and male, 14 years of age, with clinical signs of aseptic skin necrosis in the area of the thoracic, the lumbar departments of the back and abdominal wall. The female had a much larger area of the affected skin than the male. Diagnosis was put on the basis of anamnesis, a clinical picture of the disease. The veterinarian of the clinic prescribed complex therapy, which included the drug Applikus-vet for application to the wounds of animals. The general course of treatment with the test drug lasted 4–8 weeks. \u0000The drug Applikus-vet showed good therapeutic efficiency in the complex treatment of cats with clinical signs of aseptic skin necrosis. Depending on the complexity of the disease, epithelialization and reduction of the inflammatory process in the affected area of animal’s skin were observed on 21–35 days of treatment. A significant decrease in the area of the affected skin and the formation of scars and scab was noted on 28–50 days of treatment. Pazderska","doi":"10.36359/scivp.2023-24-1.30","DOIUrl":"https://doi.org/10.36359/scivp.2023-24-1.30","url":null,"abstract":"The National Reference Laboratory of Drug Residues Control has implemented a system of \"classical\" honey safety control based on the parameters of antimicrobial drug residues: samples testing by the screening enzyme-linked immunosorbent assay (ELISA) method and, in the case of positive or possibly positive samples occurring, – retesting by confirmatory UPLC-MS/MS multi-method developed in the laboratory. The article presents the results of ELISA screening method validation for the determination of residual amounts of tetracycline group antibiotics (tetracycline, oxytetracycline, chlortetracycline, doxycycline) in honey samples using Tetracyclin® RIDASCREEN test kits by R-Biopharm (Germany) and Oxytetracycline ELISA by EuroProxima (the Netherlands) and its verification by means of the confirmatory method. \u0000In accordance with the Commission Implementing Regulation (EU) 2021/808, the validation of test kits proposed by the manufacturers was carried out taking into account the minimum required limit of detection. The fitness-for-purpose of the methods was confirmed based on the establishment of main validation parameters for screening methods (technical threshold and cut-off factor) using control (blank) and fortified honey samples with standard solutions of tetracycline at the level of ½ of the minimum required limit of detection, according to the \"added-received\" criterion. The proposed methods are the simple in carrying out, time- and cost-effective, which are their main advantages. \u0000The reliability of the results obtained by the proposed methods was verified using the developed confirmatory UPLC-MS/MS multi-method. Martynyk","doi":"10.36359/scivp.2023-24-1.18","DOIUrl":"https://doi.org/10.36359/scivp.2023-24-1.18","url":null,"abstract":"Drugs that have antimicrobial, anti-inflammatory and wound-healing effects deserve special attention, sodium-containing drugs are one of them, which is why they are used in veterinary medicine. \u0000The article presents the results of experimental studies to study the sub-acute toxicity of sodium-containing drugs: known – glycetinate 2%, its analogues – ovocid 2%, ovocid 3%, potassium-sodium 1.5% (Na / K, 1.5%). \u0000The study of sub-acute toxicity of drugs was carried out by applying drugs to the skin of animals. The experiment used white rats weighing 250-270 g. 4 groups of rats with 3 animals in each were formed: rats of group I on the skin was applied ovocid 3%, group 2 - Na / K drug 1.5%, 3rd group – ovocide 2%, and the 4th group of rats (control) was applied glycetinate 2%. The animals were subjected to clinical observations, assessing their general condition and skin reaction. The effect of drugs on morphological and biochemical parameters of blood of rats was studied. \u0000Evaluating the general condition, behavior, local skin reaction of rats for the study of subacute toxicity, local irritant action of sodium-containing drugs, it was noted that they did not cause visible violations of physiological functions, the appearance of clinical signs of intoxication. No significant clinical signs of inflammation or irritation (erythema, edema, and cracks) were recorded on the skin of the animals. Various pathologies of the mammary gland do not lose their relevance. They are among the leading causes leading to economic losses in farms due to lack of milk, reduction of its grade and culling of cows in the first lactation, or even after the first calving. The work was carried out throughout 2021 in the conditions of farms with intensive technologies for the production of marketable milk. The frequency of occurrence of mastitis was studied in the course of the research, the main etiological factors that influenced the frequency of their occurrence were determined. As a result of a complex of studies of cows at different stages of lactation, it was found that a combination of factors influenced the manifestation of mastitis. The problems were similar and the dynamics of detection of mastitis in cows also did not differ in both farms. \u0000The main cause that provoked the occurrence of mastitis was found to be the influence of the association of microorganisms