Sudatri","doi":"10.24843/metamorfosa.2022.v09.i01.p08","DOIUrl":"https://doi.org/10.24843/metamorfosa.2022.v09.i01.p08","url":null,"abstract":"Akrilamida merupakan senyawa kontaminan yang ditemukan pada beberapa jenis makanan mengandung karbohidrat dan diolah melalui proses penggorengan atau pemanggangan pada temperatur tinggi. Senyawa akrilamida dapat menyebabkan terjadinya stres oksidatif yang dapat menyebabkan terjadinya neurotoksisitas. Stress oksidatif yang terjadi dapat diatasi dengan pemberian antioksidan salah satunya adalah vitamin C. Penelitian ini dilakukan untuk mengetahui gambaran histologi korteks serebri mencit yang diberikan vitamin C dan diinduksi akrilamida serta dosis yang paling efektif sebagai neuroprotektor pada otak mencit yang diinduksi akrilamida. Penelitian ini menggunakan Rancangan Acak Lengkap (RAL) dengan empat kelompok yaitu diinduksi akrilamida dengan dosis 12,5 mg/kgBB (K+), hanya diberikan larutan normal salin (NaCl 0,9 %) (K-), dan dua perlakuan yang diberikan vitamin C dengan dosis 0,26 mg/ekor/hari (P1) dan 0,52 mg/ekor/hari (P2) serta diinduksi akrilamida dosis 12,5 mg/kgBB. Setiap kelompok dilakukan 6 pengulangan sehingga jumlah sampel yang digunakan berjumlah 24 ekor mencit. Pemberian larutan akrilamida dan vitamin C dilakukan secara oral. Larutan akrilamida diberikan selama 20 hari, sedangkan larutan vitamin C diberikan 3 hari sebelum induksi akrilamida hingga hari ke-20 bersamaan dengan pemberian akrilamida. Mencit akan dibedah pada hari ke-21 untuk diambil organ otaknya kemudian dilakukan preparasi histopatologi dengan metode parafin dan menggunakan pewarnaan Hematoksilin-Eosin. Parameter yang diamati adalah nekrosis pada sel pyramid dan sel glia. Data kualitatif yang didapatkan dinalisis secara statistik dengan One Way ANOVA dan dilanjutkan dengan Duncan. Jenis ini termasuk kategori langka sehingga perlu dilestarikan. Salah satu upaya konservasi yang dapat dilakukan adalah pemilihan komposisi media tanam untuk budidayanya. Penelitian bertujuan mencari media tanam alternatif yang dapat mendukung pertumbuhan kantong semar selain media sphagnum moss. Penelitian dilaksanakan di green house Desa Ngablak, Kecamatan Ngablak, Kabupaten Magelang pada bulan Agustus hingga November 2020. Penelitian menggunakan Rancangan Acak Lengkap (RAL) dengan 4 perlakuan media tanam yaitu sphagnum moss, sekam bakar, sekam bakar dengan cocopeat (3:1), sphagnum moss dengan cocopeat (3:1) masing-masing diberikan 4 ulangan. Parameter penelitian meliputi jumlah daun, waktu kemunculan daun, panjang daun, lebar daun, dan tinggi tanaman diukur pada 12 Minggu Setelah Tanam (MST). Data penelitian dianalisis dengan Analysist of Varian (ANOVA) dan Duncan Multiple Range Test (DMRT) pada tingkat kepercayaan 95%. Hasil penelitian menunjukkan bahwa semua media perlakuan memberikan pengaruh nyata terhadap jumlah daun, panjang daun, lebar daun, dan tinggi tanaman. Media tanam terbaik adalah sphagnum moss dengan waktu kemunculan daun baru (2,39 minggu), jumlah daun terbanyak (11,25 helai), dan tinggi tanaman (1,6 cm). Eksperimen dengan model rancangan acak lengkap digunakan dalam penelitian ini. Hasil penelitian menunjukan bahwa adanya pengaruh lama penyimpanan dengan pemberian perasan jeruk nipis terhadap jumlah angka bakteri pada daging ayam kampung. Kandungan asam organik, asam