Clarke leaf methanol extract on cancer cell lines. In vitro cytotoxicity of Andrographis lineata leaf extract was evaluated against A375 (melanoma), A549 (lung), HCT116 (colon), HeLa (cervix), HepG2 (hepatoma), Jurkat cells (T-cell acute lymphoblastic leukemia), MCF7 (breast cancer), PA1 (ovary), PC3 (prostate) and U87MG (Glioblastoma) cancer cell lines. Andrographis lineata leaf extract showed cytotoxicity activity against all cell lines, however, optimum cytotoxicity was observed against the ovarian cancer cell line (IC50=39.7 µg/ml) cell line. Cell cycle and terminal deoxynucleotidyl transferase dUTP end labeling assays confirmed the cytotoxic activity of Andrographis lineata leaf extract. Reverse phase-high performance liquid chromatography analysis was performed to recognize the active ingredient in the Andrographis lineata leaf extract and the results reveal that andrographolide was the major component. {"title":"Baicalein Suppressed Cervical Cancer Tumourogenesis by Modulating circ_0007364/microRNA-665/Tumor Protein D52 Axis","authors":"X. However, the mechanism of baicalein action has not been fully clarified in cervical cancer. Tumor protein D52, circ_0007364 and microRNA-665 expression were determined by quantitative reverse transcription-polymerase chain reaction. Tumor protein D52 and apoptosis-related proteins were detected by Western blot. Cell proliferation was measured via methyl thiazolyl tetrazolium, 5-ethynyl-2'-deoxyuridine and colony formation assays. The associations among microRNA-655, circ_0007364 and tumor protein D52 were determined by dual luciferase report assay. Cell invasion and apoptosis were assessed by Transwell assay and flow cytometry. The function experiments were carried out in vivo and in vitro . Baicalein significantly decreased circ_0007364 and tumor protein D52 expression and increased microRNA-665 expression in cervical cancer cell samples. Circ_0007364 or tumor protein D52 introduction and microRNA-665 down regulation weakened the suppressive effects of baicalein in cervical cancer cells. Moreover, circ_0007364 functioned by binding to microRNA-665 to regulate tumor protein D52 and affect cellular process in cervical cancer. Methanol was employed as the solvent in this proposed process, and detection was carried out at a wavelength of 248 nm. The validation parameters linearity, limit of detection and limit of quantitation, accuracy, precision and specificity were all the subject of experiments according to International Council on Harmonisation guidelines. The stability of flurbiprofen was examined under acidic, alkaline, thermal, and photolytic conditions. The kinetics in the concentration range of 2-10 ug/ml were defined as linear with a correlation value of 0.992. The percentage of relative standard deviation for both intra-day and inter-day accuracy was under less than 2 % and 99.6 %-100.4 % of the drug was retrieved from the study. The system was able to identify variations related to stress conditions although the stress condition of degradation products hadn't been determined. The proposed approach can be employed as a stability indicating method for flurbiprofen screening in conventional pharmaceutical analysis of eye drops with a significant level of linearity, accuracy and precision. The method was found to be both affordable and convenient. A total of 74 patients with perianal and anal canal condyloma acuminatum who were admitted to our hospital from January 2021 to December 2021 were selected as the research objects. They were randomly assigned to the observation group and the control group, with 37 cases in each group. The observation group was treated with 5-aminolevulinic acid photodynamic therapy combined with carbon dioxide laser and the control group was treated with carbon dioxide laser therapy. Both groups were treated once a week for 3 w. The therapeutic effect, recurrence rate and adverse reactions were compared between the two groups. The cure rate of condyloma acuminatum in the observation group was 86.48 %, which was higher than 54.05 % in the control group, with statistical significance (p<0.05). After follow-up, it was found that the recurrence rate of the observation group (13.51 %) was significantly lower than that of the control group (43.24 %), with statistical significance (p<0.05).
