Bermudagrass (Cynodon dactylon) has long been used for soil restoration in saline-alkali land. Urbanization and the compound planting pattern combining trees, bushes, and grasses induced shading are becoming one of the most significant environmental constraints on the management of bermudagrass, which directly affects photosynthetic characteristics. Salinity and shade have become the most important environmental constraints on lawn development and implementation. Previous studies have shown that the plant physiological response under combined stress was different from that under single stress. The purpose of this research was to investigate the effects of salinity stress, shade stress, and the combined stress on bermudagrass. Shade nets were used to simulate shade stress to 85% shade. The NaCl concentration gradient for salinity stress was 1.0% for 7 days, 1.5% for 7 days, and 2.0% for 13 days, respectively. The combined stress combines the two approaches mentioned previously. The results showed that the salinity stress significantly inhibited the plant height, leaf relative water content, chlorophyll content, the chlorophyll a fluorescence induction (OJIP) curve and other photosynthetic parameters of bermudagrass while increasing electrolyte leakage when compared with control. Shade stress significantly enhanced the plant height, chlorophyll content, electrolyte leakage, the OJIP curve, and other photosynthetic parameters. Under the combined stress, the plant height and relative water content did not change significantly, but the photosynthetic parameters such as chlorophyll content and the OJIP curve increased. Furthermore, under the combined stress, the photosynthesis-related genes were regulated. Salinity stress inhibited the photosynthetic ability of bermudagrass more than shade stress, while the combined stress exhibited a considerably better photosynthetic ability. Although ZF-HDs have been investigated in several model plants, no systematic studies have been reported in apple (Malus ×domestica). In this study, 14 putative ZF-HD genes were identified in the apple genome and characterized using bioinformatics tools. All members harbored complete canonical structures of the ZF-HD motif. Phylogenetic analysis demonstrated that ZF-HD genes in the genome of apple could be classified into four subfamilies, with high intragroup similarities. Gene-structure analysis revealed that although 11 MdZHDs had only one exon, MdZHD6 and MdZHD13 had two exons and MdZHD8 had six exons, suggesting limited variation among the apple ZHD genes. The expression profiles of MdZHD genes revealed their involvement in the growth and development of different tissues. Numerous binding sites for transcription factors, such as MYB, bZIP, and AP2, were found in the promoter region of the putative MdZHD genes. Nearly all putative MdZHDs were predicted to localize in the nucleus. Finally, the expression levels of the MdZHD genes under abiotic stress were examined in apple rootstock Malus hupehensis and the results showed that the expression of 10 MdZHD genes was induced in response to three abiotic stress factors. Exceptionally, the expression of MdZHD11 was not induced in response to any of the abiotic stress treatments, MdZHD12 was only induced in response to salt stress, and MdZHD7 and MdZHD9 were induced in response to both drought and salt stress. English walnuts were stored at 5, 15, or 25 °C, and at 40%, 60%, or 80% relative humidity within each temperature. Principal component analysis was used to compare taste, texture, and aroma attributes evaluated by the taste panel to objective indicators of English walnut quality including water activity, moisture content, free fatty acids, peroxide value, hexanal content, and kernel color. Temperature was found to significantly impact English walnut oxidation and perceived rancidity, whereas storage at high relative humidity affected English walnut texture and accelerated quality loss. Water activity was more strongly correlated to textural changes than moisture content. The effect of relative humidity was more pronounced at lower temperatures, leading to increased hydrolytic rancidity and free fatty acids. Peroxide value had higher and more significant correlation to sensory attributes related to rancidity than hexanal. Free fatty acids were not correlated to the rancid sensory attribute, but were significantly correlated to bitter. English walnuts stored at 5 °C with 40% or 60% relative humidity were associated with the sweet sensory attribute and L* value (light color). Kernel darkening was associated with bitter and rancid, but a causal relationship is unknown. Ortiz","doi":"10.21273/jashs05246-22","DOIUrl":"https://doi.org/10.21273/jashs05246-22","url":null,"abstract":"East African banana (Musa sp.) breeding efforts have focused mainly on enhancing ‘Matooke’ productivity through the development of high-yielding, pathogen-resistant cultivars with adequate stability to contribute to regional food security. Before a breeding program can recommend promising cultivars for release, they must pass the sensory screens; be evaluated in the target population environments; and the data analyzed for yield, adaptability, and