It is mainly caused due to an imbalance between the physiological vasoconstrictor and vasodilator molecules. Nitric oxide and hydrogen sulphide are well established vasodilatory endogenous gasotransmitters that exhibit reduced production from intrauterine tissues during preeclampsia. Aims and objectives: The aim of the study was to determine the serum levels of nitric oxide and hydrogen sulfide in preeclampsia cases compared with age matched controls and to rule out, whether there is a significant relationship if any, between these two gasotransmitters in preeclampsia. Materials and methods: Serum levels of NOx and H2S were measured in 100 pregnant women with preeclampsia and the values were matched with controls. Results: The mean serum NOx levels of preeclamptic patients was 45.88 ± 17.72 µmol/L which were significantly lower (p<0.001) than those observed in control with a values of 161.09 ± 27.46 µmol/L. The mean serum H2S level was 32.31 ± 12.62 µmol/L in patients which was significantly lower (p<0.001) when compared to control where mean we observed 114.50 ± 20.35 µmol/L. A positive correlation exists between serum NOx and H2S levels in preeclampsia (r=0.691, p<0.001). Conclusion: The present study has elucidated that the serum levels of nitric oxide and hydrogen sulfide decrease in preeclampsia compared to normal pregnant women and these two molecules show a positive correlation in their levels in preeclampsia.","PeriodicalId":89896,"journal":{"name":"Biochemistry and analytical biochemistry : current research","volume":"52 1","pages":"1-6"},"PeriodicalIF":0.0,"publicationDate":"2019-01-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"69980509","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Pac, Iwuoha Ag","doi":"10.35248/2161-1009.19.8.381","DOIUrl":"https://doi.org/10.35248/2161-1009.19.8.381","url":null,"abstract":"Introduction: Hepatotoxicity can be caused by drugs overdose (in some cases even when the drugs are administered in therapeutic doses). This may damage the liver temporarily or permanently. The reason for this study is to evaluate the amount of damage caused by carbon tetrachloride (CCl4) on liver and establish a dose-dependent treatment and prevention with Justicia carnea leaves extract. Aim: Hepatoprotective effect of methanol extract of Justicia carnea leaves on carbon tetrachloride intoxicated albino rats was investigated. Methods: A total of thirty-five female wistar albino rats were used for the study and were grouped into 7 groups (I-VII) of 5 rats each. Group I served as normal control which received only feed and water, group II served as CCl4- treated control receiving 0.5 mg/kg bodyweight (bw) in of CCl4 alone in olive oil (1:1 v/v), group III served as extract control which received 1000 mg/kg bw of methanol extract of Justicia carnea (JC) only, groups IV, V and VI received 200, 500 and 1000 mg/kg bw of methanol extract of Justician carnea respectively prior to CCl4 administration while group VII received 100 mg/kg bw of silymarin as a standard drug prior to CCl4 administration. Some biochemical parameters were evaluated using biochemical methods. Results: The activities of aspartate transaminase (AST), alanine transaminase (ALT), alkaline phosphatase (ALP) and bilirubin levels showed a significant (p<0.05) increase in the group treated with CCl4 only when compared with the normal control while total protein and albumin levels showed a significant (p<0.05) decrease in the group treated with CCl4 only when compared with the normal control. However, groups pre-treated with different doses of 200, 500 and 1000 mg/kg bw of Justicia carnea showed a significant (p<0.05) decrease in the activities of ALT, AST and ALP, and bilirubin level while total protein and albumin levels increased significantly (p<0.05) when compared with the rats treated with CCl4 alone. Histopathological examinations of sections of the liver collected from the different groups showed that the normal control had a normal liver architecture compared to the group treated with CCl4 alone which showed a diffuse vacuolar degeneration and coagulative necrosis of the hepatocytes. However, groups pre-treated with different doses of the extract showed lesser necrosis and a better liver architecture compared with the group treated with CCl4 alone. Conclusion: The results of this study suggest that the methanol extract of Justicia carnea may exhibit hepatoprotective properties that may be effective in the management of liver toxicity.","PeriodicalId":89896,"journal":{"name":"Biochemistry and analytical biochemistry : current research","volume":"1 1","pages":""},"PeriodicalIF":0.0,"publicationDate":"2019-01-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"69980029","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Yoo","doi":"10.4172/2161-1009-c2-019","DOIUrl":"https://doi.org/10.4172/2161-1009-c2-019","url":null,"abstract":"Protein glycosylation, one of the most time-honored posttranslational modifications in proteins, plays vital roles in organic systems through a variety of processes, such as adhesion, signaling via cellular recognition, and response to unusual biological states. However, due to the complexity and heterogeneity of a glycoprotein, contemporary analyses focus in most cases on the identification of both glycosites or the released glycans only. In this study, we have developed MS-based high throughput technique for intact N-glycopeptides analysis, named GlycoProteomeAnalyzer (GPA) for analysis of N-and O-glycosylation in proteomics, which combines tandem Mass Spectrometry (MS) with a database search and algorithmic suite. We created novel scoring algorithms for confident identification of N- and O-glycosylation of proteins with