The rat model of HF was established, and the rats were randomly divided into model group (HF model, n=20) and ALDH2 group (intervention with ALDH2, n=20), with a normal group (n=20) set. After successful modeling, MRI and ECG were applied to detect the cardiac function indexes of the rats. The myocardial function index creatine kinase (CK) was measured, the status of myocardial tissue injury was determined using hematoxylin and eosin staining, and the apoptosis was observed via terminal deoxynucleotidyl transferase-mediated dUTP nick end labeling (TUNEL) staining. The activity of ALDH2 was detected, and the expression levels of genes and proteins were measured through quantitative polymerase chain reaction (qPCR) and Western blotting assay. The model group had notably decreased fractional shortening (FS) and ejection fraction (EF) and remarkably increased left ventricular end-diastolic diameter (LVEDD) and left ventricular end-systolic diameter (LVESD) compared with the normal group (p<0.05). The activity of ALDH2 declined obviously in the model group. The myocardial tissue injury was severer in the model group, and the number of apoptotic cells in myocardial tissues was greater in the model group than that in other groups (p<0.05). The model group manifested higher expression levels of Caspase-3 and light chain 3 (LC3) than the ALDH2 group (p<0.05) but significantly lower expression levels of PINK1, Parkin and B-cell lymphoma-2 (Bcl-2) (p<0.05). In comparison with those in the model group, the protein expression levels of PINK1, Parkin and Bcl-2 in myocardial tissues were prominently higher in the ALDH2 group (p<0.05). In this paper, the detection of vaginal secretions and the positive rate of serum reproductive immune antibody in infertile women and normal pregnant women were used to explore the correlation between infertility and the two, so as to provide an effective scientific basis for the diagnosis and treatment of infertility. In this paper, 80 infertile patients (experimental group) and normal pregnant women (control group) were selected for the experiment. Real-time PCR was used to detect vaginal secretion and enzyme-linked immunosorbent assay. The results showed that the positive rates of UU and CT were 42.5% and 40%, respectively. The positive rate of the control group was 8.8% (Uu) and 7.5% (CT). In addition, the positive rates of ANA and AsAb in the experimental group were 40% and 44%, respectively, and those in the control group were 7.5% and 8.8%, respectively, with a significant difference (P<0.01). {"title":"Monitoring the effect of microbial culture on cleaning and sanitizing of the external ventilator circuit.","authors":"Fengling Yu, Feng Chen, Qi Yang, Qing Wen, Linghua Li","doi":"10.14715/cmb/2022.68.2.6","DOIUrl":"https://doi.org/10.14715/cmb/2022.68.2.6","url":null,"abstract":"This research aimed to investigate the microbial spectrum of microorganisms that cause ventilator-associated pneumonia (VAP) among ICU patients in the selected hospital, antimicrobial susceptibility, genetic diversity of common isolates, and the monitoring effect of microbial culture on cleaning and sanitizing of external ventilator circuits in order to reduce the occurrence of hospital infections. For this purpose, endotracheal aspirate (ETA) specimens were sampled from ICU patients with clinically suspected VAP in the hospital between August 2020 and August 2021 and then investigated for microbiological content. This was followed by Kirby-Bauer testing for determining drug sensitivity and ERIC-PCR for genotyping. Afterward, microbial culture was performed on cleaned, sanitized and dried ventilator external ventilator pipelines and those stored aseptically for 4 weeks to evaluate the cleaning and disinfection effect and measure the bacterial content. Results showed that in the 64 confirmed VAP cases, Klebsiella was the most frequently isolated organism, followed by P. aeruginosa and Acinetobacter baumannii, while Candida is the most widely isolated fungus. The antimicrobial susceptibility spectrum revealed that 40% of the isolates were multidrug-resistant (MDR). ERIC-PCR showed no genetic relationship between pneumococcal isolates. Through microbial culture, no pathogenic bacteria were detected among cleaned and sanitized ventilator external ventilator pipelines and those stored aseptically for 4 weeks, indicating a 100% pass rate. It was concluded that ventilators in intensive care units (ICU) are susceptible to contamination, exposing patients to bacterial contamination and other comorbidities. Gram-negative bacteria are the main pathogens of VAP, which are mostly multidrug-resistant. Forty rats were the experimental subjects. They were randomly grouped as a control group (Group C), an endotoxin group (Group E), an inhibitor group (Group I), and an activator group (Group A), with 10 rats in each group. The endotoxin-educed ALI rat model was built. Arterial Blood Gas Test (ABGT) was performed, and the Wet/Dry (W/D) ratio of lung weight was determined. The pathological variations in rat pulmonary tissues were scrutinized and scored. PMN in peripheral blood was isolated; its apoptosis was