A, Birteksöz Tan","doi":"10.16883/JFPIU.31198","DOIUrl":"https://doi.org/10.16883/JFPIU.31198","url":null,"abstract":"SUMMARY In this study, 5-chloro-1'-methyl-5'-nitro-3H-spiro[1,3-benzothiazole- 2,3'-indole]-2'(1'H)-one (3u) was synthesized by the reaction of 1-methyl- 5-nitro-1H-indole-2,3-dione (1m) with 2-amino-4-chlorothiophenol (2) in ethanol. The structure of 3u was confirmed by the spectral (IR, 1H NMR, HSQC-2D, LCMS-ESI) data and elemental analysis. The new spiroindolinone derivative 3u, along with previously reported spiroindolinone derivatives 3a-t bearing benzothiazole or 5-chlorobenzothiazole moiety were tested for in vitro antimicrobial activity against selected strains. Among the tested compounds, 3i and 3l displayed the highest efficacy against Staphylococcus aureus and Candida albicans. Only 3b was found to be significantly active against Staphylococcus epidermidis. 3a-n were evaluated for in vitro antituberculosis activity against Mycobacterium tuberculosis H37Rv, but most of the tested compounds showed weakly antitubercular activity. All compounds were also evaluated against some DNA and RNA viruses in CRFK, HeLa and HEL cells. Cytotoxicities of the tested compounds were generally very high compared to standards. *Correspondence: OZET Bu calismada 1-metil-5-nitro-1H-indol-2,3-dion (1m) ile 2-amino- 4-klorotiyofenol (2)'un etanollu ortamdaki reaksiyonundan 5-kloro-1'- metil-5'-nitro-3H-spiro[1,3-benzotiyazol-2,3'-indol]-2'(1'H)-on (3u) bilesigi sentezlenmistir. 3u nun yapisi spektral (IR, 1H NMR, HSQC- 2D, LCMS-ESI) bulgular ve elemental analiz ile kanitlanmistir. Yeni spiroindolinon turevi 3u, daha once rapor edilen benzotiyazol ya da 5-klorobenzotiyazol artigi tasiyan spiroindolinon turevleri 3a-t ile birlikte secilen bakteri suslarina karsi in vitro antibakteriyel aktivite icin test edilmistir. Test edilen bilesikler icinde 3i ve 3l Staphylococcus aureus ve Candida albicans'a karsi en yuksek etkinlik gostermistir. Yalniz 3b Staphylococcus epidermidis' e karsi onemli derecede etkili bulunmustur. 3a-n nin Mycobacterium tuberculosis H37Rv karsi in vitro antituberkuloz etkileri incelenmistir; ama test edilen bilesiklerin cogu zayif antituberkuloz etki gostermistir. Tum bilesikler CRFK, HeLa ve HEL hucrelerinde DNA ve RNA viruslerine karsi ayrica test edilmistir. Test edilen bilesiklerin sitotoksisiteleri standartlara kiyasla genellikle cok yuksektir. In this study, 6 districts of Istanbul have been chosen where honey is produced by beekeepers and the plant sources were identified and kept in ISTE (the Herbarium of the Faculty of Pharmacy of Istanbul University) herbarium. According to our observations, Trifolium species are the favorite plants for honey bees in Istanbul. The proposed method was based on the formation of a yellow colored ion-pair complex by the reaction of methyl orange with mirtazapine. The ion pair complex was extracted with dichloromethane. Under the optimum condition, the ion pair complex showed an absorption maximum at 427 nm. The linearity range for concentrations of mirtazapine was found to be 2.5-12.5 I¼g/mL. The proposed method is successfully applied to the determination of mirtazapine tablet formulations. OZET Mirtazapinin farmasotik preparatlarda tayini icin duyarli spektrofotometrik bir yontem gelistirilmistir. Gelistirilen yontem, mirtazapin ile metil oranj arasinda sari renkli iyon cifti kompleksi olusturmasi esasina dayanmaktadir. Iyon cifti kompleksi diklormetan ile ekstre edilmistir. Optimize edilmis kosullarda iyon cifti kompleksinin maksimum absorbsiyon degeri 427 nm olarak tespit edilmistir. Mirtazapinin dogrusallik araligi 2.5-12.5 I¼g/mL olarak bulunmustur. Gelistirilen yontemin tabletlerde mirtazapin miktar tayininde guvenle uygulanabilir. The field works have been done from March 2010 to November 2010. During this research 52 villages have been visited and about 500 plant specimens were collected. As a result of this study's, 147 taxa of which 10 endemic have been determined for several usages. Among them 92 taxa as medicine, 52 as food, 8 as spice, 6 as dye, 9 as handicraft, 3 as fuel and 3 as tea. Additionally five plant species have been established for different usages. During this survey, besides to the usage of the wild flowering plants, their local names are also determined. This paper present list of 217 the vernacular names of useful 147 taxa that grow in Andirin. OZET Bu arastirma, Andirin (Kahramanmaras) ilcesinde bitkilerin halk arasindaki kullanimlarini saptamak amaciyla yapilan yuksek lisans tez calismasidir. Mart 2010 – Kasim 2010 tarihleri arasinda bolgeye toplam 5 arazi calismasi yapilmis, bu arazi calismalarinda toplam 52 koy, koylere bagli obalar ve daglar ziyaret edilerek toplamda ortalama 500 adet bitki ornegi toplanmistir. Arastirma sonucunda kullanilisi olan 147 takson tespit edilmistir. Bunlari 92 takson tibbi, 52 takson gida, 8 takson baharat, 6 takson boya, 9 takson el aletleri, 3 takson yakacak ve 3 takson bitki cayi olarak kullanilmaktadir. Bes turun ise diger farkli kullanimlari oldugu saptanmistir. Bu arastirma esnasinda bitkilerin kullanimlarinin yaninda yoresel isimleri de kayit edilmistir. Bu calismada Andirin ilcesinde kullanimi olan 147 taksona ait 217 yoresel isim listelenmistir. Nanotechnology based nanomaterials have been widely use in cosmetics for recent few years such as in sunscreens, hair products, skincare products, etc. However debate on their definition and insufficient quantification methods are major problems still occur in the nanomaterial field. Moreover the frequent use of cosmetics, safety of nanoscale ingredients of them has gain importance mainly by means of their dermal exposure. Although the proposed benefits that may occur by incorporating nanoparticles in cosmetics are increased efficiency, transparency, unique texture, protection of active ingredient, and overall higher consumer compliance, there still have not enough studies proved whether they are completely safe or not. As a conclusion major issues related nanomaterials such as developing, using and researching is going to increase in the near future due to their economically importance. Burm. fil. The lectin was purified 60 fold to apparent homogeneity in native polyacrylamide disc gel electrophoresis (PAGE) showing an apparent molecular weight of 45000 kDa. The fact that sodium dodecyl sulfate (SDS)-PAGE gave a subunit molecular weight of 14 400 kDa tends to propose that the lectin is composed of three subunits and thus is in agreement with Aloctin I previously partially purified and characterized by us. The lectin did not exhibit antioxidant effect as assessed by the DPPH· radical-scavenging assay. OZET Aloe vera (L.) Burm. fil. lektinin saflastirilmasi icin, yeni ve hizli bir kromatografi yontemi olan, ovalbumin bagli-siyanojen bromur (CNBr) ile aktive edilmis Sefaroz 4B, afinite kromatografisi tanitilmaktadir. Lektin, kromatografi sonucunda 60 kez saflastirildi. Poliakrilamid disk jel elektroforezi (PAGE) ile homojen oldugu gozlemlenen lektinin molekul agirligi bu yontemle, 45 000 kD olarak saptandi. Sodyum dodesil sulfat (SDS)-PAGE ile altbirim molekul agirligi 14 400 kDa olarak belirlenen lektinin, uc alt birimden olustugu ve bunun da daha once tarafimizdan kismen saflastirilmis ve karakterize edilmis olan Aloctin I ile uyum icinde oldugu sonucuna varildi. Saflastirilan lektin, DPPH· radikal supurme tayini ile degerlendirildiginde antioksidan etki gostermedi. Polymorphisms with physiological significance exist in the ADH and ALDH genes to different degrees in different ethnic groups. Specifically, ADH1B*2 (ADH1B*47His), ADH1B*3 (ADH1B*369His), ADH1C*1 (ADH1C*349Ile) and ALDH2*2 (ALDH2*487Lys) variants have been shown to confer protection against alcohol toxicity and dependence. During the past decade, the prevalence of alcoholism in Turkey has significantly increased with the economic development and industrialization of the country. Thus, the aim of this study was to investigate genetic differences in polymorphisms of alcohol-metabolizing enzymes, ADH1B, ADH1C and ALDH2, for Turkish population by using polymerase chain reaction (PCR)-restriction fragment length polymorphism (RFLP) method. Also, the frequencies of alleles were compared with those obtained from different populations. In conclusion; the allelic frequencies for ADH1B*2, ADH1B*3, ADH1C*1 and ALDH2*2 were observed 0.049, 0.004, 0.397 and 0.018, respectively. Turkish people resemble Caucasians while being different from Asians in terms of the distribution of ADH1B*2, ADH1C*1, ALDH2*2 alleles. Furthermore, Turkish people have less ADH1B* 3 allele than the other populations, especially African-Americans. Total phenolic and flavonoid contents of these extracts were determined as pyrocatechol and quercetin equivalents, respectively. The antioxidant activity of the extracts was