sitrat, minyak atsiri dan limonene yang terkandung dalam jeruk nipis mempengaruhi total bakteri yang terdapat pada daging ayam kampung. Daging ayam kampung dengan pemberian perasan air jeruk nipis yang disimpan pada suhu <40C hingga 12 hari cenderung mengalami peningkatan jumlah cemaran bakteri. Namun, jumlah angka lempeng total bakteri pada daging ayam kampung dibawah dari aturan yang dikeluarkan oleh Badan Standarisasi Nasional Indonesia mengenai batas mutu tingkat cemaran mikroba yaitu 1 x 106 CFU/g. Ekosistem di kawasan hilir Sungai Unda juga mengalami suksesi antropogenik akibat aktivitas manusia. Proses suksesi tersebut berdampak terhadap spesies tumbuhan penyusun vegetasi di kawasan tersebut. Penelitian ini bertujuan untuk mempelajari komposisi floristik vegetasi di kawasan hilir Sungai Unda. Penelitian dilakukan dengan metode jelajah pada tiga tipe ekosistem, yaitu ekosistem hutan dataran rendah, lahan tergenang, dan pantai. Hasil penelitian menunjukkan bahwa komposisi floristik di kawasan hilir Sungai Unda terdiri dari 163 jenis tumbuhan yang terbagi ke dalam 48 suku dan 128 marga. Bentuk hidup spesies tumbuhan didominasi oleh herba kecil (47 spesies), rumput (35 spesies), dan pohon (31 spesies). Fabaceae adalah suku dengan jumlah spesies terbanyak, diikuti oleh suku Poaceae dan Cyperaceae. Berdasarkan zona penelitian, hutan dataran rendah memiliki jumlah spesies terbanyak (120 spesies), sedangkan zona pantai paling sedikit (82 spesies). The use of genitri is usually as road shade, firewood, seeds for jewelry. The results showed that the diversity of chemical compounds genitri had potential for health. The aim of the study was to analyze the potential of genitri to leukocytes and lung tissue structure of wistar rats induced by influenza virus. The study used an experimental approach, with experimental animals in the form of 24 male wistar rats in 4 treatment groups, P0 negative control, P1 influenza virus induction (positive control), P2 influenza virus induction and given genitri leaf, and P3 influenza virus induction and given genitry seeds. Virus induction on day 0 and day 14. Leaves and seeds were given on the 3rd day. Surgery was performed on day 5 and day 33. The number of leukocytes were calculated using microhematocrit, and identified leukocytes and lung tissue structure staining with HE staining. Ceftriaxone, clarithromycin, dan quercetin terbukti secara in vivo dan in vitro dapat menghambat aktivitas dari pneumolysin. Tujuan dari penelitian ini adalah untuk mengidentifikasi interaksi antara ceftriaxone, clarithromycin, dan quercetin dalam menghambat aktivitas pneumolysin secara in silico. Struktur 3D pneumolysin diunduh melalui RCSB PDB (PDB ID:PQQA). Struktur 3D ceftriaxone (ID:5479530), clarithromycin (ID:84029), dan quercetin (ID:5280343) diunduh melalui PubChem. Preparasi protein dengan Discovery Studio dan ligan dengan OpenBabel. Proses docking menggunakan HEX 8.0.0 dan analisis visualisasi dengan Discovery Studio. Interaksi pneumolysin-ceftriaxone menghasilkan 4 ikatan hidrogen, 5 ikatan hidrofobik, 1 ikatan elektrostatik, dan 2 ikatan unfavourable dengan nilai energi ikatan sebesar -329,98kJ/mol. Interaksi pneumolysin-clarithromycin menghasilkan 3 ikatan hidrogen, 1 ikatan elektrostatik, dan 3 ikatan unfavourable dengan nilai energi ikatan sebesar -311,12kJ/mol.
