The young (7 days) explants were found to be suitable for culture and highest (100%) callus induction was obtained in MS medium supplemented with 5.0 mg/l BAP with 1.0 mg/l NAA. The maximum shoot regeneration (95%) and the number of shoots (10) were observed in MS medium supplemented with 6.0 mg/l BAP with 0.3 mg/l NAA. The highest shoot multiplication frequency (95%) and the number of shoots (60) were obtained in MS medium supplemented with only 2.0 mg/l BAP. The well-developed highest number of roots (5.0) per plant and average root length (5.0 cm) with 100% root induction efficiency without callusing was recorded in the half strength of MS medium devoid of any hormonal supplement. The efficiency of EC and SEs from the protocorms significantly relied on the concentration of PGRs. MS medium supplemented with TDZ (1.0 mg/l) induced optimum of EC (31.8%) and SEs (28.1/explant). Similarly, ZEA (0.5 mg/l) induced EC (27.6%) and SEs (17.1/explant). The combined treatment of TDZ (1.0 mg/l) and NAA (0.5 mg/l) resulted in the maximum induction of EC (58.6%) and SEs (39.8/explant) in an upright incubated explant. In another combined effect of ZEA (1.0 mg/l) and NAA (0.5 mg/l) induced EC (43.8%) and SEs (24.3/explant), whereas the combination of BAP (0.5 mg/l) and NAA (0.5 mg/l) produced EC (34.4%) and SEs (16.8/explants). The induced EC and SEs where healthier on medium containing TDZ + NAA than on the medium containing ZEA+NAA and BAP+NAA. The orientation of lTCL explants seemed to interact with position to affect the morphogenesis. The lTCL explants incubated in the upright orientation induced higher percentage of EC and a greater number of SEs/explants (EC-58.6% and SEs-39.8/explants) than that in the inverted orientation (EC- 16.4% and SEs-9.1/explants) irrespective of the PGRs. The SEs developed from the EC and the intermediate stage of PLBs were finally differentiated into plantlets. Different types of genotypes of lotus found in Bangladesh which represent a great variability. However, investigations on the variability of the lotus at genetic level are insufficient and limited in Bangladesh. In the current study, fourteen samples of lotus were collected from six different locations of Bangladesh were subjected to molecular analysis using codominant Simple Sequence Repeats (SSRs), as well as, dominant Random Amplified Polymorphic DNA (RAPD) markers to estimate the genetic diversity, and to test the genetic basis relationships among the germplasm. Analysis of SSRs showed a moderate level of polymorphism which was 68.12% and Nei’s genetic distance was ranging from 0.0328 to 0.7598 showing a significant level of variation among the genotypes. The germplasm collected from Kapasia identified as different and superior among the morphotypes. Moreover, RAPD analysis showed 60.44% polymorphism and Nei’s genetic distance was ranging from 0.0600 to 0.5743 indicating a moderate level of variability where yellow lotus was identified as distinct. Two Neighbor-Joining dendrograms were constructed using UPGMA method in which the germplasm were clustered into two main clades with 55% genetic similarity level for both RAPD and SSR markers. The current study was successful in establishing an efficient method for evaluating the variation and the genetic marker system to be used to identify core N. nucifera accessions. This study aims to produce the genetically identical plantlets of Dendrobium crepidatum Lindley and Paxton using in vitrodeveloped shoot explants. Coconut water (5 - 10%) supplemented Murashige and Skoog (MS) medium responded towards effective shoot formation with BAP (1.0 - 2.0 mg/l) and NAA (0.1 - 0.5 mg/l). NAA (0.5 - 1.0 mg/l) alone or in combination with BAP was found to be effective for root initiation in MS medium. Four RAPD (Random Amplified Polymorphic DNA) primers viz., G04, G10, G17 and G18 showed 100% genetic homogeneity in plants obtained from in vitro-developed shoot explant. However, G10, G17 and G18 RAPD primers showed 60, 25 and 100% genetic polymorphisms respectively in plants obtained from callus. Plants obtained from callus were genetically polymorphic where somaclonal variation might occur. This study reported the successful genetic homogeneity of shoot explant-derived plants of D. crepidatum. Cotyledonary leaf explants of two BINA varieties of tomato were cultured on MS medium containing different concentrations and combinations of BAP, Kn and IAA for multiple shoot formation via organogenesis. 