The same specificity was observed in more sensitive passive haemagglutination tests. The activity was confined to the monoclonal IgM present in the sera, and all of the monoclonal IgM in each serum had the characteristic activity. (2) The monoclonal IgM proteins induced typical capsular swelling reactions of similar specificity with Klebsiella bacteria. (3) In both precipitation, passive haemagglutination and capsular swelling reactions, the IgM Ro reacted both with type 12 and type 13 polysaccharides. Absorption experiments in situ in crossed immunoelectro-phoresis indicated that the same macroglobulin molecules reacted both with type 12 and type 13 polysaccharides. The fragments came from unprimed, NIP-Ficoll (polymer of sucrose and epichlorhydrin) or NIP-CG (chicken globulin) primed mice. Unprimed spleen fragments from C57BL/6 mice gave a good anti-NIP response to NIP-Ficoll, whereas CBA fragments did not. Priming with NIP-Ficoll made CBA fragments responsive and enhanced slightly the response of C57BL/6 fragments when stimulated with the same antigen. This memory effect could be seen only after a small priming dose. Priming the mice with NIP-Ficoll made their spleen fragments responsive to a protein conjugate of NIP (NIP-CG), but this effect was seen only after priming with a high dose. The antibody class distribution and the kinetics of the appearance of different immunoglobulin classes were similar in the primary and secondary responses in vitro. The peak responses of IgM, IgA and IgG were reached on day eight and the relative amount of IgG was the same in the primary and in the secondary responses. Spleen fragments derived from NIP-CG primed mice produced more IgG anti-NIP antibodies than fragments derived from untreated mice when immunized in vitro with NIP-Ficoll. In Crohn's disease, the total number of lymphocytes was found to be reduced and an almost equal reduction in all the lymphocyte subpopulations studied was found. In ulcerative colitis neither the number of T-lymphocytes nor of B-lymphocytes were found to differ significantly from normal values and the same was true of all B-lymphocyte subpopulations. However, the number of lymphocytes carrying neither surface immunoglobulin nor sheep red blood cell receptors was found to be significantly increased. In the group of prospective donors, the IEOP technique detected HBsAg at a frequency of 0.5 per cent, while a more sensitive solid-phase radioimmunoassay (RIA) detected a further 0.1 per cent. The RIA technique used in this investigation showed an incidence of false positives of 0.5 per cent. Most of the falsely positive sera probably contained antibodies against guinea pig globulin thus interfering with the test. The incidence of suspected transfusion hepatitis decreased after the screening for HBsAg among the blood donors was introduced. During a three-week period their weight increased from about 8 to about 10 g, while mice on normal feeding increased to about 16 g. This malnutrition resulted in a 10-fold increase of the susceptibility to intraperitoneally injected E. coli 06:K2a,2c:H1 bacteria compared to conventionally fed controls. Immunization as well as transfer of fresh immune serum to malnourished or normally-fed mice resulted in an increase in resistance, 6 to 10 fold that of non-immunized controls, while transfer of normal serum did not have a great impact on the resistance. Antibody titers to El coli 06 antigen in mice on poor and normal feeding were similar, as were there non-specific cellular responsiveness measured as lymphoblast transformation by ConA, PHA and LPS. The effect of the malnutrition seemed to be an impairment of the clearance of live E. coli 06 bacteria 1 h after a sub-lethal infection compared with that of normally-fed controls. None of the beta-lactam antibiotics studied caused significant alterations of granulocyte migratory response when incubated with the cells at concentrations of up to 128 microgram/ml. The two aminoglycoside preparations, and the two tetracycline preparations caused significant depressions of the migration response. Production of chemotactic factors was stimulated from growing cultures of E. coli by sub-minimal inhibitory concentrations of beta-lactam antibiotics and from Ps. aeruginosa by the cephalosporin derivatives only. Ten rabbits were immunized and bled every fifth week. Eight pools of St-Ab were produced during nearly four years. All the St-Ab pools were mutually compared by means of quantitative immuno-electrophoretic methods. All St-Ab pools were found to present St-Ag/St-Ab immunoprecipitate patterns which, qualitatively and quantitatively, were similar. None of the obtained immunoprecipitates between St-Ag and the pooled antisera disappeared during the procedures of purification of IgG and IgA. Quantitative and qualitative comparison of the polyvalent St-Ab pools (\"titre\") was made by counting the total number of immunoprecipitates obtained by St-Ag, followed by their identification, and by calculating the mean area of 12 selected and representative immunoprecipitate. The abilities of the rabbits to produce antibodies were found to vary individually. None of the rabbits produced precipitating antibodies against all the 64 known P. aeruginosa antigens in St-Ag. On the average, each of the rabbits lacked precipitating antibodies against 5 of the antigens. Analysis of the data showed that extraction of the cells with detergents provided yields of the serotype protein substantially exceeding those obtained by simple salt extraction of the bacteria. Routinely, more than 2 mg of end product per g of cell mass (wet weight) may be recovered by the present method. By gel chromatographic analysis, the serotype determinant protein was shown to interact with the capsular polysaccharides derived from Group A or C Neisseria meningitidis microorganisms, forming high molecular weight complexes. This interaction markedly enhanced the solubility of the serotype determinant protein. Combined vaccines of the type-specific protein with the group- specific polysaccharides were evaluated for their immunogenic potential in the subcutaneous steel spring implant model. In guinea pigs, amounts corresponding to 10 micrograms completely prevented infection upon challenge with homologous organisms four weeks after immunization. Partial protection was observed with immunizing doses corresponding to 2 micrograms or 0.4 micrograms/animal, respectively. Compared to lyophilized preparations, vaccines adsorbed to a mineral carrier were slightly less effective in inducing protection, whereas inclusion of Bordetella pertussis as a component of the vaccine stimulated the immune response.","PeriodicalId":75411,"journal":{"name":"Acta pathologica et microbiologica Scandinavica.
