The probe was designed using Primer3Plus program, labeled with the non-radioactive molecule digoxigenin (DIG) and used in the hybridization method with the dot blot technique. The test samples consisted of two types, i.e. genomic DNA samples from pure bacterial cultures and from artificially infected tomato seeds with C. michiganensis subsp. michiganensis. Samples derived from pure bacterial cultures showed positive hybridization results at all levels of DNA concentration; while samples from tomato seeds only showed positive reactions at concentrations of 10, 8, 6, and 4 g L-1. Erwinia carotovora subsp. carotovora) merupakan salah satu penyakit penting pada tanaman sawi putih. Bakteri ini menghasilkan enzim pektinase yang dapat menguraikan pektin pada dinding sel tanaman bagian lamela tengah. Salah satu alternatif pengendalian penyakit busuk lunak ialah menggunakan agens biokontrol dari filosfer. Penelitian ini bertujuan menyeleksi dan menguji bakteri filosfer dari daun kubis yang berpotensi menghambat penyakit busuk lunak P. carotovorum pada sawi putih. Bakteri filosfer diisolasi dari wilayah Cianjur, Tegal, dan Bogor. Bakteri filosfer diuji keamanan hayati berdasarkan pada reaksi hipersensitivitas dan kemampuan lisis pada agar-agar darah, serta diuji kemampuan penghambatannya terhadap P. carotovorum berdasarkan pada uji antagonis secara in vitro dan uji penghambatan penyakit secara in vivo. Bakteri filosfer dikarakterisasi berdasarkan pada morfologi koloni dan sifat Gram. Kemampuan hidup yang tinggi sebagai saprob pada tunggul tanaman dan sisa-sisa akar yang mati menjadikan bagian ini sebagai sumber inokulum penting, baik untuk tanaman karet yang sehat di sebelahnya maupun pada saat melakukan penanaman ulang. Oleh karena itu, pengurangan sumber inokulum ini menjadi kunci penting untuk mencegah penyebaran lebih lanjut. Solarisasi tanah merupakan salah satu metode disinfeksi tanah sebelum tanam dalam pengendalian beberapa hama penyakit tertentu dan dapat mengurangi penggunaan senyawa sintetik di dalam tanah. Penelitian ini bertujuan menentukan solarisasi tanah dan penambahan bahan organik terhadap kelangsungan hidup R. microporus. Solarisasi tanah secara nyata menekan kelangsungan hidup R. microporus pada kedalaman lokasi patogen, yaitu 5 dan 15 cm dari permukaan tanah. Sementara itu perlakuan bahan organik tidak berpengaruh nyata dalam menekan perkembangan R. microporus. Semakin lama waktu solarisasi, kemampuan penekanan terhadap daya hidup inokulum semakin tinggi. Pemencaran penyakit ini diketahui terjadi melalui bibit tanaman terinfeksi dan serangga vektor kutudaun.  Penelitian dilakukan untuk menguji efisiensi penularan PRSV melalui dua spesies kutudaun, yaitu Aphis gossypii dan Myzus persicae dan membuktikan bahwa PRSV tidak dapat ditularkan melalui biji. Penularan PRSV isolat Medan melalui kutudaun dilakukan pada tanaman pepaya var. California dengan periode makan akuisisi dan periode makan inokulasi masing-masing selama 10 menit. Minimal 5 ekor A. gossypii dan 10 ekor M. persicae diperlukan untuk keberhasilan penularan PRSV. Penularan PRSV melalui A. gossypii menghasilkan insidensi penyakit yang lebih tinggi dan gejala penyakit yang lebih parah dibandingkan dengan penularan melalui M. persicae. Bibit pepaya dari biji yang berasal dari buah yang menunjukkan gejala bercak bercincin tidak menimbulkan gejala penyakit dan tidak diperoleh fragmen DNA spesifik PRSV pada deteksi PRSV menggunakan metode reverse transcription polymerase chain reaction. This pathogen can survive in the soil for several years without a host plant. This study aimed to examine the viability and virulence of 21 isolates of F. oxysporum f. sp. zingiberi after being preserved for 17 years in sterile soil. Fungal viability was determined by descriptive method, while the experiment using randomized block design was conducted to examine the virulence of fungal isolates.  