The probe was a <sup>32</sup>P-labelled cloned DNA fragment of CMV strain AD169. This probe did not hybridize to DNA from uninfected cells or other herpesviruses (herpes simplex virus, Epstein-Barr virus). Specific hybridization was obtained with unselected bronchoalveolar lavage specimens, but the sensitivity of the test (33%) was lower than that of culture (80%) and immunofluorescence (60%) assays which are routinely performed in our laboratory. The detection procedure was also carried out with pharynx biopsy specimens which had been kept frozen at −70°C. CMV DNA was detected in 14% of tumour specimens and only in 1.7% of control specimens (p<0.05). The indications of DNA hybridization for CMV diagnosis are discussed.</p></div><div><p>Une méthode simplifiée d'hybridation a été utilisée pour la détection du cytomégalovirus (CMV) dans les prélèvements humains. La sonde est un fragment cloné d'ADN issu de la souche AD169 du CMV et marqué par le <sup>32</sup>P.</p><p>Cette sonde n'a donné aucune hybridation avec l'ADN des cellules non infectées ni avec celui d'autres virus Herpes (Herpes simplex virus, virus Epstein-Barr). Une hybridation spécifique a été obtenue sur des lavages broncho-alvéolaires n'ayant fait l'objet d'aucune sélection mais la sensibilité du test (33%) est inférieure à celle de la culture virale (80%) et à celle de l'immunofluorescence (60%) qui sont pratiquées en routine dans notre laboratoire. La même technique a été appliquée à des biopsies pharyngées qui avaient été conservées congelées à −70°C. L'ADN du CMV a été détecté dans 14% des prélèvements de tumeurs et seulement dans 1,7% des prélèvements témoins (p<0.05). Bouloy","doi":"10.1016/S0769-2617(88)80006-6","DOIUrl":"10.1016/S0769-2617(88)80006-6","url":null,"abstract":"<div><p>The cDNA coding for the yellow fever virus (YFV) envelope protein (E) was inserted into an SV40 vector under the control of the late promoter in place of the VP1 gene. The recombinant virus expressed a 52-Kd polypeptide which was detected by immunoprecipitation with a monoclonal antibody raised against the E protein. Surprisingly, this protein was visualized in the nuclear of the infected cells. The possible presence of a sequence involved in nuclear migration of the E protein and naturally ignored during virus infection is discussed.</p><p>The sequence coding for the mature E protein is directly preceded in the YFV genome by a sequence of 45 nucleotides coding for a 15-amino-acid long hydrophobic oligopeptide. The sequence of this oligopeptide was inserted into the SV40 recombinant virus between the ATG codon and the first codon for the E protein. The E protein expressed by this new SV40 recombinant virus was found to be localized in the cytoplasm of the infected cells in association with intracellular membranes. These results strongly suggest that the 15-amino-acid long hydrophobic region naturally plays a role as a signal peptide for the E protein.</p></div><div><p>Expression de la protéine d'enveloppe du virus de la fièvre jaune à l'aide de virus recombinants SV40 L'ADN complémentaire de la région du génome viral codant pour la protéine d'enveloppe (protéine E) du virus de la fièvre jaune a été inséré dans un vecteur SV40, à la place du gène VP1 et sous la dépendance du promoteur tardif.</p><p>Le virus recombinant obtenu exprime un polypeptide de 52 Kd que l'on peut détecter par immunoprécipitation avec un anticorps monoclonal anti-E. De façon très surprenante, cette protéine est retrouvée dans le noyau de la cellule infectée. L'existence possible d'une séquence de migration nucléaire dans la protéine E, existence normalement ignorée lors de l'infection naturelle, est discutée.</p><p>La région codant pour la protéine E mature est directement précédée, dans le génome du virus de la fièvre jaune, par une séquence de 45 nucléotides codant pour un oligopeptide hydrophobe de 15 acides aminés. Dans le virus SV40 recombinant, nous avons inséré la séquence codant pour cet oligopeptide, entre l'ATG initiateur et le premier codon de la protéine E. La protéine E exprimée par ce nouveau virus SV40 recombinant se trouve maintenant dans le cytoplasme, associèe à des membranes intracellularires.
