Forty patients with NAFLD, 38 patients with NAFLD combined with T2DM, and 30 patients with normal liver tissue were selected. They were set as Group A, Group B and Group C respectively. The expression level of glucose-regulated protein 94 (GRP94) and biochemical indicators such as alanine aminotransferase (ALT), free fatty acids (FFA), triglycerides (TG), gamma-glutamyl transferase (GGT), aspartate aminotransferase (AST), fasting blood glucose (FBG), and fasting insulin (FINS) were measured. Also, the calculation of the homeostatic model assessment for insulin resistance (HOMA-IR) index was performed. The reactive hyperemia index (RHI) was detected to evaluate the vascular endothelial function of patients. The comparison between groups and multi-factor analysis of the influencing factors of RHI was conducted. Compared with Group C, the expressions in Group A and Group B were distinctly enhanced (P <0.05). Also, the expression of GRP94 protein in Group B was distinctly higher than that in Group A (P <0.05). The average optical density values of Groups A, B, and C were 0.327 ± 0.007, 0.350 ± 0.009, and 0.299 ± 0.006, respectively. A comparison between the three groups was performed. The differences had statistical significance (P <0.05). The differences in the TG, ALT, AST, GGT, FINS and HOMA-IR between Group A and Group B had statistical significance (P <0.05). The RHI values of Groups A, B, and C were 1.59 ± 0.23, 1.79 ± 0.32, and 2.05 ± 0.47, respectively. A comparison between the three groups was performed. The differences had statistical significance (P <0.05). FFA, ALT and FBG in patients with NAFLD are risk factors for endothelial dysfunction (P <0.05). The liver damage caused by NAFLD may be related to the expression of GRP94. With the rapid development of medical technology, targeted therapy is a common choice in treatment plans due to its low toxicity and high pertinence (3). EGFR can reduce the possibility of autophagy by activating downstream factors such as P13K and MAPK (4, 5). TKI inhibitors are targeted therapies for EGFR receptors, which bind to receptor target genes to block downstream pathways and improve tumor development (6, 7). Low expression of membrane proteins on PD-1T cells is an important molecule in regulating the autoimmune system (8, 9). Existing literature shows that the expression of PD-L1 is increased in tumor patients, and this protein can combine with PD-1, resulting in the occurrence of immune escape (10). The activation of YAP can be involved in tumor development. Studies have confirmed that the increased expression of YAP in serum and tissues of lung cancer patients can promote cell proliferation and metastasis (11, 12), so most drugs can inhibit tumor proliferation through HIpo-YAP. At present, there are few studies on TKI inhibition and its relationship with HIpo-YAP or PD-L1 at home and abroad. Invasive metastasis of colon cancer is the main reason affecting the prognosis. Therefore, it is very important to study the molecular mechanism of SDF-1/CXCR4 and integrin αvβ6 in liver metastasis of colon cancer. Floating cells were used to detect the appearance of β6, and the relationship between SDF-1/CXCR4 and the molecular mechanism of colon cancer redirection was analyzed. Use immunohistochemistry to detect the appearance of SDF-1/CXCR4 and αvβ6 protein, and combine the data with clinical-pathological data for statistical analysis. Experimental data showed that the positive expression rates of αvβ6 protein in well-differentiated and poorly differentiated colon cancer tissues were 21.4% and 30.6%, respectively, and the difference was statistically significant (P<0.01). The results show that the appearance of SDF-1/CXCR4 in colon cancer cells (CCC) has nothing to do with the type of cancer cells, and increases with the decrease of cell differentiation. This has a great relationship with the classification of the clinical TNM disease stage. The later the disease stage, the higher the expression level. Our work aims to screen key biomarkers related to neutrophils in sepsis using bioinformatics analysis. For this purpose, the microarray datasets related to neutrophils in sepsis patients were downloaded from the Gene Expression Omnibus (GEO) database. According to the Bayesian test, the Limma package in R was used to screen differentially expressed genes (DEGs). Then, DEGs were uploaded to the DAVID online diagnostic tool for subsequent Gene Ontology (GO) and the Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway enrichment on the selected DEGs. Next, protein-protein interaction (PPI) network was established based on the selected DEGs using the STRING website and the Cytoscape software. Furthermore, according to the function of the iRegulon plug-in in Cytoscape, our study further predicts and established regulatory networks related to transcription factors and regulatory genes. In addition, the miRWalk2.0 database was used to search for miRNA-DEG pairs, associated with the conduction of intersections of miRNAs predicted by TargetScan, Miranda, miRDB and RNA22 databases. Then, these miRNA-DEG pairs were also displayed in the form of a regulatory network through Cytoscape. Finally, two datasets were selected to verify the screened genes, regulatory factors, and miRNAs, to plot receiver operating characteristics (ROC) curves and compute the area under the curve (AUC) values. The results showed that AKT1, MMP9, ARG1, ETS1 targeting AKT1, and has-miR-124-3p targeting RPS6KA5 may have diagnostic value for patients with sepsis and septic shock. Reperfusion therapy is a standard method to treat cardiovascular diseases and has achieved high clinical effects. However, this treatment method is likely to cause myocardial ischemia-reperfusion injury. It has been reported that the Rho kinase inhibitor fasudil can interfere with cardiomyocyte apoptosis through the Rho-ROCK signaling pathway, so it is often used to treat cardiovascular diseases. The essay aims to research this specific influence of fasudil on cardiac damage in myocardial ischemia-reperfusion mouses through the Rho-ROCK signal path and its related mechanisms. Forty rats were taken as the research object, and the mouses were separated into control clusters. In the observation cluster of fasudil, the rat heart device was perfused by surgery. The rat coronary artery was ligated for 20 minutes to make the rat myocardial ischemia. Then, the ligation was loosened for myocardial perfusion to create a rat myocardial ischemia-reperfusion model. Observation group rats were perfused with quantitative fasudil, 80 minutes after ischemia-reperfusion, the ultrastructural changes and myocardial ischemic area of the rat myocardium were observed under a microscope, and the dynamic changes of the mouse heart were examined by flow cytometry. The PCR fluorescence method was used to explore the outlook layer of Rho-ROCK kinase activity to detect rat cardiomyocyte apoptosis. It is shown that under this intervention of fasudil, this expression level of Rho-ROCK kinase activity in the observation group was reduced by 18.3%, the myocardial cell apoptosis rate was decreased by 26.4%, and one area of myocardial ischemia can be reduced by 32.5%. The ultrastructure of the new object in rats is improved, and the left ventricular diastolic and systolic effect is enhanced. Studies have shown that insulin resistance has an important relationship with polycystic ovary. It is of great significance to explore the changes of inflammatory factors, oxidative stress, glucose and lipid metabolism, and insulin resistance in patients with PCOS. In the study process, 642 polycystic ovary patients in the first half of 2019 were divided into insulin resistance (n=357) and non-insulin resistance (n=285) groups. Oxidative stress index, glucose, and lipid metabolism index, and inflammatory factors were detected during the study process. The results showed that the levels of hs-CRP, TNF- α, and IL-6 in the IR group were 5.9mg/L, 9.2μg/L, and 87.2ng /ml, while those in the non-IR group were 4.6mg/L, 6.3μg/L and 51.5ng/ml, respectively. Thus, IL-6 and insulin levels maintain a dynamic balance. Low levels of IL-6 can promote insulin secretion, while high levels can inhibit its secretion. From January 2019 to April 2021, 100 female patients with common hematological tumors in our hospital were selected as the observation group, and 50 healthy women in our hospital during the same period were selected as the control group. The serum levels of follicle-stimulating hormone (FSH), estradiol (E2) and luteinizing hormone (LH) were detected and compared between the observation group and the control group before chemotherapy, patients with different disease