{"title":"How toxicity drives evolution: past, present and future.","authors":"Jorge Herkovits","doi":"10.1007/s10646-025-02918-4","DOIUrl":"https://doi.org/10.1007/s10646-025-02918-4","url":null,"abstract":"<p><p>The evolutionary context of environmental toxicology and chemistry in the Anthropocene is expanded towards a global perspective through three complementary approaches: (i) environmental toxicity as a mechanism to explain selectivity in mass extinctions with the K-Pg event as a case study; (ii) evidence that certain features of embryonic development could be considered biomarkers of adaptation to specific environmental agents of the evolutionary process, e.g. the transition from the anaerobic to the aerobic metabolism in early embryonic stages versus from the anoxic toward the oxic Earth, and (iii) the capacity to build favorable conditions for life by the collaboration of a population of free-living embryos reducing environmental toxicity and its evolutionary implications from an onto-phylogenetic perspective. Focusing on the future, the article highlights the relevance of environmental toxicology and chemistry for sustainability, to understand the characteristics of eventual extraterrestrial life better, and for space colonization initiatives. The Mexican salamander, Ambystoma dumerilii is endemic to Lake Pátzcuaro, Michoacán, Mexico and is exposed to nutrient runoff and discharges from surrounding agriculture and municipal waste. This study evaluated the toxicity of nitrate and phosphate singly and in mixtures to embryonic and the first-stage larval development of A. dumerilii to determine whether measured surface water concentrations comparable to those found in Lake Pátzcuaro pose a risk and whether Mexico's water quality standards are adequate to protect the species. Eggs and larvae of A. dumerilii were exposed for 26 days to nitrate and phosphate individually as well as to dilutions of a mixture based on their LC<sub>50</sub> concentrations. The Probit determined LC<sub>50</sub> for NO<sub>3</sub>-N was 887 mg/L and for PO<sub>4</sub>-P was 2015 mg/L. The LC<sub>50</sub> for the mixture was 1.0 toxic unit (TU), while the EC<sub>50</sub> for the hatching inhibition mixture was 0.26 TU. Effect thresholds were 0.1 and 0.04 TU respectively. Concentrations of nitrate and phosphate mixtures in selected Mexican surface waters were above the hatching inhibition threshold and the equivalent Mexican standards expressed as mixtures also exceeded this threshold. The toxicity responses to the mixtures suggest that nitrate and phosphate anions compete for the same sites and act in an additive manner. Nitrate and phosphate need to be managed as mixtures rather than singly to protect species with similar sensitivities to A. dumerilii. However, there is limited knowledge about the adverse effects of composite MPs on soil-plant systems. To address this lack of data, a pot experiment was conducted using polyethylene (PE), polypropylene (PP), polyvinyl chloride (PVC), and polyolefin (PO) in various combinations: PP-PE-PVC, PP-PE-PO, PE-PVC-PO, PVC-PO-PP, and PP-PO-PE-PVC. The study investigated composite MPs effects on soil properties and physiological-biochemical indicators of Chinese cabbage (Brassica napus L.). The results indicated that 2% concentration of different types of composite MP pollution significantly increased soil pH while reducing soil water content, organic matter, ammonium nitrogen, nitrate nitrogen, available phosphorus, sucrase activity, urease activity, and soil microbial numbers. Soil catalase (CAT) activity initially increased and then decreased. The effect of composite MP pollution on Chinese cabbage revealed that it significantly reduced water content, plant height, and chlorophyll content, while promoting root length growth. Additionally, it had a negative effect on the activities of CAT, peroxidase, and superoxide dismutase activity in Chinese cabbage, with a significant declines. The study found that the degree of risk associated with composite MP pollution was related to the types of MP combinations. An experiment was conducted to evaluate the combined effects of these contaminants on survival, growth, hematology, intestinal and liver histology, oxidative stress, and immunity of Nile tilapia (Oreochromis niloticus). Fish were exposed to T1 (10 mg/L polyamide microplastic; PA-MP), T2 (8 mg/L hexavalent chromium; Cr-VI), T3 (8 mg/L Cr-VI + 10 mg PA-MP) with a control group in triplicates. After 42 days of exposure, it was observed that the ingestion of microplastics in the gastrointestinal tract (GIT), feed conversion ratio (FCR) and mortality were highest in T3 with the co-exposure of Cr-VI with PA-MP. There was an increasing trend in blood glucose concentration and a decreasing trend in hemoglobin concentration observed from T1 to T3. Nuclear and cellular abnormalities became more frequent and histological observation was also aggravated in T3. The relative mRNA expression of antioxidant genes like SOD (superoxide dismutase) and CAT (catalase) was significantly upregulated in T2 and T3 and the immune genes such as TNF-α (Tumor necrosis factor- α), IL-1β (Interleukin-1β) and IFN-γ (Interferon- γ) were also significantly upregulated in T2 and T3 with the exposure of Cr-VI alone and co-exposure of Cr-VI and PA-MP. The guideline monitors soil microbial nitrogen transformation over 28 days, potentially missing long-term effects of persistent pesticides. Additionally, nitrate alone may be not sensitive enough to detect disruptions in microbial functions. We investigated whether functional gene analysis could provide a more sensitive bioindicator of pesticide impact. To compare this method with the standard test, we conducted a microcosm experiment following the OECD 216 experimental setup. To capture long-term effects beyond the typical test period, we extended the incubation duration to 56 days. Four different concentrations of the persistent fungicide boscalid were added based on predicted environmental concentration. We also assessed microbial responses to fungicide exposure by measuring classical soil microbial parameters. According to the standard test, boscalid had no harmful long-term effects on soil microbiota. In contrast, our analysis of functional genes found an overall reduction in the acid phosphatase-encoding phoN gene abundance on Day 56, and correspondingly, in acid phosphatase activity in the highest fungicide treatment. Simultaneously, we observed a tendency towards lower fungal abundance based on measured copy numbers of an ITS region of nuclear ribosomal DNA (rDNA) and increased cumulative CO<sub>2</sub> production. These results indicate a fungicide-related response of the microbial community and impaired microbial phosphorus cycling.