investigated by using four methods including DPPH free radical and ABTS cation radical scavenging, I²-carotene bleaching and cupric reducing antioxidant capacity (CUPRAC) assays. The methanol extract showed the highest antioxidant activity in two assays. The anticholinesterase effect of the extracts was determined by using Ellman method via inhibition of acetyl- (AChE) and butyryl-cholinesterase (BChE) enzymes. The strongest inhibition against BChE was observed for the methanol extract. The antioxidant and anticholinesterase potential of bilberry bloom extracts was determined for the first time. OZET Bu calismada, yaban mersini bitkisinin cicek kisimlarinin petrol eteri, diklorometan, metanol ve sicak su ekstreleri hazirlandi. Bu ekstrelerin toplam fenolik madde miktarlari pirokatekole esdeger, toplam flavonoit madde miktarlari kersetine esdeger olarak belirlendi. Antioksidan tayin yontemleri olarak, DPPH serbest radikal ve ABTS katyon radikali giderim, I²-karoten renk acilim ve CUPRAC yontemleri kullanildi. Metanol ekstresi bu yontemlerin ikisinde en yuksek antioksidan aktiviteyi gosterdi. Ekstrelerin antikolinesteraz aktiviteleri, asetilkolinesteraz ve butirilkolinesteraz enzimlerinin inhibisyonuna dayanan Ellman yontemi ile belirlendi. Metanol ekstresi butirilkolinesteraza karsi en yuksek aktiviteyi gosterdi. Yaban mersini bitkisinin cicek kisimlarinin ekstrelerinin antioksidan ve antikolinesteraz aktiviteleri ilk kez bu calisma ile belirlendi. Alper","doi":"10.16883/JFPIU.32047","DOIUrl":"https://doi.org/10.16883/JFPIU.32047","url":null,"abstract":"SUMMARY The present work was designed to examine possible effects of methiocarb on lipid peroxidation, reduced glutathione (GSH), antioxidant enzymes such as superoxide distumtase (SOD), catalase (CAT), glutathione peroxidase (GSH-Px) ve glutathione reductase (GSH-Rd) and histological changes in Wistar albino rats after 5-day exposure. Moreover, we evaluated possible protective effects of vitamin E and taurine on methiocarb-induced oxidative damage. Fourty-eight rats were randomly divided into six groups as follows: control; methiocarb; vitamin E; vitamin E+methiocarb; taurine and taurine+methiocarb. 5-day exposure of methiocarb (1/10 of LD50) significantly increased lipid peroxidation in liver and kidney when compared to control group. Levels of GSH and activities of SOD and CAT were found to be decreased, while GSH-Px increased and GSH-Rd remained unchanged in rat tissues treated with methiocarb. Pre-treatment of vitamin E (100 mg/kg b.w.) and taurine (50 mg/kg b.w.) resulted in a significant decrease on lipid peroxidation, alleviating effects on GSH and antioxidant enzymes. Administration of methiocarb caused to the degenerative changes in liver and kidney tissues. Pre-treatment of vitamin E and taurine were partially protected both tissues against methiocarb-induced liver and kidney injury. In conclusion, the study has demonstrated that 5-day methiocarb exposure in Wistar rats caused oxidative damage on liver and kidney which were partly ameliorated by pre-treatment of vitamin E and taurine. Özçelik","doi":"10.16883/JFPIU.43215","DOIUrl":"https://doi.org/10.16883/JFPIU.43215","url":null,"abstract":"SUMMARY From the roots of pale purple-flowering Aconitum orientale sample, the norditerpenoid alkaloids septentriodine, lappaconitine, finaconitine, ranaconitine, puberanidine and delstaphinine together with the new alkaloid aconitorientaline obtained for the first time from the white-fowering sample, were isolated. All alkaloids isolated from the white-flowering sample were also obtained from the pale purple-flowering Aconitum orientale sample.All these results show, although they have different flower colors, both samples belong to Aconitum orientale and they are not different species. OZET Kirli-mor cicekli Aconitum orientale koklerinden norditerpenoid alkaloitler septentriodin, lappakonitin, finakonitin, ranakonitin, puberanidin, delstaphinin ve ilk defa beyaz cicekli Aconitum orientale koklerinden yeni madde olarak izole edilen akonitorientalin izole edilerek tanimlanmislardir. Beyaz cicekli Aconitum orientale koklerinden elde edilen tum alkaloitler yeni madde de dahil olmak uzere kirli-mor cicekli ornegin koklerinden de elde edilmistir. Tum bu sonuclar cicek renkleri farkli olmalarina karsin her iki ornegin de Aconitum orientale turune ait oldugunu ve bu orneklerin farkli turler olmadigini kanitlamaktadir.
