All antisera had the same titre by immunofluorescence, 1/400 or 1/800, whether tested against the homologous or the heterologous virus. An antiserum prepared with the French strain neutralized the Hawaiian virus at a dilution of 1/800 both <em>in vitro</em> and <em>in vivo</em>. The Hawaiian virus was not neutralized to a significant extent by reference mouse ascitic fluids for 26 different vertebrate rhabdoviruses.</p></div><div><p>Des immunsérums ont été préparés, chez le lapin, à partir de deux souches de virus sigma d'origine géographique très différente (France et Iles Hawaii), ces deux souches ayant été purifiées après production sur cultures cellulaires. Les titres des immunsérums sont identiques (1/400 ou 1/800, selon le lapin considéré) qu'ils soient utilisés contre la souche homologue ou la souche hétérologue de virus sigma. L'immunsérum préparé contre la souche française neutralise la souche originaire de Hawaii jusqu'à la dilution 1/800 <em>in vivo</em> et <em>in vitro</em>. La souche hawaiienne n'est pas neutralisée d'une manière significative par des ascites immunes de référence obtenues chez la souris avec différents rhabdovirus de vertébrés.</p></div>","PeriodicalId":77667,"journal":{"name":"Annales de l'Institut Pasteur. Rodier","doi":"10.1016/S0769-2617(88)80080-7","DOIUrl":"10.1016/S0769-2617(88)80080-7","url":null,"abstract":"<div><p>In order to determine whether HTLV-I (a recently discovered retrovirus causing T-cell leukaemias and lymphomas, as well as tropical spastic paraparesis) was present in Djibouti in 1988, we investigated 576 subjects belonging to various groups at risk of acquiring HIV, a related retrovirus transmitted analogously to HTLV-I. Sera were screened by a commercial agglutination assay and confirmation of repeatedly reactive sera was performed by specific Western blot analysis. Four sera were strongly positive for HTLV-I by Western blotting, while one serum displayed equivocal reactivity. All positive sera came from young women who engaged in prostitution. This accounted for an HTLV-I seropositivity rate of 0.7% (95% CI, 0.03–1.4%) in the total study population and 1.2% (95% CI, 0.02–2.4%) in the population engaged in prostitution. We concluded that in Djibouti in June 1988, HTLV-I was present but that the prevalence of infection in high risk indivduals was very low.</p></div><div><p>Afin de savoir si HTLV-I était présent à Djibouti en 1988, nous avons examiné 576 sujets appartenant à divers groupes à risque pour un HIV, un rétrovirus apparenté à HTLV-I et qui est transmis de manière similaire. Les sérums ont été soumis à un test rapide de dépistage, et la confirmation des résultats positifs a été obtenue par la méthode du «Western blotå. Quatre sérums réagissent fortement pour HTLV-I au Western blot, tandis qu'un cinquième sérum donne une réaction non significative. Tous les sérums positifs sont ceux de jeunes femmes qui admettent être prostituées. Girard","doi":"10.1016/S0769-2617(88)80008-X","DOIUrl":"10.1016/S0769-2617(88)80008-X","url":null,"abstract":"<div><p>A chimaeric poliovirus carrying a type-2-specific neutralization epitope on a type 1 capsid was created by site-directed mutagenesis of the Mahoney strain of poliovirus type 1. An <em>Eco</em>RV and a <em>Hin</em>dIII restriction sites were first constructed in the cDNA of poliovirus type 1 at nucleotide positions 2756 and 2786, respectively, <em>i.e.</em> on either side of the sequence encoding neutralization epitope C3 (VP1 amino acids 93–103), which is part of neutralization site NImI. The cDNA sequence framed by the two sites was next taken out and replaced by custom-made oligonucleotides encoding the equivalent region of VP1 from the Lansing strain of polivirus type 2. The DNA from the plasmid carrying such a hybrid construct was transfected onto CV1 cells generating a chimaeric virus, v510. Neutralization of v510 with a panel of monoclonal antibodies showed that v510 has lost the poliovirus type 1 C3 epitope but acquired a new, poliovirus type-2-specific neutralization epitope. Preliminary results indicate that v510 also shows neurovirulence for mice, which is a specific trait of the Lansing strain of poliovirus type 2.</p></div><div><p>Pour créer un poliovirus chimère exprimant des épitopes de neutralisation du type 2 à la place de ceux d'une capside de type 1, on a créé, par mutagenèse dirigée à l'aide des oligonucléotides de synthèse voulus, un site <em>Eco</em>RV et un site <em>Hin</em>dIII en positions respectives 2.756 et 2.786 du cADN de la souche Mahoney de type 1. Ces deux sites encadrent la séquence de l'épitope C3 (acides aminés 93 à 103 de VP1) correspondant au site de neutralisation NImI de la capside du poliovirus. On a ensuite retiré la séquence ainsi encadrée, et on a inséré à sa place la région équivalente de la souche Lansing de type 2, sous form d'oligonucléotide de synthèse. On a ensuite transfecté des cellules CV1 avec le plasmide porteur du cADN hybride, générant ainsi un virus chimère, v510, dont la séroneutralisation par des anticorps monoclonaux montre qui'il a perdu l'épitope C3 du type 1 et acquis, à la place, un épitope de type 2.