100% shoot induction was observed when the explants were cultured on MS medium supplemented with BAP, Kn and IAA. Earliest shoot induction was observed within four weeks. The MS media supplemented with 1.5 mg/l BAP + 0.5 mg/l Kn + 0.2 mg/l IAA found to be the best medium with regards to number of days required for the initiation of regeneration, no. of multiple shoot formation and regeneration frequency in case of both the varieties. Shoot elongation was obtained in the same regeneration medium. The elongated shoots were rooted on half strength MS medium supplanted with 0.2 mg/l IBA. The in vitro rooted plantlets were successfully established in soil with 97% survival rate. This AI-MAS process reactor consisted of a small anaerobic compartment (first stage) placed at the corner of the intermittent aeration tank (second stage) with a magnetic separator in the middle. Better sludge separation was observed at 1-6 rpm of the magnetic drum. Using AI-MAS process, continuous removal of about 90% T-P, 96% T-N and 94% soluble CODCr were achieved with the loading rate of 3.5 mg/l T-P, 16 mg/l T-N and 250 mg/l CODCr, for the period of 160 d at the rate of 6 hrs hydraulic retention time (HRT). Simultaneous removal of phosphorus and nitrogen from the wastewater was possible due to coexistence of both phosphorus and nitrogen removal bacteria within the same sludge biomass. Phosphorus removal was performed through phosphate accumulation in aerobic condition and released again in anaerobic condition with the concomitant uptake of organic substances. However, all the banana varieties cannot be cultured in media with the same composition to get the optimum shoots in multiplication cycleswith desirable shoot qualities. Banana varieties like Robusta grow optimally in Murashige and Skoog medium containing 3-5 mg/L BA (6- benzyl adenine). Under the same condition, the growth of Nendran is found to be poor with a low number of rootable shoots. The poor multiplication ratio causes a higher cost for the micropropagated plantlets of var. Nendran. Hence the current study was carried out to optimize the shoot multiplication of the two varieties of banana (Nendran and Robusta) in MS medium supplemented with various combinations and concentrations of plant growth regulators. The multiplying shoots were cultured in MS medium supplemented with different concentrations of BA, combinations of BA, NAA (- naphthalene acetic acid) and combinations of KN (kinetin) and NAA. Combinations of BA and NAA (0.5 mg/L BA+0.1 mg/L NAA or 0.5mg/L BA+0.5 mg/L NAA) was found to result in responses ideal for commercial production of var. Nendran. For the var. Robusta, 2 mg/L BA was ideal for shoot multiplication and 1 mg/L BA+0.5 mg/L NAA could be optimal for use at elongation stage to get maximum desirable shoots for rooting. The study could optimize a suitable plant growth regulator regime for the commercial production of banana var. For callus induction, leaf explants were inoculated on MS medium containing different concentrations of 2,4-D. Highest callus induction frequency (90%) was recorded on MS medium supplemented with 4.0 mg/l 2,4-D. Remarkable results on indirect shoot organogenesis and multiplication (86.67%) with maximum shoot number per unit callus (11.64 ± 0.37) and shoot length (10.13 ± 0.24 cm) were observed when the leaf derived callus were transferred on MS medium fortified with 1.5 mg/l BAP and 0.5 mg/l NAA. Additionally, the incorporation of 10% coconut water with the medium showed satisfactory shoot growth and development. The best response towards root induction (85%) was achieved on ½ MS medium fortified with 3.0 mg/l IBA, 2.0 mg/l IAA and 2.0 mg/l NAA with an average 22.20 ± 0.87 roots per unit shoot. Amylases and proteases are among the most important enzymes. During this study, indigenous amylase and protease producing bacteria were isolated from common decayed fruits and vegetable wastes viz. Papaya, Brinjal, Cucumber, Potato and Snake Gourd. Bacterial load ranged in between 0.67 × 109 and 9.06 × 109 cfu/g on NA and 1.50 × 109 and 7.00 × 109 cfu/g on PYG agar medium. Maximum mean bacterial load on both NA (6.19 ± 2.60 × 109 cfu/g) and PYG agar (4.54 ± 1.03 × 109 cfu/g) were observed in decayed papaya. A total of 113 bacterial isolates were primarily isolated. Considering better amylase and protease activity 16 isolates were selected for detailed study. The starch hydrolysis ratio (SHR) of the isolates ranged in between 1.25 ± 0.37 and 2.47 ± 0.23 while casein hydrolysis ratio (CHR) ranged in between 2.35 ± 0.12 and 6.44 ± 1.16. The highest SHR was 2.47 ± 0.23 found in Bacillus sp. of snake gourd and the highest CHR was 6.44 ± 1.16 found in Bacillus subtilis also from snake gourd. Out of the 16 isolates 13 were Gram positive and 3 were Gram negative. Gram positive isolates were identified as Bacillus acidocaldarius (4), B. firmus (2), B. lentus (4), B. subtilis (2) and B. alcalophilus (1). Gram negative isolates were identified as Edwardsiella hoshinae, Proteus myxofaciens and P. mirabilis. Six isolates having higher SHR and CHR were authenticated through molecular identification and were identified as Chryseobacterium sp. S29.2, Bacillus sp. X8, Bacillus sp. strain GA1B, Bacillus sp.TdEND26, Bacillus subtilis strain BPA28 and Bacillus subtilis BAB-881. The maximum amylase (61.33 ± 2.14 U/ml) and protease (56.91 ± 0.23 U/ml) production were observed in Bacillus sp. TdEND26 rd. In case of co-production, the highest amount of amylase (54.13 ± 1.23 U/ml) protease (81.80 ± 4.54 U/ml) production was observed in Bacillus sp.
