However, the causal relationship between them remains unclear. Objective To investigate whether genetically predicted uveitis is related to the risk of PsA, and vice versa. Methods A two-sample bidirectional Mendelian randomisation (MR) design was employed, conducting a meta-analysis on data sourced from three distinct origins, followed by sensitivity analyses to ensure the robustness of the findings. Results MR analysis revealed a positive causal effect of uveitis on PsA. Meta-analysis results from three data sources revealed an odds ratio (OR) of 1.63, with a 95% confidence interval (CI) ranging from 1.22 to 2.19, and a statistically significant P-value of 0.001. Inverse MR results indicated a positive causal relationship between PsA and uveitis. Meta-analysis results: OR = 1.55, 95%CI = 1.07-2.24, P-value = 0.02. Limitations This study exclusively included individuals of European ancestry, thereby potentially limiting its generalisability to other populations, such as those of Asian or African descent. Secondly, Uveitis is a collective term for various intraocular inflammations, including anterior uveitis, intermediate uveitis, posterior uveitis, and pan uveitis. Conclusions The outcomes of our study indicate a significant association between uveitis and an elevated risk of PsA. Conversely, PsA is associated with an increased risk of uveitis. Aim To correlate the change trends of immunological biomarkers with timing of pemphigus relapse, and identify its predictors following rituximab treatment Methods This prospective cohort study included 44 patients with pemphigus treated with a rituximab biosimilar (Rheumatoid arthritis protocol). Clinical data were recorded at baseline, and immunological biomarkers (anti-DSG1, anti-DSG3, anti-AChRM3, CD19+ B-cells, CD19+27+ memory B-cells) were estimated at baseline and every three months till 12 months. Patients were clinically followed-up for 2 years or until relapse and grouped as early (<12 months) relapse, relapsing between 12-24 months, and no relapse till 24 months. Results Twenty-four (54.5%) patients relapsed by 2 years, with 18 (75%) relapsing within the first year. Early (3-6 months) rise in CD19+ B-cells, anti-DSG3 and anti-DSG1 levels, and delayed (9-12 months) anti-DSG3 rise distinguished early from late relapse, while patients remaining relapse-free exhibited no significant immunological changes. At month 6, 73.3% of early relapsing patients had anti-DSG3 >120 RU/mL, as compared to 27.3% of patients without early relapse (p=0.008). Early relapse rates were significantly higher in patients with incomplete B-cell depletion at 3 months (100% vs 35%; p=0.02). There was no significant difference in the relapse rates based on B-cell repopulation (p=0.20). Incomplete B-cell depletion at 3 months and/or anti-DSG3 >120 RU/mL at 6 months had a positive and negative predictive value of 64.7% and 87.5% respectively, for early relapse. This integrative review used the electronic databases MEDLINE/PubMed and the Virtual Health Library (VHL), including 12 studies conducted predominantly in Brazil, China, India, and Ethiopia. We focused on the role of regulatory T (Treg) cells in Hansen's disease, specifically examining their frequency of expression and the functional characteristics of their subpopulations. Findings reveal the ubiquitous nature of Treg across clinical forms, particularly the CD3+CD4+CD25+FoxP3+ phenotype, with a pronounced frequency in multibacillary cases, including borderline-lepromatous and lepromatous leprosy. In these cases, a notable upregulation of inhibitory molecules (CTLA-4, PD-1) and the production of immunoregulatory cytokines (IL-10, TGF-β) are evident in skin lesions and/or in vitro-cultured cells. The intricate interplay of local stimuli, cytokine profiles, antigenic nature, receptor expressions, co-inhibitory molecules, and macrophage types modulates the cellular immune response, influencing disease severity. The increased presence of Treg and their associated products appears to contribute to bacillary multiplication in patients with the disseminated form, fostering disease persistence. Skin ageing is characterised by a decrease in the levels of collagen, elastin fibre, and hyaluronic acid (HA), resulting in a loss of skin elasticity and hydration. Prior studies suggest that oral collagen supplements can help increase collagen, elastin, and HA levels, ultimately improving skin health. Objective To assess the impact of collagen-based supplements on the signs of skin ageing and determine the effective dosage. Methods This systematic review and meta-analysis utilized PubMed, Clinical Key, ProQuest, and Cochrane Library databases to identify studies published from 2010 to 2021. It includes ten randomised controlled trials (RCTs) with a total of 646 participants. Results The findings indicate that collagen supplements were statistically effective in increasing skin hydration (SMD 1.25 (CI: 0.77-1.74)) and elasticity (SMD 0.61 (CI: 0.21-1.02)). The most commonly administered dose was 4 g/day, with a median dose of 3.5 g/ day. Limitations Most of the studies could not control for weather-related factors. The findings are more applicable to females. Moderate heterogeneity (I2 = 55.5-56.5%) may limit generalisability. The risk of bias was generally unclear, and publication bias could not be assessed due to the small number of studies.