assessed, and its total NF-κB p65 and p-NF-κB p65 expressions were assessed. The expression of SNHG11 mRNA in pulmonary tissues was assessed. Results: Compared to Group C, the W/D ratios and pathological scores of Group E, Group I, and Group A boosted notably (P <0.05), while their ABGT indicators and PMN apoptosis rates dropped (P <0.05). Compared to Group E and Group I, the W/D ratio and pathological score of Group A dropped notably (P <0.05), while its ABGT indicators and PMN apoptosis rate boosted (P <0.05). Compared to Group C, the p-NF-κB p65 and SNHG11 expressions were boosted in Group E, Group I, and Group A (P <0.05); compared to Group E and Group I, the p-NF-κB p65 and SNHG11 expressions in Group A dropped (P <0.05). A role for vitamin D related to the immune system has been established. Vitamin D modulating effect is on the adaptive and innate immune responses. Our study aimed to compare serum levels of vitamin D in patients having different clinical symptoms of OLP (symptomatic or asymptomatic) with healthy individuals. Also, in this study, for further evaluation, the expression level of interleukin-17A and interleukin-6 (IL-17A and IL-6) was evaluated because the presence of active vitamin D reduces the expression of these pro-inflammatory factors. This study was included three groups with 30 volunteers in each. The first group included asymptomatic oral lichen planus patients (reticular or plaque-like lesions). The second group consisted of symptomatic oral lichen planus patients (atrophic or bullous-erosive lesions). In contrast, the third group consisted of healthy control subjects. The serum 25-hydroxyvitamin D was measured between the three groups and then correlated with clinical manifestation of oral lichen planus, either symptomatic or non-symptomatic. The Real-Time PCR technique was used to evaluate the expression of IL-17A and IL-6. Patients with symptomatic OLP (second group) had statistically significantly lower Vitamin D levels than asymptomatic OLP patients (first group). Healthy Controls (third group) exhibited statistically significantly higher vitamin D levels than OLP groups. The results of IL-17A and IL-6 genes expression showed that the presence of vitamin D had a statistically significant effect on reducing the expression of these two pro-inflammatory cytokines among symptomatic and asymptomatic OLP patients. Also, the results showed that there was a statistically significant difference between OLP patients (group I and II) and the control group (group III). In the study, the ovarian cancer cell lines (BT 549) were rolled into a control group (normal culture-defined as BT549/NC) and a response group (transfected with non-coding RNA LINC00857 cultured cells-defined as BT 549YAP cells). The expression and proliferation ability of the transcription factor YAP-TEAD in the two groups of cancer cells were analyzed and compared. The results showed that the YAP-TEAD expression rate was the highest in Bt549 cells; the YAP content grade (0.18) in BT 549-YAP cells was lower than BT 549/NC (0.2) after transfection (P< 0.05); and the apoptotic rate of the response group (80%) was higher than that of the control group (25%) after the intervention. With the extension of culture time, the expression of CCN1 mRNA decreased (P< 0.05), and CCN2 mRNA increased (P< 0.05). After 12, 24, 36, and 48 hours, the apoptosis rate of the reaction group at different time points was higher than that of the control group (P< 0.01). When YAP-TEAD was down-regulated, the in vitro proliferation ability of BT 549-YAP cells was weakened compared with BT 549/NC and parental cells. For this purpose, 30 SD healthy rats were selected, and 10 of them were randomly selected as the control group. The remaining 20 rats were established as acute myocardial infarction model rats, and randomly divided into model group and activated SIRT1 group. Interventions were performed on rats in each of the 3 groups. ROS staining, inflammatory factors [IL-6, IL-1β levels], H9c2 cell viability, Caspase3 and Caspase8 activity, antioxidant enzyme indexes [SOD, CAT, MDA levels], SIRT1, PI3K, MTOR, HIF-1α, HO-1, GLUT1 mRNA expression were compared between groups. Results showed that IL-6 and IL-1β levels were abnormally elevated in the model group compared with the control group (P<0.05). IL-6 and IL-1β levels decreased in the activated SIRT1 group compared with the model group (P<0. 05). H9c2 cell viability decreased and Caspase3 and Caspase8 activities increased in the model group compared with the control group(P <0.05). H9c2 cell viability increased and Caspase3 and Caspase8 activities decreased in the activated SIRT1 group compared with the model group (P<0.05). SOD and CAT levels were abnormally decreased and MDA levels were abnormally increased in the model group compared with the control group (P<0.05). SOD and CAT levels were abnormally increased and MDA levels were decreased in the activated SIRT1 group compared with the model group (P<0.05). PI3K and SIRT1 expression decreased and MTOR expression increased in the model group compared with the control group (P < 0. 