on the mammary gland. After carrying out a complex of bacteriological studies of milk samples obtained from cows in experimental farms, we found that cultures of microorganisms were identified in milk: S. aureus, S. agalactiae, E. сoli, S. lactis, S. epidermidis. The species spectrum of microorganisms identified from milk depended on the form of mastitis. In clinical forms of mastitis, cultures of S. aureus (58.4%), S. agalactiae (28.3%) and E. сoli (13.2%) were more often identified. In subclinical forms of mastitis, the spectrum of isolated microflora was somewhat different, in particular, cultures of S. aureus were isolated in 40.2%, S. agalactiae - 24.5 %, S. lactis - 19.3 %, S. epidermidis - 10.2 %, E. сoli - 5.8%. A predisposing factor to the occurrence of mastitis in cows was the use of straw as bedding, which is a very good organic substrate for the development of microorganisms. Since we found the remains of damp bedding in the stalls on both farms, an additional factor of influence through bedding is also relevant. Mamchur","doi":"10.36359/scivp.2023-24-1.26","DOIUrl":"https://doi.org/10.36359/scivp.2023-24-1.26","url":null,"abstract":"The recipe of mineral-vitamin premix, made on the basis of salts of mineral elements (Cuprum, Zinc, Cobalt, Iodine and Selenium) and fat-soluble vitamins (A and D), which are deficient in the Pre-Carpathian zone, has been developed and its influence on individual metabolic links and productivity of dairy cows has been investigated. The experiment was conducted on two groups of highly productive cows of Simental breed, analogues in origin and productivity in the summer-pasture period of keeping. The main diet for animals of both groups consisted of pasture grass, mowed mass of cereal-bean mixtures of green conveyor, hay cereal-mixed grass, molasses and standard feed K 60-32-89 with premix P 60-5M. The experimental group received a similar amount of this feed, but with an experimental mineral-vitamin premix. It was found that in the herbal-concentrate type of diet in the control group, the deficiency of Cuprum, Zinc, Cobalt, Iodine and Selenium was 16.6, respectively; 13,2; 6,9; 26,2; 51.8%, and the deficiency of vitamins A and D - 60.0 and 16.3%. Introduction of mineral-vitamin premix into the compound feed, which contained salts of the specified elements deficient for the Pre-Carpathians, made it possible to better balance the rations according to the mineral part, which significantly influenced both the intensity of metabolic processes in the rumen and blood, and the milk productivity of cows and the quality of milk. In particular, the positive effect of its feeding on the level of fermentation, the activity of enzyme systems and the intensity of synthetic processes in the rumen of cows of the experimental group was noted. The study of the hematological picture showed a tendency to increase the level of red blood cells and hemoglobin in the blood of cows of the experimental group, an increase in the concentration of nitrogen fractions and urea (P < 0.01). Analyzing the results of research, it can be argued that ensuring the optimal level of mineral-vitamin nutrition led to an increase in the intensity of metabolic processes in the body of animals and therefore contributed to an increase in dairy productivity. \u0000The average daily yield of natural milk in the experimental group for 90 days of the accounting period was 21.6 kg and was 10.8% higher than in the control (19.5 kg). The chemical composition of milk of cows that received a mineral-vitamin premix as part of the feed was preferable. Yanovych","doi":"10.36359/scivp.2023-24-1.12","DOIUrl":"https://doi.org/10.36359/scivp.2023-24-1.12","url":null,"abstract":"This manuscript presents a developed method for determining cefquinome sulfate in piglets blood plasma intended for clinical and pharmaceutical research of veterinary drugs based on it. Blood plasma proteins were precipitated twice with a solution of trichloroacetic acid. The supernatant was further purified by a series of solid-phase extraction. Separation was performed on an inverted phase Kinetex EVO С18 column using acetonitrile and 0,1 % trifluoroacetic acide solusion as the mobile phase. The gradient mode of eluents was used during 10 min at a flow rate of 1,4 ml/min. The peak retention time of cefquinome sulfate is 4,2 min. The specificity of the analytical method was checked by comparing the chromatographic separation of a sample of blood plasma enriched with a standard solution of cefquinome sulfate and a sample of blood plasma placebo. The preparing loaded blood plasma samples procedure for building a calibration graph is described in the article. The validation parameters of the method “recovery” and “coefficient of variation” were considered in accordance with the criteria of Council Directive 2002/657/EC. The procedure of sample preparation of fortified blood plasma to construct calibration graph is described in the manuscript. The mean recovery from fortified blood plasma samples in the range of 0.1-2.0 μg/ml cefquinom sulfate was 102.3 %. The method is linear in the concentration range of 0.1 – 4.0 μg/ml of cefquinome sulfate. The correlation coefficient for the determination method is 0.9998. The results obtained