The treatments consisted of control, 21 isolates of F. oxysporum f. sp. zingiberi from Boyolali and Temanggung, each treatment was replicated three times. The variables consisted of colony colors and diameters, macroconidia and microconidia shapes, growth time, dry weight of mycelia, conidia density, incubation period, affected area, rhizome wet weight difference, and waste index. The results showed that all fungal isolates which were stored in sterile soil for 17 years still had the ability to grow well on PDA medium and fill up petri dishes in 11–36 days. Moreover, all the isolates caused infection and disease symptoms development in ginger rhizome var. Penelitian ini bertujuan mengidentifikasi spesies Meloidogyne secara morfologi dari tanaman seledri yang berasal dari sentra sayuran di Banjarbaru. Pengambilan sampel tanaman seledri yang terinfeksi NPA dilakukan dengan metode purposive sampling pada tiga lokasi di Kelurahan Landasan Ulin Utara, yaitu Desa Kurnia, Desa Sukamara, dan Desa Sukamaju Ujung. Identifikasi spesies Meloidogyne diperlukan dalam merancang strategi pengendaliannya yang efektif. Penelitian ini bertujuan mendeteksi dan mengidentifikasi spesies Meloidogyne pada kentang secara morfologi, morfometri, dan molekuler. Sampel ubi kentang bergejala bintil, malformasi bentuk ubi, permukaan ubi tidak rata, serta permukaan ubi bergelombang dikumpulkan dari tiga sentra produksi kentang di wilayah Sumatra, yaitu Karo (Sumatra Utara), Solok (Sumatra Barat), dan Kerinci (Jambi). Ekstraksi nematoda dilakukan dengan teknik pembedahan jaringan ubi berbintil. Identifikasi morfologi dilakukan berdasarkan pola perineal nematoda betina. Pengukuran morfometri dilakukan terhadap juvenil 2 berdasarkan formula de Man. Identifikasi molekuler dilakukan dengan teknik PCR dilanjutkan dengan perunutan nukleotida dan analisis filogenetika. Tiga spesies Meloidogyne yang diidentifikasi ialah Meloidogyne arenaria, M. incognita, dan M. javanica. Amplifikasi DNA menggunakan primer spesifik CO1 berhasil mengamplifikasi pita DNA sebesar ±360 pb untuk M. arenaria, ±326 pb untuk M. incognita, dan ±170 pb untuk M. javanica. Hasil perunutan nukleotida menunjukkan bahwa isolat M. incognita asal Karo-Indonesia berkerabat sangat dekat dengan spesies sejenis dari negara Cina, Amerika Serikat, Vietnam, Inggris, Brazil, dan Afrika Selatan. Penyakit hawar pelepah ini menjadi masalah yang cukup serius di berbagai negara penanam padi, termasuk Indonesia. Pengendalian yang mengandalkan penggunaan pestisida sintetis dan varietas tahan sering kali menimbulkan permasalahan baru baik pencemaran lingkungan maupun munculnya populasi patogen resisten sehingga masih diperlukan pengendalian yang lebih aman dan ramah lingkungan. Penelitian ini melaporkan potensi bakteri endofit asal tanaman padi sehat dalam menghambat pertumbuhan R. solani secara in vitro dan kemampunnya dalam menekan infeksi R. solani pada perkecambahan padi. Percobaan menggunakan metode kultur ganda antara galur-galur bakteri endofit dan R. solani. Pengamatan dilakukan terhadap pertumbuhan koloni R. solani dan pertumbuhan miselium secara mikroskopis. Pengujian dilanjutkan dengan memperlakukan benih padi menggunakan bakteri endofit. Benih padi yang telah dilapisi dengan bakteri endofit kemudian ditumbuhkan pada medium agar-agar dekstrosa kentang yang telah ditumbuhi R. solani. Tingkat gangguan penyakit hawar pada benih menunjukkan bahwa hampir semua galur bakteri yang diuji dapat menghambat pertumbuhan koloni R. solani dan juga menyebabkan malformasi pada miseliumnya. Aplikasi bakteri endofit pada benih padi juga secara nyata menurunkan tingkat infeksi R. solani pada benih padi. Di antara bakteri yang diuji, OS7 berpotensi dikembangkan sebagai agens pengendali hayati hawar pelepah yang disebabkan oleh R. solani. Antraknosa sangat merugikan karena menurunkan hasil produksi. Tujuan penelitian adalah mendapatkan cendawan laut yang berpotensi menghambat pertumbuhan C. acutatum dan menekan penyakit antraknosa, serta mengidentifikasi isolat cendawan laut yang potensial sebagai agens pengendalian hayati. Potensi agens hayati mengacu pada hasil penapisan seperti uji antagonis dan senyawa organik volatil (SOV), sedangkan penghambatan penyakit antraknosa pada cabai didasarkan pada hasil pengukuran insidensi penyakit dan diameter gejala antraknosa. Hasil uji menunjukkan semua isolat cendawan laut berpotensi menghambat pertumbuhan C. acutatum dan menghasilkan SOV, serta dapat menekan insidensi penyakit antraknosa pada cabai. Empat galur cendawan laut, yaitu 4145, B3st2, GN322, Z521 memiliki potensi menjadi agens pengendalian hayati penyakit antraknosa pada cabai.  Hasil identifikasi empat galur tersebut berdasarkan analisis sikuen nukleotida adalah Fusarium proliferatum 4145, Trichoderma harzianum B3st2, fungal endofit GN322, dan Fusarium solani Z521.","PeriodicalId":31619,"journal":{"name":"Jurnal Fitopatologi Indonesia","volume":" ","pages":""},"PeriodicalIF":0.0,"publicationDate":"2022-07-27","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"45158712","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}
