The number of diarrheas, Bristol stool, traditional Chinese medicine (TCM) syndromes, clinical efficacy indexes, self-rating anxiety scale (SAS) score, the MOS item short from health survey (SF-36) scale score, peripheral blood cell levels of CD4+, CD8+, and Th17, gastrin (GAS), motilin (MTL), and cholecystokinin (CCK) levels was assessed before and after treatment. The adverse events were also recorded. Compared with those before treatment, all indexes of both groups were significantly improved (P<0.05). Compared with those of the control group, the number of diarrheas, Bristol stool, TCM syndrome score, SAS score, and CD8+ cell levels was significantly decreased after treatment in the experiment group (P<0.05). The clinical cure rate (48.0% vs. 73.1%), SF-36 score, GAS, MTL, CCK contents, and CD4+, and Th17 cell levels were significantly increased (P<0.05). No significant difference was in the incidence of adverse events between the two groups (P>0.05). The interaction among the four was analyzed to provide a research basis for the clinical treatment of neuroblastoma at the molecular level. The human neuroblastoma SK-N-SH cells were collected and cultured. According to the transfection methods, the cells were divided into control group (without any treatment), si-LINC0196 group (si-LINC0196 transfection), si-LINC0196-NC group (si-LINC0196 vector transfection), miR-584-5p group (miR-584-5p mimic transfection), miR-584-5p-NC group (miR-584-5p inhibitor transfection), miR-34a-5p group (miR-34a-5p mimic transfection), and miR-34a-5p-NC group (miR-34a-5p inhibitor transfection). The proliferation, migration, and apoptosis of SK-N-SH cells in each group were compared. The effects of LINC0196, miR-584-5p, miR-34a-5p, and TRIM59 were evaluated. The expressions of LINC0196 and TRIM59 in SK-N-SH cells in si-LINC0196, miR-584-5p, and miR-34a-5p groups were up-regulated. miR-584-5p and miR-34a-5p in si-LINC0196-NC, miR-584-5p-NC, and miR-34a-5p-NC groups decreased significantly (P < 0.05). The proliferation rate, migration rate, and invasiveness of SK-N-SH cells in miR-584-5p and miR-34a-5p groups were lower than those in si-LINC0196-NC, miR-584-5p-NC, and miR-34a-5p-NC groups, while the apoptosis rate increased (P < 0.05). After miR-584-5p and miR-34a-5p transfections, the relative activities of WT-LINC0196 and WT-TRIM59 dual luciferase were greatly inhibited (P < 0.05). For this purpose, forty-eight rats were randomly divided into sham group, autologous nucleus pulposus transplantation model group (NP group), neurotropin treatment group (NP+NT group), and solvent [normal saline (NS)] control group (NP+NS group). After 7 days of intervention, the mechanical paw withdrawal threshold (PWT) and thermal paw withdrawal latency (PWL) of the rats were measured, and the expression levels of Iba-1, c-JNK and CXCL1 in spinal cord tissues were measured by Western blot. The levels of tissue-associated inflammatory and anti-inflammatory factors in the spinal cord were detected by ELISA. Results showed that Neurotropin significantly alleviated mechanical and thermal hyperalgesia induced by NP transplantation and reduced levels of Iba-1, c-JNK, and CXCL1 proteins in the spinal cord tissue. In addition, neurotoxins also lowered concentrations of the inflammatory factors IL-1β, IL-6 and TNF-α. For this purpose, 30 healthy male SD rats were reared in a quiet and clean environment with alternating light for 12 hours. They were rolled randomly into 3 groups, each with 10 rats, namely the control (Ctrl) group, the experimental group, and the Dex group. The rats in the Ctrl were not treated, and the rats in the experimental group were intraperitoneally injected with 50μg/kg saline. After that, cardiac surgery was performed under CPB. Rats in the Dex group were injected with 50 μg/kg Dex intraperitoneally and underwent cardiac surgery under CPB. The Morris water maze (MWM) experiment was performed to test the learning and memory abilities and spatial positioning abilities of SD rats. Enzyme-linked immunosorbent assay (ELISA method) was adopted to detect the contents of TNF-α, IL-6, and IL-1β. Fluorescence quantitative PCR was applied to determine the mRNA expression levels of TNF-α, IL-6, and IL-1β in the hippocampus. Results showed that in the MWM experiment, in contrast with the Ctrl, the escape latency of the experimental group and the Dex group after surgery were prolonged (P<0.05), and the times