Here, we investigate their antibacterial activity, action at the transcript level and effects on membrane and biofilm to better control L. monocytogenes.</p><p><strong>Methods and results: </strong>RNA sequencing data revealed that the OPAC and DOPAC treatments to L. monocytogenes led to the identification of enormously differently expressed genes in cellular components (membrane) and phosphotransferase system pathway at their 1/2 MIC (7.805 mM l-1). {"title":"Genetic relatedness and antimicrobial resistance profiles of β-lactamase-producing Escherichia coli from environmental and animal sources in West Texas: a One Health approach.","authors":"Yamima Tasnim, Md Kaisar Rahman, Babafela Awosile","doi":"10.1093/jambio/lxaf190","DOIUrl":"10.1093/jambio/lxaf190","url":null,"abstract":"<p><strong>Aims: </strong>We aimed to determine the genetic relatedness among β-lactamase-producing Escherichia coli isolated from soil, lake water, and feces of geese, pigs, dogs, cattle, coyotes, wild hogs, and horses at one health interface in West Texas.</p><p><strong>Methods and results: </strong>Previously isolated 56 β-lactamase-producing E. coli isolates from the feces of geese, pigs, horses, coyotes, dogs, cattle, and wild hogs, and soil and lake water from different locations in West Texas were analyzed for genetic relatedness using whole-genome sequencing, core genome multilocus sequence typing, and phylogenetic single-nucleotide polymorphism (SNP) analysis using the publicly available bioinformatic platforms. Among 56 β-lactamase-producing E. coli isolates, 82.1% carried blaCTX-M genes, 42.86% carried blaTEM genes, 7.14% carried blaOXA-10, and 3.57% contained blaCMY-2 genes. Nine different types of phylogroups, 35 serotypes and 36 sequence types, 32 distinct plasmid groups, 75 types of virulence genes, and 46 core genome multilocus sequence types were identified. Using 100 maximum of SNPs between the isolates to define a clone, 14 clones were identified in 13 E. coli isolates from animal feces and environmental samples. Infantis) against acid, oxidative, increased osmolarity, and thermal stresses.</p><p><strong>Methods and results: </strong>25 strains isolated from human, food, and veterinary sources in Brazil between 2013 and 2018 were analyzed. 8 log10 CFU ml-1 inoculums were exposed for 10 min, 1 h, or 24 h to acid stress (HCl pH 2.6); oxidative stress (15 mmol l-1 H2O2); NaCl 9%; refrigeration (4°C); freezing (-20°C); and heating (63/74°C). Most strains (n = 22) survived the acid stress after 10 min and 1 h. Strains maintained high populations under oxidative stress (>87.7%) and NaCl 9% (111.4%). No strains survived heating, while refrigeration prevented multiplication (∼101%), and freezing reduced populations (<92.6%). Strain 1143/14 had its transcriptome sequenced under acid and oxidative stresses and NaCl 9%. 14 up-regulated and 38 down-regulated transcripts under acid stress, and 407 up-regulated and 143 down-regulated under oxidative stress were expressed, associated with biological processes of response and adaptation. No transcripts were differentially expressed with NaCl 9%.</p><p><strong>Conclusions: </strong>Salmonella Infantis survival against most stresses represents a challenge for its control. The efficacy of heating reinforced its importance to control Salmonella in food. When the concentration of CaCO3 was 50 mmol·L-1, the cumulative hydrogen production was 3726.27 mL·L-1, which was 193% higher than that of the control group. By exploring the effects of adding CaCl2, Na2CO3, and pH control on the hydrogen production of C. butyricum, it was found that the main reason why CaCO3 promoted the hydrogen production of C. butyricum was that it could control the pH of the fermentation broth within a suitable range. Transcriptomic analyses were performed on C. butyricum with and without the addition of CaCO3, respectively. After the addition of CaCO3, a total of 222 genes were significantly expressed, of which 141 genes were significantly upregulated and 81 genes were significantly downregulated. Among them, genes related to carbohydrate metabolism were upregulated (pgm by 1.95-fold, eno by 2.33-fold, and pflD by 3.43-fold).</p><p><strong>Conclusions: </strong>CaCO3 can accelerate the process of glycolysis by upregulating the pgm and eno genes, and further promote the increase in hydrogen production by upregulating the pflD gene. Listeria monocytogenes was detected more frequently in cold smoked (4.9%) compared to hot smoked (2.0%) samples. The majority (63%) of samples had a water activity that would not prevent the growth of L. monocytogenes. Elevated levels of Enterobacteriaceae (>10 000 CFU g-1) were detected in 4.7% of samples. Listeria monocytogenes was detected more frequently in samples with elevated levels of Enterobacteriaceae. Isolates of L. monocytogenes were compared using whole genome sequencing to historic isolates. Isolates from three samples were the same strain as that associated with a clonal complex (CC) 217 outbreak under investigation and detected despite an earlier product recall. Twelve isolates were CC121 strains and 10 of these belonged to six single nucleotide polymorphism single-linkage clusters with one strain linked to a human case from 2021. One CC415 strain was also linked to a human case. The remaining isolates belonged to CC8, CC9, CC14, CC37, CC31, CC398, CC87, and CC475 with no links to human cases. Investigation of historical isolates revealed that 11 strains were associated with specific manufacturers for up to 10 years. The presence of quaternary ammonium compound efflux genes showed an association with CC.