Pancreatic B-cells express the MTNR1B gene, which expresses the receptor 1B of melatonin. Multiple large-scale genome-wide association studies and meta-analyses revealed a strong connection between the rs10830963 of the MTNR1B gene with fasting glucose and T2DM in many ethnic groups. This study aims to detect the link between rs10830963 and early-onset type 2 diabetes (EOD) in the Egyptian population. The rs10830963 was identified using polymerase chain reaction (PCR) followed by DNA sequencing technology on 12 T2DM patients and 18 healthy controls group with an age range of 30 to 39 years. Fasting blood sugar (FBS), glycated hemoglobin (HbA1c), serum creatinine (CREAT), triacylglycerol (TG), total cholesterol (TC), high-density lipoprotein cholesterol (HDL-C), low-density lipoprotein cholesterol (LDL-C), and very low-density lipoprotein cholesterol (VLDL-C) were determined for both healthy and diabetic groups. Four T2DM patients (one male and three females) in addition to four healthy individuals (one male and three females) were found to contain the risk G allele when compared to the NCBI reference sequence using the T-Coffee online tool for multiple sequence alignment. Furthermore, the RNA secondary structure for reference and sample sequences with the rs10830963 was predicted in terms of minimum free energy (MFE) using the Vienna online tool, finding that there is a slight difference in MFE between the compared sequences, which may explain the upregulation of mRNA of the MTNR1B gene in presence of rs10830963. The Middle-East respiratory syndrome (MERS) and severe acute respiratory syndrome (SARS) outbreaks in 2003 and more recently have demonstrated how lethal CoVs can be when they infect humans across the species barrier. The novel severe acute respiratory syndrome coronavirus 2 has threatened the world in many ways (SARS-CoV-2). Zonulin is a member of a protein family whose first member, pre-haptoglobin 2 (HP2), was discovered nearly ten years ago. (Rittirsch D 2013). Materials and Methods: A total of 120 Covid-19 patients' serum samples were collected and an apparently healthy group (n=60) with an age range (of 35-75) years, was admitted from ALAmal Hospital. Zonulin levels were measured by enzyme-linked immunosorbent assay (ELISA) (kit. Metabolic parameters were measured by enzymatic spectrophotometer methods. The correlation coefficients between serum Zonulin levels and age, BMI, Elements and electrolytes were also evaluated. Results: Serum Zonulin, CRP, D-dimer and ferritin levels were significantly higher in Patients with COVID-19 (324.4±12.46) vs in control (79.69±11.77), (42.67±1.84) vs in control (3.36±0.25), (4188.21±198.73) vs in control (289.43±251) and (738±20.09) vs in control (130.66±9.2) (P <0.001). The correlation of Zonulin levels in COVID-19 patients was significantly positive with age, CRP, D-dimer and ferritin levels but negative with Iron, Ca and Na levels. The serum of Zonulin levels in moderate COVID-19 patients significantly high compared with the critical and severe patients group. Conclusions: Serum Zonulin levels increased in COVID-19 patients, especially in severe cases. Therefore, Zonulin levels demonstrate a prognostic value for predicting the severity of COVID-19. Continuous Zonulin results throughout the study period revealed that the severe group's values were higher than those of the non-severe group. Underlying molecular mechanism of HCV-induced-HCC by temporal expression mRNA profiling of mock and HCV-infected Huh7.5.1dif cells was unraveled. A catalogue of bio-markers of HCC was analyzed to interpret the clinical relevance of the bio-markers. The percentage of over-expression and co-occurrence for targeted bio-markers within The Cancer Genomic Atlas (TCGA) were mapped using cBioPortal for cancer genomics. Spotting of differentially expressed genes (DEGs) and related temporal pathways enrichment analysis was accomplished employing MATLAB functions (e.g. mattest). Module analysis of the designed co-expression network using Cytoscape software was carried out. The overall expression of targeted biomarkers was recorded in 60% of the cases. Significantly overexpressed biomarkers recognized were CLK2, E2F5, CDK5, E2F3, MCM3, PCNA and CDK4. CCNB2, CLK2, CDK4, CDC7, E2F3, PCNA, and MCM3 predominantly controlled the co-expression of the listed bio-markers. After the initial phase of infection with a detrimental expression changing pattern, stability in expression followed by consistency in the level of expression of a set of genes was observed.
