It is believed to have range of toxicity from acute to chronic and also can cause DNA strand breaks and chromosomal aberrations in humans. This study was aimed at characterizing the potential of previously isolated bacteria for butachlor degradation. The isolates from culture collection, labelled A-F were screened for butachlor degradation on Bushnell Hass agar media with butachlor as a sole carbon source. Isolate A (molybdenum-reducing and aniline-degrading Pseudomonas sp.) was observed to grow best and tolerated the highest concentration of butachlor supplemented in the media after 72 h of incubation at 37 ℃. Characterization study revealed that the Pseudomonas sp. can utilize and grow with butachlor at optimum pH between 6.0 - 6.5, temperature between 30 – 37 ℃ and can tolerate up to 600 mg/L butachlor concentration with increase in growth as inoculum size increases. Additionally, this bacterial strain shows no lag phase regardless of the concentration of the herbicide used and reach its maximum growth after 24 h of incubation. In addition, sigmoidal curves are often seen in these tests. In the event of asymmetry, a logistic (5PL) curve, or a logistic (4PL) curve, may be the best way to represent a distinctive sigmoidal relationship. It is possible that the linearization of an otherwise nonlinear connection by log transformation may result in a disruption of the error structure of the curve, and that this will have the opposite effect of decreasing or even eliminating error in the relationship. Previously, a surface plasmon resonance biosensor for the detection of squamous cell carcinoma antigen (SCCa) using nanoparticle technology was developed. However, based on the calibration curve, it conforms to the majority on sigmoidal shape curve for antibody-type sensing system. The resultant curve showed a sigmoidal calibration curve but was not modelled according to any of the sigmoidal models available. When its concentration is elevated, it poses a great danger as it can interfere with the processes of spermatogenesis and oogenesis in several animals including fish. Its remediation using molybdenum-reducing bacterium is an emerging important tool. One of the parameters that are important in governing molybdenum reduction is temperature. Several models may be used to simulate the growth rate of microorganisms on various media as a function of temperature. Arrhenius is one of the very popular models since it has minimal parameters. Temperature generally affects microbial growth and metabolic activity on their substrates. The small nature of microbes makes them susceptible to change in the surrounding temperature. Growth on molybdenum by Serratia marcescens strain DRY6 is described, with a discontinuous chevron-like graph of apparent activation energy with a breakpoint at 26.25 oC. Regression analysis resulted in two activation energies: 15-25 oC and 30–40 oC with the activation energies of 52.7 kJ/mol and 13.33 kJ/mol, respectively. For the examined temperature range (15-25 oC), a Q10 value of 2.08 and a theta value of 1.02 was calculated. This process is impacted by a change of the surrounding temperature. In general, to model the impact of small temperature ranges on the growth rate of microorganisms on substrates, several models are available. Arrhenius is one of the models and is very common because it has few parameters. An apparent activation energy based on the Arrhenius plot on the rate of molybdenum reduction by Pantoea sp. strain HMY-P4 is reported in this study. A linear plot of ln qmax against 1/T at the temperature range studied (25–40 oC) resulted in an activation energy value of 45.69 kJ per mol (95% Confidence Interval or C.I., 32.04 to 59.35). The plot also yields the Arrhenius frequency factor (A) which was calculated as 2.71 × 107 (95% C.I., 1.25 × 105 to 5.86 × 109). The corresponding Q10 value of 2, which represents a relative 10 ˚C increase in the surrounding temperature and a doubling of the reaction rate. The Q10 value of 1.82 (95% C.I., 1.52 to 2.18) and a theta value of 1.06 (95% C.I., 1.05 to 1.08) was calculated. These issues have warranted the development of sensitive detection and therapeutic devices. A fluorescence-based detection of the pathogen E. coli is previously developed using a mini-emulsion technique to create fluorescent/electroactive poly(3-hexylthiophene) P3HT nanoparticles (NPs) stabilized with CTAB or cetyltrimethylammonium bromide, a quaternary ammonium salt forming CTAB-P3HT NPs. Limits of detection for the bacterium based on fluorescence spectroscopy is 5 CFU/mL. The curve showed a sigmoidal calibration curve but was not modelled according to any of the sigmoidal models available. The aim of this study is to use the Four-Parameter Logistic Model to determine the LOD value more accurately. The modelling exercise gave values of the parameters a, d, Log EC50 and Hillslope representing the maximum and minimum responses, value that produces a 50% signal response, and a slope-like parameter (Hill coefficient) of -0.01427 (DF/F0), 0.6189 (DF/F0), 3.473 and 0.413, respectively. The LOD value was 30 CFU/mL with the 95% confidence interval from 13 to 64 CFU/mL. The usage of the 4Pl model in this study was successful, since it was able to represent the entire date curve. The correlation coefficient values of 0.986 indicated good fitting of the experimental data to the 4PL model. This organism produced endoglucanase (CMCase) and exoglucanase (FPase) when grown at a different range of parameters pH (6, 7, 8, 9, 10 and 11), temperature (30, 40, 50 and 60OC), incubation time (4th 6th 8th 10th and 12th day) and nitrogen source (NH4SO4, NH4Cl, NaNO3 and NH4HCO3). Carboxymethylcellulose (CMC) and cellulose powder were used as the sole carbon source. In this research, statistical tools called Response Surface Methodology (RSM) was used for the optimization of cellulase enzyme by selecting three important parameters after one factor at a time (OFAT) approach. Using OFAT, optimum production of both CMCsase (3.52U/mL) and FPase (4.07U/mL) were achieved after 8 days incubation at pH 8, temperature 30OC and 1.0g/L ammonium sulphate while RSM produced CMCase 3.91U/mL