P. aeruginosa strains isolated from CF patients were more sensitive than strains from non-CF patients to the bactericidal activity of normal human serum. The bactericidal activity was heatlabile. As regards the sensitivity to normal human serum, mucoid and non-mucoid variants were not found to differ. Strains originating from chronically infected CF patients with many precipitins against these bacteria did not differ with respect to serum sensitivity from strains originating from intermittently colonized CF patients without P. aeruginosa precipitins. Compared with normal sera, CF sera showed similar or higher bactericidal activity against a panel of P. aeruginosa strains. In this respect, any difference between CF sera with precipitins and CF sera without precipitins against P. aeurginosa was not found. Sera from three CF patients chronically infected with P. aeruginosa, and with many precipitins against these bacteria, showed a selective inability in bactericidal activity against the patients' own P. aeruginosa isolate possibly reflecting the presence of \"bactericidal blocking\" antibodies.","PeriodicalId":75411,"journal":{"name":"Acta pathologica et microbiologica Scandinavica. In the present work it is shown that some 85-90% of the haemolytic plaque-forming cells (PFC) survived the freeze-thaw process. Irradiated rats were restored with fresh and frozen-thawed cells and immunized against sheep red blood cells (SRBC). Evidence is presented that a restrictive control of the PFC response by suppressor cells present in the spleen cell suspension is lost during the freeze-thaw process, giving a higher number of PFC/spleen in recipients of frozen-thawed mixed spleen and lymph node cells than in rats receiving the corresponding fresh preparations.","PeriodicalId":75411,"journal":{"name":"Acta pathologica et microbiologica Scandinavica. Their size as well as their content of phase-lucent and phase-dense granules increased. The cell content of the lysosomal enzyme acid phosphatase was increased. In contrast, macrophages from the same source and cultured in a medium containing 50 per cent foetal calf serum (FCS) were not activated in this way. NBCS-activated macrophages had a markedly enhanced capacity to phagocytose Toxoplasma gondii and Listeria monocytogenes, as compared with macrophages cultured in FCS. The effector cells could be divided into at least two subpopulations. 1) host-derived macrophage-like cells and 2) thymocyte-derived donor cell T lymphocytes. These two subpopulations could be separated and identified by means of commonly used immunofractionation techniques. Purified suspensions of these all produced anti-B allo-antibodies which could both haemagglutinate red blood cells and strongly inhibit the GVH reactivity of blood lymphocytes of the original donor type. Absorption studies of the sera using cells of the same four sources showed that they could all completely remove the GVH inhibitory antibodies. A fact of particular importance is that B-haplotype determined antigens which are common to erythrocytes and lymphocytes seem to include the gene products of the allo-aggression locus, as earlier defined (Simonsen 1975).","PeriodicalId":75411,"journal":{"name":"Acta pathologica et microbiologica Scandinavica. Anti-human IgM immunoglobulins were bound on the wells of polyvinylchloride microtiter plates. Serum specimens were incubated in the anti-human IgM coated wells and bound IgM antibodies were then assayed for antigen specificity by subsequent incubations with HAV antigen and 125I-labelled human anti-HAV IgG. The test showed a high sensitivity and specificity for anti-HAV IgM antibodies. No false-positive reactions were observed either in the sera from patients with hepatobiliary disorders other than HAV infection or in the sera containing both rheumatoid factor and anti-HAV IgG antibodies. In acute HAV infections specific IgM antibodies were present already in the first specimens taken within a few days after the onset of jaundice. The persistence of the IgM antibodies was from 4 to 6 months. IgM antibody titers up to 1,000,000 were observed in the acute phase of HAV infection. In routine diagnostic work the titration of the sea was not necessary, since a reliable qualitative result was obtained by testing the sera in a single dilution of 1:100. A similar \"reverse\" immuno-assay principle may be adaptable for the diagnostic determination of IgM antibodies to different viral and microbial antigens.","PeriodicalId":75411,"journal":{"name":"Acta pathologica et microbiologica Scandinavica. Animals immunized and treated with CVF had an infection resistance similar to untreated controls but considerably less than mice immunized only. The CVF treatment did not affect the antibody formation after immunization. Untreated or immunized AKR mice deficient in C5 did not show decreased resistance to the infection. Exposure of NMRI mice to preformed antigen-antibody complexes unrelated to the infection did not increase the susceptibility of the animals to infection, neither did it impair their C3 levels. Two out of five freshly frozen (-70 degrees C) normal rabbit sera gave significant, heat-labile protection of mice to the E. coli infection without containing any antibodies detectable with the ELISA. This protective capacity was not abolished with zymosan treatment affecting the levels of complement factors and could be \"natural antibodies\" stimulated from the environment. In testing whether similar protective factors as those found in normal rabbit serum could be raised in CBA mice, such animals were orally given killed E. coli 06 bacteria. St-Ag did not undergo proteolytic degradation by plasmin which is present in the corresponding polyvalent rabbit antiserum (St-Ab) against St-Ag. Storage at -30 degrees C for up to 4 years caused no qualitative changes in St-Ag and, after 1 year, only minor quantitative changes could be revealed. Repeated thawing and freezing caused no changes in St-Ag whereas storage at 4 degrees C, or at room temperature, caused gradual degradation of most of the antigens of St-Ag, beginning after 1 week. Heating up to 40 degrees C for 2 min did not change St-Ag, but heating for longer periods of time or at higher temperatures gradually destroyed most of the antigens of St-Ag. None of the beta-lactam antibiotics studied caused significant alterations of granulocyte migratory response when incubated with the cells at concentrations of up to 128 microgram/ml. The two aminoglycoside preparations, and the two tetracycline preparations caused significant depressions of the migration response. Production of chemotactic factors was stimulated from growing cultures of E. coli by sub-minimal inhibitory concentrations of beta-lactam antibiotics and from Ps. aeruginosa by the cephalosporin derivatives only.