types in the observation group, postmenopausal patients in the observation group, and postmenopausal patients in the observation group. Results showed that the serum FSH, E2 and LH levels of the observation group had no significant changes before chemotherapy (P > 0.05). Compared with before chemotherapy, the levels of serum FSH and LH in patients with different disease types in the observation group after chemotherapy were significantly higher, while E2 was significantly lower (P < 0.05), and the serum FSH, E2 and LH levels of postmenopausal patients in the observation group did not change significantly after chemotherapy (P > 0.05). The levels of FSH and LH in the observation group after chemotherapy were significantly higher, and E2 was significantly lower (P < 0.05). For this purpose, 60 SD rats were divided into four groups; the control group and treated groups with low, medium, and high concentrations of Danzikang. The histopathology of rats was analyzed and TGF-β signaling pathway-related proteins were determined. Results showed that the average optical density in serum of the Danzikang Granule intervention group was significantly higher than the control group (P<0.05), and the average optical density increased with drug concentration increasing (P<0.05). Compared with the control group, Danzikang knee granule cell survival in the intervention group was elevated the serum and reduced cell apoptosis rate (P < 0.05). Danzikang knee infusion concentrations were positively correlated with bone marrow mesenchymal stem cell survival rates (P < 0.05), and negatively correlated with apoptosis rate (P < 0.05). TGF-β1, BMP2, and BMP4 were significantly increased in the three concentrations of the Danzikang Granule serum intervention group (P<0.05). TGF-β1, BMP2 and BMP4 were significantly increased in the high concentration group, while TGF-β1, BMP2 and BMP4 were significantly decreased in the low concentration group (P<0.05). The Wakitani histological score of the control group was significantly lower than the other three groups (P<0.05). For this purpose, from June 2017 to December 2020, fresh NSCLC tissues and paired adjacent NSCLC tissues from 45 patients were collected. DNA and total RNA were extracted from non-small cell lung cancer (NSCLC) and paired non-cancerous lung tissues. The DNA library combined with a biotinylated probe was collected by Dynabeads m270 streptavidin beads. The concentration of the final library was determined by qubit dsDNA HS assay. Quantitative analysis of DMR methylation in 45 paired tumor and normal lung tissues was performed. RT qPCR and Western blot were used to verify the mRNA expression of candidate genes. Results showed that the methylation rate of CpG 7 in stxbp6 in stage III NSCLC was higher than that in stage I and early-stage II NSCLC; The methylation rates of cpg1 and 38-39 units in fzd10 were higher in stage I NSCLC than in stage II and III NSCLC; The methylation rates of CpG 6 in stxbp6 and CpG 4 and 20-21 in bcl6b in patients with tumor diameter > 3cm were higher than those in patients with tumor diameter < 3cm; Methylation of CpG unit 3 in stxbp6 is associated with age. Stxbp6, bcl6b, fzd10 and hspb6 mRNA expression were down-regulated in patients under 45 years old. The methylation rates of CpG 7 in stxbp6, CPG 6 in stxbp6 and CpG 4 and 20-21 in bcl6b were negatively correlated with the survival time of patients; The methylation rates of CpG 1 and 38-39 units in fzd10 were positively correlated with survival time (P<0.05). During the study, 30 pairs of primary cervical cancer tissues and adjacent tissues diagnosed by pathology were collected, and then placed in a cryopreservation tube, immediately placed in liquid nitrogen to freeze, and then moved to a -80 °C refrigerator for storage until use. They were dipped with crystal violet staining solution for 20 minutes, washed 3 times with PBS, and dried at room temperature. The polycarbonate film on the chamber was gently cut, placed on a glass slide, and covered with neutral resin. The number of cells in 5 random fields was counted under a microscope, and the differences between the groups were compared. The qRT-PCR results showed that compared with the normal cervical tissue cell line HCCR1, the expression levels of miR-494 in cervical cancer cell lines HCCR1 and C-33A were significantly reduced.