calculation of False Discovery Rate (FDR). In our approach, all amino acid sequence as well as glycosylation website records had been got from the Uniprot database. From the Swiss-Prot accession variety of human protein, our GPA software automatically construct tryptic N- and O-glycopeptide database for the proteins in human plasma sample. It lets in automatic identification of site-specific N- and O-glycopeptides of protein combos using HCD, CID, and ETD MS/MS spectra with GPA- DB from Uniprot with estimated FDR ≤ 1%. GPA has been designed to easily handle high -throughput glycoproteomic statistics with a graphical user interface and verified on internet site (https://www.igpa.kr/). It can also be integrated with cloud computing carrier that eliminates the need for local clusters and will increase throughput of data analysis. Glycosylation, a particular top-down analysis, which affords the most direct method for obtaining records on glycans in a glycoprotein. By calculating the molecular weight variations between the peaks, it is viable to decide the types of glycan modifications on that protein. Such an analysis, however, regularly lacks sensitivity and structure information. Because of this, glycoprotein analyses are often divided into two primary strategies for accumulating glycoprotein information using MS techniques. One entails an evaluation of launched glycans, while the other includes characterizing glycopeptides that are got by proteolytic digestion of the unique glycoprotein.","PeriodicalId":89896,"journal":{"name":"Biochemistry and analytical biochemistry : current research","volume":" ","pages":""},"PeriodicalIF":0.0,"publicationDate":"2018-06-02","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"47958495","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Therefore, identification and characterization of placental components including proteins and their multi-protein complexes is an important step to understanding the placenta function. Here we analyzed for the first time human placentas extremely stable multi-protein complex (SPC, ~1000 kDa) by MALDI MS and MS/MS spectrometry using proteins tryptic hydrolyzates after proteins separation by SDS-PAGE and 2-D electrophoresis. The formation of such a very stable complex due to the random association of several various proteins is very unlikely. It was shown that SPCs contain twelve proteins: hemoglobin, alkaline phosphatase, cytoplasmic actin, human serum albumin, chorionic somatomammotropin hormone, heat shock protein beta-1, peroxiredoxin-1, 78 kDa glucose-regulated protein, protein disulfide isomerase A3, serotransferrin, annexin A5, and IgGs. These twelve proteins have in themselves many different and important biological functions, which can be inherent for these proteins in the complex. In addition, the complex demonstrated nine different enzymatic activities: DNase, RNase, ATPase, phosphatase, protease, amylase, catalase, peroxidase (H2O2-dependent) and oxidoreductase (H2O2-independent). The efficiency of the catalysis of each of these reactions by SPC preparations from three placentas was comparable. It was shown that hydrolysis of r(pU)23, r(pA)23, and r(pC)23 leads to the formation of 1-22-mer oligonucleotides, while digestion of microRNA mirR137 is a site-specific (3A-4U > 9U-10A > 8U-9U ≥ 15U-16A) resulting in the formation of only four major products. A large number of potentially possible functions of the complex in accordance with the functions of its individual proteins are considered. Progress in the study of placental proteins complexes can promote understanding of their biological functions.","PeriodicalId":89896,"journal":{"name":"Biochemistry and analytical biochemistry : current research","volume":"7 1","pages":"1-9"},"PeriodicalIF":0.0,"publicationDate":"2018-03-09","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"49360279","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

LA complex interacts with DNA in tumor cells with histones and impaired the chromatin structure. There are no any data on how LA recognizes DNA and interact with histones and DNA of chromatin. The approach of stepwise increase in ligand complexity was used for estimation of the relative contribution of every DNA nucleotide unit to its total affinity for human LA. It was shown that the LA DNA-binding site minimal ligands are orthophosphate and all dNMPs and rNMPs (Kd=(5.0-43)×10-5). Maximal contribution to the total affinity was observed for three nucleotide units of all (pN)n with a significant decrease in the order 1>2>3, at n=4-6 it was remarkably lower and at n ≥ 6-7 all dependencies of -logKd upon n reached plateaus. Double-stranded (pN)n showed significantly lower affinity comparing with singlestranded ligands. The thermodynamic parameters characterizing the specific contribution of (pN)1-6 every nucleotide link (ΔGo) to their total affinity for LA were estimated. The spatial model of LA-DNA complex was calculated. LA protein sequence has homology with those of five histones (H1-H4) involved in the chromatin nucleus interactions between themselves and their complex with DNA. It is assumed that the homology may be the main reason for the interaction of LA with chromatin DNA, leading to a breakdown in its structure, as well as the proper binding of histones between themselves and with DNA.","PeriodicalId":89896,"journal":{"name":"Biochemistry and analytical biochemistry : current research","volume":"07 1","pages":"1-8"},"PeriodicalIF":0.0,"publicationDate":"2018-01-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"https://sci-hub-pdf.com/10.4172/2161-1009.1000363","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"70555792","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}
