Our work purpose is to evaluate the level of LNC (TUG1) in SLE patients. This research includes 20 control and 70 SLE subjects. The serum level of LNC (TUG1) was measured by polymerase chain reaction. Down-expression of LNC (TUG1) was observed in SLE patients more than in control. This study aimed to evaluate the genetic association of single nucleotide polymorphism rs3807598 (C:G) of HOTTIP among rheumatoid arthritis patients. Method: The study included 120 subjects from 2 groups: 60 RA patients and 60 healthy controls. Real-time PCR with TaqMan allelic discrimination assay was used to perform the genotyping. The odds ratios (ORs) models and 95% confidence interval (CIs) were used to test the associations. Results: The results indicated that lncRNA HOTTIP SNP rs3807598 (C:G) exhibited a statistically nonsignificant association with the risk of RA. Conclusion : Up-to-date, the role of HOTTIP SNPs in RA risk is still unknown, It is important to consider how epigenetic modifications including non-coding RNAs polymorphism and expression affect the RA risk and how well it responds to treatment. {"title":"The Incidence and Molecular Characterization of Banana Bunchy Top Virus (BBTV) Infecting Banana (Musa spp.) in Ismailia governorate, Egypt","authors":"H. Amin, Azza G. Banana bunchy top disease, caused by the banana bunchy top virus (BBTV), is one of the most damaging infections on banana plantations, resulting in significant crop losses. The current study was conducted to characterize the incidence of banana bunchy top disease (BBTD) and to study the molecular characteristics of BBTV from banana plantations in Ismailia. This study was conducted during the period 2019-2021 in three locations (Abu-Suwayr, El Tal-El kabir and Fayed) in Ismailia Governorate. The occurrence of BBTV was initially studied using visual inspection of external virus symptoms. TAS-ELISA, PCR and nucleic acid hybridization were used to detect the virus. Further molecular characterization of BBTV was performed based on Rolling Circle Amplification (RCA) and nucleotide sequencing. BBTD incidences ranging from 3.33 to 5.14% were observed in two localities Abu-Suwayr and El Tal-El kabir. While a relatively low incidence of 1.48% was observed in Fayed. Our results showed that the BBTD incidence is directly related to the percentage of winged aphids inside the aphid colonies regardless the percentage of the aphid's occurrence. The PCR product (973 bp) of BBTV-component 3 (DNA-S) was used for the creation of a non-radioactive Dig labeled-DNA probe for further use for BBTV detection using dot blot and Southern blot hybridization. The sequence comparison analysis of DNA-S component showed that the present Ismailia isolate (BBTV-3F) shared nucleotide identity 98.4%- 91.9 % with three isolates from Qalubya Egypt and shared 97.9% nt identity with east African isolates (Rwanda and Malawi). The phylogenetic analysis revealed that the DNA-component 3 (DNA-S) of the BBTV-3F isolate from Ismailia belongs to the South Pacific group (SPG) and is markedly separated from those of the Asian group (AG). There has been a clear interest in assessing the relationship between antibiotic resistance phenotype and biofilm production. The aim of this paper was to present additional experimental results on this topic and to test the ability of Salmonella typhi isolates to biofilm formation using in vitro methods in the context of their antibiotic resistance. In the result of the study, 28 isolates of S. typhi were included. All 28 clinical specimens 100% showed strong biofilm formation and all clinical specimens showed the presence of pml gene by clear bands in gel electrophoresis. The results of the antibiotic sensitivity test of S. typhi isolates by disk diffusion method against 18 types of commonly used antibiotics showed a large variation in their rates of antibiotic resistance, as the highest percentage of resistance to the antibiotic to S . typhi resistance to the most common antibiotics used in treatment. Aim: To assess the effect of cigarette smoking on nerve conduction velocity of median and ulnar nerve by nerve conduction study. Methodology: 50 smokers and 50 non-smokers were recruited and nerve conduction velocity of the median and ulnar nerve were studied in both groups. Result: The sensory nerve conduction velocity among smokers was significantly reduced when compared to non-smokers (p<0.05). Discussion: The chemical oxidants that are released from cigarette smoke cause neural ischemia and also enhance oxidative stress. The sensory nerve is a thin nerve and it is more prone to damage by the chemicals released from cigarette smoke. Therefore, it may be a useful biomarker for diagnosing disease severity in COVID-19 patients. lipocalin-2 was initially identified as a secreted protein from human neutrophils. Alveolar type II cells that have been damaged primarily express this substance. To verify lipocalin-2's potential as a diagnosing biomarker for COVID-19 patients, Lipocalin-2 levels in the blood were examined in this pilot investigation. To examine the relationship between serum lipocalin-2 levels and the severity of COVID-19 infection to see if this protein may be utilized as a disease indicator. This study was done in a case-control study with One hundred and twenty patients (79 males, 41 females) with COVID-19 who participated in the research. The COVID-19 patients were divided into three groups based on the severity of the illness: critical disease (n = 30), severe disease (n = 30), and mild/moderate disease (n = 60), with (n = 60) healthy volunteers serving as the control group (35 males, 25 females). Between January 2022 and May 2022, the patients were obtained from Al-Amal hospitals and the AL-Shefaa centre in ALNajaf City, Iraq., All of the patients' fundamental clinical and demographic data were collected along with blood samples. Enzyme-linked immunosorbent tests were used to measure the blood's level of LCN2 (ELISA). The levels of total cholesterol, triglycerides, and High-Density lipoprotein were assessed using colorimetric methods. Ichroma was tested for serum ferritin, D-dimer, and CBC by Swelab. ran a statistical study to see if they were related to the severity of the disease. Higher lipocalin-2 levels were observed in the patient group, particularly in cases of mild/moderate (1.32±0.30) (P. 0.001), severe(2.16±0.42) (P. 0.001), and critical(4.71±1.01) (P. 0.001) comparing cases to healthy controls (0.86±0.51) respectively, groupings. (SPO2 %, Hb, TC, HDL, LDL, and lymphocyte) levels were found to significant negative correlation with one another in the COVID-19 patient group, with p-values=0.001 for each of these relationships. Moreover, a significant positive correlation between (TG, VLDL-C, WBCs, neutrophil, platelet, N/L ratio, D-dimer, Ferritin, and CRP, p.value=0.001 for each one of them) levels with lipocalin-2 in the COVID-19 patients group. a cut-off value of 1.215 (ng/mL) for lipocalin-2 predicted severe COVID-19 with a sensitivity of 81.7 % and a specificity of 80.2 % (AUC: 0.9, 95%CI 0.852-0.949;p<0.0001).
