UV and NMR spectra revealed the presence of hydrogen bonds in these complexes. The dissolution rates of various PVP-gossypol complexes in water at 37 degrees C were determined. The results showed that the presence of PVP in the complex enhanced the dissolution rate of gossypol in water greatly. In addition, the in vitro spermicidal activity on human semen, antifertility effect on male rats, and the toxic reaction of male mice by taking large dosage were preliminarily studied. The experimental results indicated that the PVP-gossypol complexes not only possessed the similar antifertility effect and acute toxicity to male mice as gossypol-acetic acid, but also reduced the irreversibility of the fertility as well. The sorbent contains antidigoxin antibodies covalently bound to polyacrolein microspheres, 0.2 mu diam. Thousands of microspheres are matrix-encapsulated in crosslinked agarose to form beads of 500 to 800 mu diam. Digoxin intoxicated dogs showed heart block, ventricular and atrial tachycardia and extended runs of PVCs. During hemoperfusion the abnormal ECG abated; at 2 h of hemoperfusion the ECG tracings returned to normal. Up to 25% of the total digoxin burden was removed. The numbers of the blood cells and the content of a battery of relevant soluble components of the blood remained constant. Dogs were alive and well after the hemoperfusion. Non hemoperfused dogs, which received antiarrythmic agents, did not survive the intoxication. It involves skeletization of the vein and its collaterals by longitudinal blind and blunt dissection, thus separating the vein from its surroundings. This is followed by elevation and cutting of the subdermal tissue with an electrosurgical pencil. The device consisted of Ti-6A1-4V alloy acetabular and femoral components connected with a Bion component. At necropsy the Bion component was found to have fractured. The tissue surrounding the Bion was granulomatous with numerous blackened regions. {"title":"Thrombogenicity and the interaction of proteins, platelets and white cells.","authors":"J Long, S DeSantis, E Shors, M Uszler, C Wuest, S Klein, R White","doi":"10.3109/10731198309118795","DOIUrl":"https://doi.org/10.3109/10731198309118795","url":null,"abstract":"<p><p>Radioactive labeled protein and platelet studies suggest that an ideal vascular surface absorbs albumin and prevents platelet adherence and aggregation. This study examines the interaction of serum and plasma proteins, white cells, and platelets in the thrombotic response to small internal diameter vascular prostheses. Thirty-seven replamineform silicone rubber vascular prostheses, 4 mm internal diameter and 6 cm length, had 5% aspirin and 5% theophylline by weight dispersed in a luminal coating (D); the remaining 37 control (C) prostheses had an identical silicone rubber coating without drugs. Scanning electron and darkfield microscopy confirmed similar surface topography in C and D grafts. One C and one D graft were studied in parallel using a modified canine arteriovenous fistula. Consistently, D prosthesis had increased bleeding through the graft wall. The 7 serum and 8 plasma experiments demonstrated greater accumulation on the D prostheses, the greatest deposition occurring within the first 30 minutes. In 8 platelet experiments, D grafts accumulated more radioactivity at both the 5 and 30 minute intervals. Six mononuclear and 8 PMN experiments had less accumulation on the D grafts at 15 minutes and no consistent difference at 60 minutes. Elution of C and D Grafts demonstrated significant drug levels in only the D grafts at the beginning of the experiments and after 15 minutes of exposure of blood. The calves were chronically instrumented to measure (or derive) heart rate, stroke volume, cardiac output, iliac, superior mesenteric and renal flows and resistances, mean aortic, right atrial, pulmonary artery, and left atrial pressures, the systemic and pulmonary pressure gradients, and total systemic and total pulmonary resistances. The calves were also instrumented to produce reversible partial constriction of the ascending aorta and for cooling of the cervical vagus nerves. The hemodynamic responses to increased afterload were characterized before and during bilateral cervical vagus nerve cooling to 6-7 degrees C. Aortic constriction causes stroke volume, cardiac output and aortic pulse pressure to decrease. Left atrial pressure and total peripheral resistance increase. Mean aortic pressure is constant during aortic constriction alone, despite a continued decrease in pulse pressure, due to a balance between sinoaortic reflexes which attempt to increase arterial pressure in response to the decreased pulse pressure and cardiopulmonary reflexes which attempt to decrease arterial pressure in response to the increased left atrial and cardiopulmonary pressures. Vagal cooling removes cardiopulmonary reflex modulation of the sinoaortic reflexes.