05). PI3K and SIRT1 expression increased and MTOR expression decreased in the activated SIRT1 group compared with the model group(P<0.05). The expression of HIF-1α, HO-1 and GLUT1 mRNA increased in the model group compared with the control group, and the difference was statistically significant (P <0.05). The expression of HIF-1α, HO-1, and GLUT1 mRNA decreased in the activated SIRT1 group compared with the model group, and the difference was statistically significant (P<0.05). TB patients and HIV-infected/AIDS patients registered in Fuzhou Center for Disease Control and Prevention were selected as research subjects. The population characteristics of TB/HIV dual infection and mutation of drug-resistant genes were discussed. It was found that TB patients aged 20-40 years had the highest HIV infection rate, followed by those aged over 40 years. The rate of HIV infection in smear-negative TB patients was higher than that in smear-positive TB patients. HIV/AIDS patients aged 20-40 had the highest TB infection rate. In addition, men had higher rates of HIV than women, and married people had lower rates of HIV than single people. Mycobacterium tuberculosis (MTB) had the highest resistance to isoniazid (42.86%), followed by ofloxacin (34.82%), streptomycin (33.81%), and rifampicin (32.15%). Among the 113 cases of multi-drug resistant strains, 82 cases had mutations in the rpoB gene, with a gene mutation rate of 55.75%. The mutations ranged from codon 511 to codon 569. A total of 31 cases had mutations in the katG/inhA gene. Of which, there were 17 cases of katG single gene mutation, 9 cases of inhA single gene mutation, and 5 cases of combined katG and inhA gene mutation. It was suggested that it was necessary to carry out key TB/HIV two-way screening for TB patients older than 40 years old/smear-negative and male, single, and HIV-infected/AIDS patients aged 20-40. The resistance of MTB to antiTB drugs in this area was generally high, and the drug resistance of retreated patients was significantly higher than that of newly treated patients. For this aim, placenta samples were collected from five pregnant patients co-infected with syphilis and five undergoing full-term delivery before RT-PCR was performed to detect the mRNA expression of TLR2, TARC and MDC genes. The protein expression of TLR2, TARC and MDC genes was examined by Western Blotting. Results showed that TLR2, TARC and MDC were expressed in placental syncytiotrophoblast cells of patients with pregnancy-associated syphilis infection. TLR2 level was found significantly higher in placenta tissue of patients with pregnancy-associated syphilis infection compared with normal placenta tissue (P<0.05), so were TARC (P<0.05) and MDC genes (P<0.05). For this aim, the endometrial stem breast cancer cell line MCF-7 was cultured in vitro, and the overexpression mimic miR-138 mimics and the inhibitor anti-miR-138 were transfected into the endometrial stem breast cancer cell line MCF-7, which was set to overexpress miR-138 group and interfere with miR-138, and set up negative control of overexpression and negative control of inhibitor. Observe the cell proliferation and apoptosis ability of each group, and the changes in tumor necrosis factor-α (TNF-α), interleukin 1β, 6, 18 (IL-1β, IL-6, IL-18) levels, and compare the Bax of each group, NF-κB, VEGF protein expression level. Results showed that the proliferation ability of the miR-138 overexpression group was significantly lower than that of the miR-138 overexpression control group (P<0.05); the proliferation ability of the miR-138 interference group was significantly higher than that of the miR-138 interference control group (P<0.05). The apoptosis rate, caspase-3 and caspase-9 expression levels of the miR-138 overexpression group were significantly higher than those of the miR-138 overexpression control group (P<0.05); the apoptosis rate, caspase-3 and caspase-9 expression levels of the miR-138 interference group were significantly lower than those of the miR-138 interference control group (P<0.05). The expression levels of IL-1 β, IL-6, IL-18 and TNF - α in the miR-138 overexpression group were significantly lower than those in the miR-138 overexpression control group (P < 0.05). The protein expression levels of Bax, NF-κB and VEGF in the miR-138 overexpression group were significantly lower than those in the miR-138 overexpression control group (P < 0.05); the protein expression levels of Bax, NF-κB and VEGF in the miR-138 interference group were significantly higher than those in the miR-138 interference control group (P <0.05). The proliferation ability of the miR-138 overexpression group was significantly lower than that of the miR-138 overexpression control group (P < 0.05); the proliferation ability of the miR-138 + NF-κB overexpression group was significantly higher than that of the miR-138 overexpression group (P<0.05). The apoptosis rate of the miR-138 + NF-κB overexpression group was significantly lower than that of the miR-138 overexpression group (P < 0.05). Then MiR-138 can significantly inhibit the proliferation of breast cancer cells, promote apoptosis, and regulate the expression of inflammatory factors in the cells.