stability. Twenty-four primary and secondary triploid hybrids [NARITA (N)] derived from ‘Matooke’ bananas, six triploid local ‘Matooke’ cultivars, and one exotic cultivar were evaluated for their yield, adaptability, and stability across the East African region at three highland sites in Uganda’s western and central regions, as well as at three sites in Tanzania’s northeastern and southern highlands regions, from 2016–19. A randomized complete block design with four replicates was used for multisite trials. The mixed-model restricted maximum likelihood/best linear unbiased prediction approach, along with additive main effect multiplicative interaction model biplots, were used to dissect and visualize genotype-by-environment patterns. Following the likelihood ratio test, both genotype and interaction effects were highly significant, confirming the influence of genotype and site heterogeneity for selecting specific and broadly adapted cultivars. N23 had the greatest yield across all sites associated with adaptability and stability, outperforming the overall mean yield of all genotypes by 34.2%. In Tanzania, N27 (second), N7 (third), N18 (fourth), N4 (fifth), N12 (sixth), and N13 (seventh); and in Uganda, N17 (second), N18 (third), N2 (fourth), N8 (fifth), N13 (sixth), N12 (seventh), N4 (eighth), and N24 (ninth) demonstrated good adaptability and stability, as well as high yield. Furthermore, the fungal pathogen Pseudocercospora fijiensis had no significant effect (P > 0.05) on yield, stability, and adaptability of the hybrids. As a result, they can be introduced into areas where black leaf streak constrains banana production significantly and threatens farmers’ livelihoods. The average site yield potential ranged from 9.7 to 24.3 t⋅ha–1 per year. The best discriminating sites for testing breeding clones were Lyamungo in Tanzania and Sendusu in Uganda. To improve commercial traits, many novel cultivars have been produced through hybridization by commercial breeders. However, the genetic relationship of most cultivars is unclear, thus hindering the progress of Denphal-type Dendrobium breeding programs. Therefore, the development of molecular markers is encouraged to identify different cultivars. In this study, based on the transcriptome database of the Denphal-type Dendrobium ‘Red Bull’, the polymorphisms expressed sequence tag-derived simple sequence repeat (EST-SSR) were developed from 100 pairs EST-SSRs that were randomly selected from the EST-SSR database. The genetic relationship of 42 Denphal-type Dendrobium cultivars was analyzed according to the developed EST-SSRs. Then, the transferability of EST-SSRs was analyzed by performing a relationship analysis of 40 Dendrobium species. The results showed that a total of 5174 potential EST-SSR markers were identified with 4486 unigene sequences, and 5289 primer pairs were successfully designed. Of the selected 100 pairs of EST-SSRs, a total of 86 pairs produced the expected polymerase chain reaction products of the primary screening, 58 pairs produced the expected fragment size, and 20 pairs showed polymorphisms. Furthermore, the dendrogram of 42 cultivars showed that at a genetic distance of 0.15, the cultivars collected were grouped into five clusters of three major clusters and two minor clusters; all these clusters had the same characters of each cluster. The transferability analysis showed that 18 of the 20 EST-SSR markers among the 40 Dendrobium species were polymorphic. However, associated research in this area is still lacking in rabbiteye blueberry (Vaccinium ashei). In this study, we identified miRNAs, predicted their target genes, performed functional enrichment analysis of the target genes, and screened for miRNAs related to phytohormone signaling and self-incompatibility. A total of 491 miRNAs were identified, of which 27 and 67 known miRNAs as well as 274 and 416 new miRNAs were found in the rabbiteye blueberry cultivars Brightwell and Premier, respectively. Compared with ‘Premier’, 31 miRNAs were upregulated and 62 miRNAs were downregulated in ‘Brightwell’. Kyoto Encyclopedia of Genes and Genomes (KEGG) functional enrichment analysis indicated that the 4985 target genes predicted were involved in biosynthesis of amino acids, plant–pathogen interaction, and spliceosome pathways. A total of 10, one, one, five, two, five, and two candidate miRNAs related to auxin, cytokinin, gibberellin, abscisic acid, ethylene, brassinosteroid, and salicylic acid signaling, respectively, in rabbiteye blueberry pollen were identified. Further analysis indicated that novel_miR_49 was a candidate miRNA related to self-incompatibility, and their target gene was maker-VaccDscaff21-snap-gene-21.37. In addition, the KEGG enrichment analysis of the target genes of novel_miR_49 showed that they were involved in the ribosome, aminoacyl-tRNA biosynthesis, and glycosylphosphatidylinositol-anchor biosynthesis pathways. The results revealed that the microRNAs of rabbiteye blueberry pollen regulated to phytohormone signal transduction and self-incompatibility signal transduction based on related to auxin, cytokinin, gibberellin, abscisic acid, ethylene, brassinosteroid, and salicylic acid signaling. However, soft