Tanaman sakit menunjukkan gejala antara lain: tanaman kerdil, daun menguning, nekrotik akar dan puru akar. Ekstraksi nematoda dilakukan dari 100 mL sampel tanah dan 5 g sampel akar tanaman jambu biji. Identifikasi dilakukan secara morfologi dengan mengamati karakter kunci dari setiap genus fitonematoda yang ditemukan. Penghitungan jumlah nematoda dilakukan pada setiap genus fitonematoda. Enam genus fitonematoda berhasil diidentifikasi, yaitu Criconemoides, Helicotylenchus, Hoplolaimus, Meloidogyne, Rotylenchulus, dan Xiphinema dengan jumlah berkisar 8–2300 ekor. Salah satu cara penanganan pascapanen untuk mempertahankan kualitas buah salak pondoh ialah dengan penambahan bahan tambahan pangan kalsium klorida dan bahan pelapis kitosan. Penelitian ini bertujuan menguji pengaruh CaCl2 dan kitosan untuk mengendalikan T. paradoxa, penyebab busuk hitam pada buah salak pondoh. Perlakuan terdiri atas CaCl2 6%, kitosan 1.5%, CaCl2 6% + kitosan 1.5%, dan kontrol.  Hasil penelitian menunjukkan bahwa kitosan 1.5% lebih efektif dalam mempertahankan kualitas buah salak selama 12 hari penyimpanan dibandingkan dengan perlakuan lainnya. Perlakuan ini menghambat peningkatan populasi mikrob, persentase kerusakan daging buah, persentase susut bobot, dan penurunan nilai organoleptik dibandingkan dengan kontrol pada suhu ±28 °C dan kelembapan relatif 65%–75%. Penelitian bertujuan memperkirakan penurunan hasil padi akibat penyakit hawar pelepah berdasarkan keparahan penyakit yang terjadi pada stadium masak susu. Penelitian dilaksanakan di rumah kawat dan lahan percobaan Balai Besar Penelitian Tanaman Padi pada musim hujan 2019/2020. Penelitian di rumah kawat menggunakan tiga varietas padi yaitu “Ciherang”, “Inpari 32 HDB”, dan “Baroma”; sedangkan penelitian di lahan percobaan menggunakan “Ciherang”. Inokulasi R. solani dilakukan pada tanaman padi stadium anakan maksimum dengan menyisipkan tanaman terinfeksi sebagai sumber inokulum penyakit diantara rumpun padi pada tingkat insidensi antara 5%–50%. Tingkat keparahan penyakit pada tanaman di rumah kawat dan lahan percobaan terus berkembang selama periode pengamatan dan mencapai berturut-turut 60.09% dan 70.56%.  Lebih lanjut, keparahan penyakit yang tinggi menyebabkan penurunan hasil yang tinggi pula. Materi genetik yang diuji yaitu 19 varietas padi yang sudah dilepas dan 50 aksesi plasma nutfah padi koleksi Balai Besar Penelitian Tanaman Padi Badan Litbang Pertanian-Kementerian Pertanian. Pengamatan mengikuti metode skoring SES IRRI 2014. Gen AB FAR-1 diketahui sebagai gen penting yang mengendalikan patogenisitas A. besseyi.  Penelitian dilakukan untuk mengetahui karakter gen AB FAR-1 yang diisolasi dari nematoda yang berasal dari benih padi.  Ekstraksi nematoda dilakukan dengan metode corong Baerman dari benih 5 varietas padi “Ciherang“, “Inpari Sidenuk“, “Sintanur“, “Hibrida Prima“ dan “Pak Tiwi“.  Ekstraksi DNA total nematoda menggunakan metode CTAB dilanjutkan dengan amplifikasi gen AB FAR-1 menggunakan primer spesifik FAR-F1/R1 dan analisis  urutan nukleotidanya.  Pita DNA spesifik gen AB FAR-1 berukuran 150 pb berhasil diamplifikasi dari semua sampel nematoda.  Analisis sekuen menunjukkan bahwa gen AB FAR-1 tersebut memiliki homologi tertinggi (92.5 – 100%) dengan aksesi Genbank JQ686690.1, yaitu gen AB FAR-1 A. besseyi asal Cina.  Walaupun memiliki homologi yang tinggi, terdapat beberapa perbedaan nukleotida pada sampel gen AB FAR-1 A. besseyi asal “Ciherang“, “Inpari Sidenuk“ dan “Hibrida Prima“.  Analisis pohon filogenetika lebih lanjut mengelompokkan gen AB FAR-1 A. besseyi menjadi 2 grup, yaitu grup 1 terdiri atas gen AB FAR-1 A. besseyi asal Cina, “Sintanur“, “Hibrida Prima“ dan “Pak Tiwi“ dan grup 2 gen AB FAR-1 A. besseyi asal “Ciherang“, dan “Inpari Sidenuk“.","PeriodicalId":31619,"journal":{"name":"Jurnal Fitopatologi Indonesia","volume":" ","pages":""},"PeriodicalIF":0.0,"publicationDate":"2021-07-25","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"45627466","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Infeksi Meloidogyne incognita (nematoda puru akar) pada tanaman kenaf menyebabkan tanaman kerdil sehingga menurunkan produksi tanaman. Penelitian ini bertujuan menentukan ketahanan tujuh genotipe kenaf terhadap M. incognita. Percobaan dilakukan dengan menginfestasi tanaman kenaf yang berumur 15 hari setelah tanam (HST) dengan M. incognita pada populasi 40 nematoda juvenil 2 per 100 g tanah. Medium tanam yang digunakan ialah tanah berpasir dengan komposisi pasir 55%, debu 36%, dan liat 17%. Variabel ketahanan terdiri atas indeks puru akar dan faktor reproduksi nematoda. Analisis asam salisilat, fenol, lignin serta beberapa variabel pertumbuhan tanaman dilakukan pada umur tanaman 75 HST. Diantara tujuh genotipe tanaman kenaf yang dievaluasi terdapat 3 genotipe yang toleran (KR4, KR15 dan KR5) dan 4 genotipe sangat rentan (KR1, KR6, Kin2, dan DS028). Genotipe yang memiliki respons toleran terhadap M. incognita menunjukkan peningkatan senyawa fenol, asam salisilat, dan lignin pada akar dibandingkan dengan kontrol. This study aimed to determine the effectiveness of the zero energy cool chamber (ZECC) in reducing the severity of anthracnose in Capsicum frutescens L. and extending the shelf life of chilies. The research was started by designing and building ZECC as a storage area for chilies. Chilies with and without fungal inoculation were stored in ZECC and storage chamber at room temperature. C. gloeosporioides experienced a growth inhibition of 40.48% after being stored in ZECC compared to its growth at room temperature. Storage of chilies in ZECC was able to reduce the severity of anthracnose by 56.2% on the 15th day, did not affect vitamin C content and total dissolved solids (TSS) and was able to reduce chilies weight loss during storage. Based on the organovisual test using the visual quality rating method and the Hedonic sensory test, consumers prefer chilies stored in the ZECC. The results showed that storage in ZECC could extend the shelf life of chilies. This storage model does not use electricity or is zero energy so it can be used by small scale farmers.