</p><p><strong>Conclusions: </strong>Diverse L. monocytogenes detected in smoked fish with persistent strains associated with some manufacturing premises. The genomes were used for in silico phylogenetic analysis (cgMLST, SNP, ST410 clade analysis), ARG screening and for comparative analysis of complete plasmids. Antimicrobial susceptibility was determined by MIC measurement. All veterinary strains (CA, veterinarian) belonged to the phylogenetic clade ST410-B2 and the human strains to either ST410-B2 or B3. Strains recovered from the veterinary setting were clonal, differing by 0-4 SNPs. A higher genetic distance (>131 SNPs) was observed between the veterinary and the clinical human strains. All veterinary and 15 human strains shared an identical IncX3 plasmid harbouring blaOXA-181. Additional multidrug resistance plasmids were detected in human strains only.</p><p><strong>Conclusions: </strong>The Ec ST410-B2-OXA-181 lineage was detected in both humans and animals. Staphylococcus pseudintermedius biofilms are associated with poor prognosis, and rising antibiotic resistance threatens public health, making the development of new antibacterial agents imperative. Sanguinarine (SAN) is a plant-derived alkaloid with a wide range of pharmacological activities, including antibacterial, anti-inflammatory, and antitumour effects. The antibacterial potential of SAN against S. pseudintermedius merits further investigation.</p><p><strong>Methods and results: </strong>The minimum inhibitory concentration and minimum bactericidal concentration of SAN hydrogel against clinically isolated S. pseudintermedius were determined as 1.25 and 5 mg mL-1, respectively. Phenol-sulfuric acid and XTT assays demonstrated that SAN hydrogel significantly delayed biofilm formation (P < 0.05) by suppressing exopolysaccharide synthesis and biofilm metabolic activity. Live/dead fluorescent staining and scanning electron microscopy (SEM) confirmed its potent disruptive effects on mature biofilms, inducing bacterial lysis and death, while reducing biofilm thickness and extracellular matrix. In a murine skin infection model, SAN hydrogel (2.5/5 mg mL-1) exhibited significant antibacterial efficacy (P < 0.001), accelerated wound healing, and reduced bacterial load (P < 0.001) and IL-6 levels (P < 0.0001) in skin tissues. In addition, this pathogen has lower susceptibility to disinfectants used in hospitals, favoring its stay in such environments. Evaluate the action of eugenol and cinnamaldehyde against Candida auris.</p><p><strong>Methods and results: </strong>The products were evaluated through sensitivity tests of broth microdilution, as well as their possible mechanisms of action using proteomic analysis. For microdilution was used 24 strains and for synergistic interactions was used 2 strains. Cytotoxicity of these products was verified in Vero cells. Eugenol and cinnamaldehyde showed fungicidal action at concentrations ranging from 31.25 to 250 µg/mL. Combinations of eugenol and cinnamaldehyde with itraconazole showed either antagonistic effects or no detectable interaction. No cytotoxicity was found for eugenol at the concentrations that caused yeast growth inhibition. On the other hand, cinnamaldehyde was cytotoxic at the same concentrations. This study investigated the release of EVs from Fusarium oxysporum, noting significant differences in their structure and biological effects in both in vitro and in vivo.</p><p><strong>Methods and results: </strong>The study found that the size of these vesicles was consistent with patterns observed in other fungal species. Notably, the release of EVs from the planktonic form increased exponentially after 120 h of growth, resulting in heightened vesicle production. In biofilm, EVs released from the biofilm matrix showed distinct characteristics from those secreted externally, with a peak in release at 24 h. The concentration of EVs inside the biofilm was significantly higher, reflecting the complexity of biofilm dynamics. Additionally, EVs from the biofilm, particularly those isolated at 48 h of maturation, were highly cytotoxic, causing over 95% cell death in human cells. Bimolecular fluorescence complementation confirmed membrane-localized FfGpr1-FfG2 and FfG2-AC interactions, with intensified fluorescence at septa. Fermentation assays revealed opposing GA3 yields: Ffgpr1Δ produced 21% less GA3 than the wild type, whereas FfG2Q204 L increased yield by 17%. qPCR(quantitative real-time PCR) analysis demonstrated that Ffgpr1Δ upregulated FfCPS/KS, FfP450-2, and FfP450-3 transcription by 6-8-fold while downregulating FfDES by 82%, whereas FfG2Q204 L induced a 6-fold increase in FfCPS/KS mRNA level. Strikingly, FfDES overexpression in Ffgpr1Δ restored GA3 production to wild-type levels but led to GA7 accumulation and suppressed FfP450-3 upregulation, suggesting feedback-regulated metabolic constraints.</p><p><strong>Conclusion: </strong>Glucose-induced cAMP production required FfGpr1. FfGpr1-FfG2 and FfG2-AC interacted on the cell membrane, with enhanced co-localization at the septal region.