they crossed platforms reduced (P<0.05). In contrast with the experimental group, the escape latency of the Dex group shortened, and the times they crossed platforms increased. ELISA suggested that in contrast with the experimental group, the concentrations of TNF-α, IL-6, and IL-1β in the Ctrl decreased (P<0.05), and those in the Dex group decreased slightly. In the fluorescence quantitative PCR experiment, in contrast with the experimental group, the mRNA expression levels of TNF-α, IL-6, and IL-1β in the Ctrl increased, and those in the Dex group decreased slightly. For this purpose, a total of 96 patients with NAFLD were selected as research subjects and randomly divided into a control group (n=48) and an observation group (n=48). The Control group was treated with rosuvastatin, based on which observation group received Clostridium butyricum capsule treatment. The efficacy in the two groups of patients was compared, and the intestinal flora, lipid metabolism, liver function and inflammation were observed. Results showed that the efficacy in the observation group was significantly better than that in the control group (p<0.05). After treatment, the content of Eubacterium rectale in the observation group was lower than that in the control group, while the content of Bacteroides thetaiotaomicron and Bifidobacteria was notably higher than that in the control group (p<0.05). Moreover, the observation group had remarkably lower levels of total cholesterol (TC), triglyceride (TG), free fatty acids (FFA), total bilirubin (TBIL), direct bilirubin (DBIL), alanine aminotransferase (ALT), aspartate aminotransferase (AST), procollagen III peptide (PIIIP), collagen-IV (C-IV), hyaluronicacid (HA) and laminin (LN) as well as lower levels of tumor necrosis factor-alpha (TNF-α), catabolite activator protein (CAP) and interleukin-6 (IL-6) in serum than the control group (p<0.05). Eighty gallbladder carcinoma patients undergoing open cholecystectomy in Heilongjiang Provincial Hospital from February 2016 to April 2019 were selected and divided into observation group (n=40) and control group (n=40) using a random number table. All patients underwent open cholecystectomy under general anesthesia and tracheal intubation. Patient-controlled intravenous analgesia was adopted after the operation in the control group, while right TPVB was performed before general anesthesia in the observation group. The changes in inflammatory factors and oxidative stress factors were compared between the two groups, the anesthesia resuscitation indexes, and the changes in the bispectral index (BIS) and Ramsay score during anesthesia resuscitation were recorded, and the changes in the hemodynamic indexes in perianesthesia and anesthesia resuscitation-related complications were analyzed. At 15 min after anesthesia, the observation group had lower levels of inflammatory factors high-sensitivity C-reactive protein (hs-CRP) and interleukin-6 (IL-6) (p<0.05), malondialdehyde (MDA) (p<0.05) and a higher level of superoxide dismutase (SOD) (p<0.05) than the control group. The anesthesia resuscitation time was shorter in the observation group than that in the control group (p<0.05). At 10 min, 20 min and 30 min after anesthesia, both BIS and Ramsay scores were significantly higher in the observation group than those in the control group (p<0.05). Moreover, the proportion of circulatory function-related complications and anesthesia resuscitation-related complications were lower in the observation group than that in the control group (p<0.05). The NRS score in the observation group was lower than that in the control group after anesthesia (p<0.05). Due to the lack of information about the frequency and resistance of Streptococcus pneumoniae to antibiotics, the present study was performed to determine the frequency of carriers of Streptococcus pneumoniae and its microbial resistance in children. For this purpose, the current descriptive cross-sectional study was conducted from November to March 2020 on 554 children aged 2-12 years in kindergartens and schools. This study collected samples with a sterile swab from the nasopharyngeal region, transported them to the laboratory by a transport medium, and then cultured them on an agar culture medium. After isolation, confirmatory tests and antibiotic susceptibility were performed. The results were analyzed using SPSS16 software and interpreted according to Mann Whitney U and