rot disease (Pectobacterium carotovorum ssp. carotovorum) is devastating for this crop. This study explored the Amorphophallus resistance mechanism against soft rot disease by analyzing transcriptome data using a weighted gene coexpression network analysis. The RNA sequencing of plants infected for 0, 12, 24, and 48 hours produced a total of 52.25 Gb of clean reads. A total of 29,096 genes were divided into 34 modules. Six modules of interest with the highest correlation with the target traits were selected to elucidate the resistance genes and pathways. The selected modules were enriched in the α-linolenic acid metabolism, phenylpropane biosynthesis, plant hormone signal transduction, and plant pathogen interaction pathways. Ultimately, AmBGLU, AmCAML, AmCDPK, AmLOX, and AmRBOHD were identified as genes of interest in the four significantly related metabolic pathways for real-time fluorescence quantitative polymerase chain reaction verification. The determination of salicylic acid (SA) and jasmonic acid (JA) in Amorphophallus muelleri and Amorphophallus konjac that suffered from soft rot disease showed that SA and JA were involved in the A. muelleri and A. konjac defense response against soft rot disease. Methyl jasmonate treatment delayed the onset of A. konjac soft rot disease. It has traditionally been used as an ornamental and medicinal plant in several Asian countries. The rapid development of high-throughput sequencing technology makes it feasible to obtain complete chloroplast (cp) genome sequences and will deepen our understanding of evolution of G. jasminoides. In this study, we sequenced the complete cp genomes of two botanical varieties of G. jasminoides. The complete cp genomes of both botanical varieties of G. jasminoides showed highly conserved structures and the length was 154,954 base pairs (bp) for G. jasminoides var. radicans (GJR) and 155,098 bp for G. jasminoides var. grandiflora (GJG). A total of 132 and 133 genes were identified in GJR and GJG, respectively. The cp genomes of two newly sequenced G. jasminoides were further compared with two published G. jasminoides cp genomes. Multiple repeats and simple sequence repeats (SSRs) were detected among different genotypes of G. jasminoides. The intron sequences of rps16 and rpl16 genes were slightly divergent among four genotypes of G. jasminoides. Phylogenetic analyses based on the complete cp genome sequences showed that G. jasminoides was closely associated with Fosbergia shweliensis, with Coffea as their close relative. The University of Missouri Center for Agroforestry in New Franklin, MO, USA, hosts an eastern black walnut cultivar repository. This collection supports an ongoing breeding program to improve economic performance for nut production (kernel weight, nut quality, precocity, and yield). In 1996, 54 cultivars were grafted and planted in a series of experimental orchards for evaluation. From 2001 until 2015, trees were evaluated for 12 phenological and eight nut quality/yield traits. Economically relevant traits including kernel weight, kernel percentage, and precocity (total nuts produced age 6 to 10 years) ranged from 1.1 to 8.6 g, 5.3% to 39.3%, and 16 to 1522 total nuts per tree, respectively. Kernel percentage was positively correlated with kernel weight (r = 0.51) and precocity (r = 0.38). Precocity was negatively correlated with the first (r = −0.39) and last (r = −0.30) female bloom. Principal component analysis and biplot analyses revealed high levels of variation among the cultivars. The first two components explain 43.4% of the total variation. Nut dimensions (nut length and nut thickness), nut and kernel weight, and kernel percentage are the largest drivers of variation in the collection. Eigenvectors for precocity and kernel percentage load together and are orthogonal to kernel weight, suggesting these three important traits can be improved simultaneously. Also, nut length loads with kernel weight, providing a candidate indirect selection parameter to increase kernel weight. We quantified daily fruit sink strength per stem (st) at m days after transplanting (SSTm_st) by modeling the fruit growth curve. The daily total dry matter production (TDMm_st) was calculated and defined as the source strength. During an experiment lasting ≈250 days, the fruit set ratio [number of fruit harvested/number of flowers (FSRm)] decreased significantly with increases in both the weekly average SSTm_st from 9 days before anthesis (DBA) to 13 days after anthesis (DAA) and the weekly average fruit number (FRNm_st) from 9 to 1 DBA. FSRm increased significantly with increases in both the weekly average TDMm_st from 1 to 13 DAA and the weekly average source-to-sink ratio [source strength/fruit sink strength (SSRm_st)] from 5 DBA to 13 DAA. During the whole experimental period, significant positive correlations with FSRm were observed for TDMm_st and SSRm_st, and significant negative correlations with FSRm were observed for SSTm_st and FRNm_st. FSRm increased until approximately the time when the weekly average SSRm_st at 1 to 7 DAA (anthesis to 156°C⋅d−1) ranged from 1.0 to 4.0; then, it showed a saturation curve at SSRm_st values more than 4.0 (R2 = 0.81).