in the study of the linearity of this technique were used to estimate the correctness and convergence. The accuracy of the measurements was evaluated by examining the known amounts of analyte added to the control blood plasma. Recovery data are acceptable because they are within ± 10% of the target value. The method has sufficient convergence (accuracy). The evaluation of the intermediate precision of cefquinome sulfate determination was assessed on three different days of analysis. The limit of detection for cefquinom sulfate is 0.05 μg/ml and limit of quantification - 0.10 μg/ml. The average CV for each compound was <10%. The procedure was confirmed and then applied to determination cefquinome sulfate in the pig blood plasma obtained during the study of the pharmacokinetics of the veterinary drug. These injuries pose challenging problems in animals because traditional treatments are lengthy and not always successful. In animals, skeletal muscles are plastic and dynamic tissues that can adapt to different conditions, such as fiber type changes and hypertrophy, and recover from injuries. \u0000The main purpose of this article is to discuss, analyze and summarize the results of research aimed at the use of allogeneic mesenchymal stem cells, a means of regenerative therapy, in veterinary practice. \u0000The main direction of cell therapy is the use of allogeneic mesenchymal stem cells (MSCs). These cells are undifferentiated and can renew themselves, which allows them to develop into cells with specific functions and become an effective treatment for tissue repair. Bacterial diseases of the pig digestive tract lead to significant economic losses due to high mortality, reduced growth, treatment and prevention costs. \u0000The main bacterial intestinal infections of pigs are anaerobic enterotoxemia (clostridiosis), colibacillosis, intestinal salmonellosis, dysentery, proliferative enteropathy (ileitis). \u0000Anaerobic enterotoxemia of pigs is an acute toxic-infectious disease mainly of newborn piglets, caused by pathogenic bacteria of the genus Clostridium and characterized by hemorrhagic-necrotic inflammation of the intestinal mucosa, diarrhea and toxicosis. \u0000Swine colibacillosis is an intestinal infection caused by the enterotoxigenic Escherichia coli, which is able to produce enterotoxins that locally affect the intestines of pigs, causing diarrheal syndrome. \u0000Intestinal salmonellosis is a factorial infection. The causative agents are enteropathogenic salmonella (mainly Salmonella enterica serotype typhimurium), which cause inflammation and necrosis of the small and large intestine, leading to diarrhea, which may be accompanied by generalized sepsis. \u0000Dysentery is a severe enteroinfection of pigs caused by the anaerobic bacterium Brachyspira hyodysenteriae, characterized by fever, debilitating mucohemorrhagic diarrhea and dehydration, leading to high mortality among animals. \u0000Proliferative enteropathy is a sporadic disease of pigs caused by Lawsonia intracellularis. The acute form of ileitis, known as proliferative hemorrhagic enteritis, is characterized by intestinal hemorrhage and sudden death, and usually occurs in pigs older than 4 months. \u0000Diarrhea in piglets can also be caused by enterococci (Enterococcus spp.) and chlamydia (Chlamydia suis). Therefore, the main aim of these studies was to determine the antibiotic susceptibility of microorganisms isolated from the pathological material. \u0000The article describes the susceptibility of isolated strains to the following antibiotics: ampicillin, amoxicillin, oxacillin, ofloxacin, streptomycin, cefazolin, gentamicin, amikacin, amoxiclav, erythromycin, enrofloxacin, lincomycin, tetracycline, doxycycline, vancomycin, rifampicin, ciprofloxacin, norfloxacin, tylosin, nitrofurantoin, polymyxin B. \u0000During the study of six Escherichia coli and three Salmonella enteritidis strains, it was ascertained that they demonstrated different susceptibility to the studied antibiotics, in particular, only two E. coli isolates, which is 33,3 %, were resistant to streptomycin, 50 % of the isolated strains were susceptible. \u0000When determining the susceptibility, it was established that all isolated E. coli isolates were resistant to cefazolin, vancomycin, oxacillin, and 83,3 % were resistant to amoxicillin, ampicillin, vancomycin, amoxiclav, rifampicin. \u000033,3 % of Salmonella enteritidis isolates were moderately susceptible to streptomycin, amoxicillin, and ampicillin, and 66,7 % were sensitive to ciprofloxacin, norfloxacin, and enrofloxacin. \u0000All isolated S. enteritidis strains were found to be resistant to cefazolin, tylosin, vancomycin, oxacillin, lincomycin, and erythromycin, 100 % susceptible to ciprofloxacin, amikacin, and ofloxacin and 68,7 % moderately susceptible to amoxicillin and gentamicin.","PeriodicalId":21617,"journal":{"name":"Scientific and Technical Bulletin оf State Scientific Research Control Institute of Veterinary Medical Products and Fodder Additives аnd Institute of Animal Biology","volume":"22 1","pages":""},"PeriodicalIF":0.0,"publicationDate":"2022-12-02","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"78984404","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}