Chi-Square Tests. Streptococcus pneumoniaewas found in 15% of samples, and the antibiotic resistance of the isolates to the antibiotics azithromycin, amoxicillin, rifampicin, amoxicillin-clavulanic acid, trimethoprim/Sulfamethoxazole, and ceftriaxone were 63.9%, 56.6%, 41%, 37.3%, 37.3%, and 3.6%, respectively. Also, 31.1% of the isolates were not resistant to any antibiotics. According to the results, excessive use of antibiotics has led to high resistance to azithromycin, amoxicillin, amoxicillin/clavulanic acid, and trimethoprim/Sulfamethoxazole, which indicates an increased risk of refractory infectious diseases. For this purpose, one hundred adult patients with primary pulmonary tuberculosis were selected for the study. The age of the enrolled cases ranged from 18 to 60 years old, with a controlled male to the female sex ratio of about 1:1. The Chinese medical evidence was considered to be a qi-yin deficiency type of pulmonary consumption. The patients were randomly divided into experimental and control groups, 50 cases each. In addition, 50 cases of healthy people were selected as a healthy control group, totaling 150 cases. In the experimental group, patients were given Baihe gujin decoction and Shengmai powder based on conventional western medicine, 1 dose for 1 day, 150mL/time for 2 times. The control group was treated with conventional Western medicine. 2 mL of fasting elbow venous blood from the subjects was taken in the morning. Peripheral blood mononuclear cells were isolated by density centrifugation. Monocytes were obtained after incubation with 5 μL of CD14+ immune magnetic beads at 4°C. The relative expression of IL1β and IL1R genes were measured using real-time quantitative PCR (RT-PCR), respectively. Results showed that the relative expression of IL1β and IL1R genes was significantly lower in the experimental group after 3 months compared with the control group, and the statistical difference was highly significant (p<0.01). Identifying microorganisms in environmental samples, particularly soil could be a valuable interest to study their effect on soil quality and plant growth. Through this study, we conducted a molecular characterization of bacteria found in the rhizosphere of Medico sativa plants grown in Hail soil and we highlighted their main properties. The sequences analyses revealed that the main bacterial isolates Pseudarthrobacter, Metabacillus, Priestia, and Massilia species. According to the sequences analysis and the phylogeny tree results, some of the identified bacteria were classified at the species level: Pseudarthrobacter was identified clearly as Pseudarthrobacter phenanthrenivorans; Metabacillus isolates grouped with Metabacillus sediminilitoris and the two Priestia isolates closely related to Priestia aryabhattai. We concluded that Hail soil is a niche of diverse bacteria with a high interest in soil environment and ecosystems. High levels of blood sugar may affect sperm quality and reduce the potential for male fertility. Increased levels of sperm DNA damage is often associated with reduced count and motility or abnormal morphology.</p><p><strong>Methods: </strong>This experimental study was conducted in Mashhad University of Medical Sciences. In this work, 40 mice (C57BL/6) were divided randomly into 4 groups: 1) Control, 2) Diabetic, 3) Diabetic + Insulin, and 4) Sham. After 35 days, the right epididymis of all specimens was used for Real-Time PCR and left epididymis for evaluation of sperm parameters using Aniline blue, Toluidine blue, Papanicolaou, and immunohistochemical study. Also, testes were applied for immunohistochemical, TUNEL studies, and biochemical assay.</p><p><strong>Results: </strong>Results of this study showed that chromatin integrity, morphology, cation channels of sperm (Catsper) expression, and biochemical factors level were significantly changed in diabetic mice in comparison to other groups (P<0.05) and treatment with insulin improved these parameters.</p><p><strong>Conclusion: </strong>Our findings showed that the sperm parameters such as DNA integrity, morphology, and Catsper expression change in diabetic mice.</p>","PeriodicalId":520584,"journal":{"name":"Cellular and molecular biology (Noisy-le-Grand, France)","volume":" ","pages":"8-18"},"PeriodicalIF":1.6,"publicationDate":"2022-02-28","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"40639877","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}
