In both cases of hepatoblastoma the serum AFP were determined by rocket immunoelectrophoresis. {"title":"Diabetic cardiopathy. Quantitative histological studies of the heart from young juvenile diabetics.","authors":"T. The groups were comparable with respect to sex-ratio, age, heart weight and blood pressure. Findings in the hearts were as follows: In arterioles in which the luminal diameter ranged between 15-50 mu, a strongly PAS-positive stained vessel wall occurred at a higher frequency among diabetics than among non-diabetics (75 per cent respectively 33 per cent). The PAS-positive structures, however, occupied the same area of the vessel wall in the two groups. In arterioles from diabetics, the number of cells in tunica media was increased as compared with that in non-diabetics (2 p less than 0.01). The amount of perivascular connective tissue was also increased in the diabetics (2 p less than 0.01). There was no indication of an endothelial cell proliferation in the PAS-positive stained vessels from diabetics and non-diabetics. The wall of the capillaries was not thickened and it was not more PAS-positive in the hearts from diabetics than in those from non-diabetics. Moreover, the number of capillaries per square millimeter of heart muscle was the same in the two groups of hearts. In the present light microscopical study, the diabetic micro-angiography of the heart was demonstrable in the arterioles. The experimental mice were examined at varying intervals after the FUDR injection. At term, the young exhibited the malformations of the limbs and skull so well-known after administration of FUDR. In addition, there was an increased number of resorptions. 24 hours after the injection on the 9th—12th day of gestation, vascular dilatations and fresh haemorrhages were found at the sites of predilection of malformations. Foetuses removed 2 or more days after the injections showed characteristic, saccular haematomas in the limbs and tail as well as flat, widespread haematomas in the skull. At increasing intervals after the injections, the haematomas decreased in number and were rarely present in foetuses at term. As the number of haematomas decreased, the number of malformations increased. Daily laparotomies, from the 12th to the 18th day, on 5 mice treated on the 11th day with FUDR revealed extremity haematomas which disappeared on the 13th—18th day. Electron microscopic pilot studies of 2 foetal limbs showed, 24 hours after the injection of FUDR, red blood cells in an extra-vascular situation. The control mice had 354 normal foetuses and 8 per cent spontaneous resorptions. This study, which was a screening experiment, showed that FUDR induces vascular injury with haemorrhages and formation of haematomas, resulting in congenital malformations. The islets of starved controls possessed larger B-cell mitochondria than those of fed ones. The earliest B-cell changes in the alloxan-treated animals were localized to the mitochondria which showed swelling, and disruption of inner and occasionally outer membranes. Later, many mitochondria were disintegrated, and the endoplasmic reticulum and Golgi complex disorganized. The secretory granules were preserved, although sometimes with atypical configuration, in degenerating but non-necrotic B-cells, suggesting that insulin stored in granules is not released until the cells are necrotic. Finally, frank necrosis was seen in some B-cells, whereas others were unaffected. The Ca2+-precipitation studied by pyroantimonate technique and x-ray analysis differed in the B-cells of the alloxan-treated animals from that in the controls; the former animals exhibited no or only sparse precipitation in mitochondria and secretory granules, but a rich precipitation in the cytoplasmic ground substance, whereas the precipitation in the controls mainly was localized to mitochondria and secretory granules. {"title":"Cell-associated immunoglobulin in human non-Hodgkin lymphomas. Jørgensen, Herman Høst","doi":"10.1111/J.1699-0463.1981.TB00194.X","DOIUrl":"https://doi.org/10.1111/J.1699-0463.1981.TB00194.X","url":null,"abstract":"Eighty-three non-Hodgkin lymphomas classified according to the Kiel classification have been studied with regard to surface immunoglobulin (sIg) on cells in suspension and cytoplasmic immunoglobulin (cIg) by the peroxidase anti-peroxidase method (PAP) on formaline-fixed tissue sections. Fifty-six out of 66 examined (i.e. 85%) revealed a monoclonal staining pattern for sIg, whereas 37/70 (53%) gave a monoclonal staining pattern for cIg by PAP. The methods combined gave a monoclonal staining pattern in 73/83, i.e. in 88%, of the biopsies tested. The discrepancies between the two methods were largest in centroblastic/centrocytic and lymphocytic lymphomas. With regard to the light chain staining patterns, complete agreement between the two methods was obtained in the 20 cases that allowed such analysis to be made. This suggests that the specificity of PAP, as carried out in this study with reagents purified by immunoabsorbent techniques, is satisfactory. On a basis of heavy chain isotypes centroblastic/centrocytic, lymphoplasmacytoid, and immunoblastic lymphomas could be divided into distinct immunological subgroups. In four biopsies the sIg heavy chains were mu + delta, whereas mu + gamma chains were detected by PAP. This finding may be relevant to the mu leads to gamma switch known to occur during normal B-cell differentiation. In all five patients slight vasculitis and a few interstitial eosinophils were found in the pulmonary tissue 1-2 weeks after the withdrawal of nitrofurantion therapy. Alveolitis, fibrinous alveolar exudate and perivascular granulomas were also seen in some specimens. The patients with acute reactions had marked blood eosinophilia. In all patients, pulmonary diffusing capacity (DLCO) was below normal, but ventilatory function was not appreciably affected. Withdrawal of nitrofurantion resulted in complete or partial improvement in 4-8 weeks. These malignant soft tissue tumours are characterized by a mucoid and nodular appearance, a coarse plexiform capillary pattern, and they are mostly seen subcutaneously (26 out of 30) in the extremities (24 out of 30) and trunk (4 out of 30) elderly people. Histochemical studies, comprising staining with Alcian blue and toluidine blue at different pH's with and without preceding digestion with testicular hyaluronidase and with the Scott technique, indicated the presence of hyaluronic acid but not sulphated glycosaminoglycans as chondroitinsulphates. Myxofibrosarcoma is believed to belong to the general category of fibroblastic and histiocytic malignant soft tissue tumours. The median diameter of the tumours was 7 cm. They were divided into 4 grades according to cellularity, cell atypia and mitotic activity. The grade III and IV tumours showed pronounced atypia, often with the bi- and multinucleated giant tumour cells and occasionally with giant cells of Touton's type, suggesting a relationship to malignant fibroxanthoma. All of the patients were treated surgically and one received also pre- and post-operative irradiation. None of the 2 grade I myxofibrosarcomas recurred, while 2 out of 7 grade II tumours, 6 out of 10 grade III tumours, and 7 out of 11 grade IV tumours recurred once and up to 9 times. Metastasis appeared in 7 out of 30 patients; grade I tumours were not seen in any of these cases. By the time of follow-up after intervals ranging from 1 month up to 27 years, 14 patients had died; 6 of these had died post-operatively or of intercurrent disease.