and FPase 4.26U/mL respectively when incubated for 8 days at pH 8.5, temperature 45 and 3% ammonium sulphate concentration. Optimization of the culture conditions using RSM leads to an increase of 0.39U/mL (CMCase) and 0.19U/mL (FPase). The use of RSM has gained considerable attention in the past decade in the optimization of various physicochemical parameters and nutritional factors. Its application in different industries may find ways of selecting different factors influencing cellulase activity. The fungus can produce a considerable amount of cellulase enzyme at a pH of up to 10 and 50OC. The concentration at which enhanced microbial activity is seen, on the other hand, results in a significant decrease in growth rate as well as an increase in lag time (due to the higher lag time). An established link exists between heavy metal toxicity in microorganisms and the process of bioremediation, which has been well-documented. Because heavy metals have an impact on bioremediation, they must be researched, and appropriate countermeasures must be implemented. Copper reduced the growth of the SDS-degrading bacteria Enterobacter sp. strain Neni-13 to a significant extent. Under varying doses of mercury, the SDS-degrading bacteria exhibited a sigmoidal pattern with time periods ranging from 7 to 10 hours. Gompertz's model was used to calculate the growth rates of copper in different concentrations. As the copper concentration rose, the growth of bacteria was suppressed with a concentration of 1.0 g/L, with virtually total stoppage of bacterial development. From the Gompertz model, we got the estimates of growth rates; after which, they were estimated according to the Han-Levenspiel, Shukor, Wang, Liu, Andrews, and Amor models. The modified Han-Levenspiel, Andrews, Liu, and Shukor models could all successfully fit the curve. Results of the statistical analysis showed that the Han-Levenspiel model was the best model based on highest adjusted correlation coefficient (adR2), the lowest values for RMSE and AICc, and values of AF and BF closest to unity. The parameters obtained from the Han-Levenspiel model were Ccrit 0.209 mg/L (95%, C.I., 0.199 to 0.219), μmax 0.209 h-1 (95% C.I., 0.199 to 0.219) and m 0.472 (95% C.I., 0.383 to 0.561. Estimating the accuracy of thermodynamic parameters is dependent upon the equilibrium constants between two phases (KL: dimensionless). In this study, thermodynamic parameters were calculated from the KL constant derived from the adsorption of Langmuir isotherms of Pb(II) on an sulphuric acid-treated brown algae Cystoseira stricta biomass at different temperatures. The conversion of the KL values to the dimensionless Kc values based on the Langmuir model was then assessed for thermodynamic parameters via the Van’t Hoff’s equation. The Pb(II) adsorption process onto the Cystoseira stricta biomass was spontaneous and feasible with DG values at 25, 30, 35 and 40 oC of -24.56 (95% C.I., -24.00 to -25.03), -22.60 (95% C.I.,-21.95 to -23.12), -20.90 (95% C.I.,-18.87 to -22.01) and -17.17 (95% C.I.,-17.71 to -21.41), respectively, and occurred in an endothermic nature (DH= -0.30 kJ/mol (95% C.I., -194.34 to 193.74) with an increased in randomness (DS= -299.47 J/mol×K (95% C.I., -553.60 to -45.35) kJ/mol). Significant parameters such as precise reduction rate, theoretical maximum reduction and whether reduction at high molybdenum concentration influenced the lag time of reduction can be discovered by mathematical modeling of the reduction phase. While common, the use of the linearization method by the use of natural logarithm transformation is inaccurate and can only provide an approximate value for the calculated single parameter, the real growth rate. In this work, a variety of models for such as logistic, Gompertz, Richards, Schnute, Baranyi-Roberts, Von Bertalanffy, Buchanan three-phase and more recently Huang were utilized to obtain values for the above parameters or constants. The Huang model was the best model in modelling the Mo-blue production curve of the Serratia sp. strain DRY5 based on statistical tests such as root-mean-square error (RMSE) (0.043), adjusted coefficient of determination (R2) (0.994), bias factor (BF) (1.00), accuracy factor (AF) (1.03) and corrected AICc (Akaike Information Criterion) (-67.02). Parameters obtained from the fitting exercise were maximum Mo-blue production rate (m), lag time () and maximal Mo-blue production (Ymax). We make use of primary growth models for modeling Mo-blue output in this work. This is an emerging method of identifying constants of parameters that control the reduction of molybdenum. For the creation of further secondary models, the constants calculated from this work will be utilized for such purpose. Mathematical models can also be utilized to estimate or predict the possible actions, to replicate a prediction or retroactivity and to schedule better experiments. Bacteria are an emerging instrument for the bioremediation of the molybdenum pollutant in the world, molybdenum reduction into insoluble molybdenum blue. In this study, the molybdenum reduction rate by the bacterium Morganella sp. was studied for its inhibitory kinetics characteristics using sodium molybdate as a substrate. Modeling kinetics showed that molybdenum reduction could be explained by several models such as Monod, Haldane, Hans-Levenspiel, Luong, Teissier, Aiba and Yano with Aiba as the best model as judged using error function analysis such as bias and accuracy factors (BF and AF), root mean square error (RMSE), adjusted coefficient of determination (adjR2), and corrected Akaike Information Criterion (AICc). The calculated value for the Teissier-Edward’s constants, which are qmax, Ks, and Ki that are maximal reduction rate, half saturation constant for maximal reduction, half saturation constant for inhibition of reduction were 7.77 (95% C.I., 4.41 to 19.95) µmol Mo-blue hr-1, 26.63 (95% C.I., 12.82 to 40.44) mM and 51.39 (95% C.I., 23.67 to 79.10) mM, respectively. The true maximal reduction rate, which occurred when the slope of the curve is zero occurs at 36 mM molybdate concentration and a corresponding value of 1.85 µmol Mo-blue hr-1.
