Radium-223 dichloride (Xofigo®, Bayer HealthCare Pharmaceuticals Inc.) is the first α-particle emitter therapeutic agent approved by the FDA, with benefits in overall survival and delay in symptomatic skeletal event for patients with metastatic castrate-resistant prostate cancer (CRPC). Recent post hoc analyses of the phase III ALSYMPCA trial support the previously established safety profile as well as therapeutic effect and clinical outcome of Radium-223. Currently, Radium-223 is approved as a single agent therapy for metastatic CRPC. Clinical trials are currently investigating Radium-223 in additional clinical settings such as earlier asymptomatic disease and in combination with other agents including hormonal therapeutic agents and immunotherapeutic as well as chemotherapeutic agents. Trials are also ongoing in patients with other primary cancers such as breast cancer, thyroid cancer, and renal cancer metastatic to bone. In this article, the physics and radiobiology, as well as a literature update on the use of Radium-223, are provided along with case presentations, aiming at a better appreciation of research data as well as the assimilation of research data into clinical practice.","PeriodicalId":89157,"journal":{"name":"International journal of molecular imaging","volume":"2016 1","pages":""},"PeriodicalIF":0.0,"publicationDate":"2016-09-27","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"https://sci-hub-pdf.com/10.1155/2016/2568031","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"64281468","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

AD is diagnosed with certitude at autopsy or via brain biopsy. However, it is now known that the AD pathological abnormalities, detected in vivo by biological markers, precede the clinical symptoms by years or even decades. The uniform lack of success in treating AD may be due to the fact that therapeutic intervention is begun too late in the course of the disease. The earliest recognizable pathological event in AD is aggregated cerebral amyloid-β deposition; this pathology may be present 20 years before the onset of dementia. A newly developed retinal camera visualizes amyloid in the retina and, through computer analysis, quantifies it yielding a number: the retinal amyloid index. The retina is an extension of the brain and early studies show that the amyloid deposition in the retinal correlates with cerebral amyloid deposition. This article introduces this imaging technology and suggests a role for early detection of the pathology associated with AD. Moreover, the utility of this technology in monitoring interventional trials is suggested. If the sensitivity and specificity of this imaging of retinal amyloid is successful enough to be embraced by the research community, it may ultimately be considered a “virtual brain biopsy.","PeriodicalId":89157,"journal":{"name":"International journal of molecular imaging","volume":"64 1","pages":"1-2"},"PeriodicalIF":0.0,"publicationDate":"2016-04-03","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"84708209","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

The aim of this study was to label and visualize PIs labeled by the contrast agents based on Chemical Exchange Saturation Transfer (CEST) owing an advantage of contrast switching on/off and simultaneous visualization of differently treated cell population in one MR experiment. \u00001.2 Material and methods: Two paramagnetic chelates with europium Eu-DO3A-ae and ytterbium Yb-DO3A-ae were tested for labeling of PIs by pinocytosis and microporation. The toxicity, labeling efficacy and detection threshold were assessed for each agent. In vitro MR imaging of the labeled islets was performed on a 4.7 T scanner using a modified turbo spin-echo (RARE) sequence. \u00001.3 Results: The sufficient labeling efficacy was observed only by using high agent concentration compromising islet viability. The microporation procedure was not effective for islet labeling because of its invasive nature. The islets labeled by pinocytosis were visualized in a phantom by CEST imaging, however, after a long acquisition time at 4.7 T. \u00001.4 Conclusion: The low sensitivity of detection and impaired cellular viability of the pancreatic islets labeled by the novel CEST contrast agents represent a challenge for the further implementation of these probes as exogenous cellular labels.","PeriodicalId":89157,"journal":{"name":"International journal of molecular imaging","volume":"8 1","pages":"0-0"},"PeriodicalIF":0.0,"publicationDate":"2016-01-16","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"90681331","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Biliary complications are the most frequent complications after common liver surgeries. In this study, accuracy of hepatobiliary scintigraphy (HBS) and impact of hyperbilirubinemia were evaluated. Methods. Between November 2007 and February 2016, 131 patients underwent hepatobiliary scintigraphy after having liver surgery. 39 patients with 42 scans after LTX (n = 13) or hepatic resection (n = 26) were evaluated in the study; 27 were male, with mean age 60 years. The subjects underwent hepatobiliary scintigraphy with Tc-99m labeled Mebrofenin. The results were compared to ERCP as gold standard performed within one month after HBS. We calculated sensitivity, specificity, PPV, and NPV. We compared LTX patients to patients with other liver surgeries. Furthermore the influence of hyperbilirubinemia on HBS scans was evaluated. Results. HBS always provided the correct diagnosis in cases of bile leak in the liver-resected group (14/14). Overall diagnostic accuracy was 76% (19/25) in this group and 54% (7/13) in the LTX group. False negative (FN) diagnoses occurred more often among LTX patients (p = 0.011). Hyperbilirubinemia (>5 mg/dL) significantly influenced the excretion function of the liver, prolonging HBS's time-activity-curve (p = 0.001). Conclusions. Hepatobiliary scintigraphy is a reliable tool to detect biliary complications, but reduced accuracy must be considered after LTX. </p>","PeriodicalId":89157,"journal":{"name":"International journal of molecular imaging","volume":"2016 ","pages":"7857849"},"PeriodicalIF":0.0,"publicationDate":"2016-01-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"https://sci-hub-pdf.com/10.1155/2016/7857849","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"34337818","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"OA","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}
