The T4/TBG ratio, determined from measurements of total thyroxine (TT4) and thyroxine-binding globulin (TBG) concentrations, is an appropriate index with improved diagnostic accuracy compared with T4. In this study, we calculated the T4/TBG ratio after determining serum TT4 and TBG by radioimmunoassay (INEP, SCG) in euthyroid persons (n=102), in patients with hypothyrosis (n=33) and hyperthyrosis (n=66), as well as in healthy pregnant women (n=346). The reference range for each group of patients was determined. Values obtained for the T4/TBG ratio in healthy subjects varied from 2.1 to 4.4. They were lower in hypothyrosis (0.5 ’2.2) and elevated in hyperthyrosis (4.7–13.1). The reference intervals of the T4/TBG ratio were also determined for each trimester of pregnancy. A significant correlation was obtained between the values for T4/TBG ratio and FT4 concentration, determined by the two-step radioimmunoassay (CIS-bio international, France), in healthy subjects and patients with thyroid disorders (r=0.93), as well as during pregnancy (0.59). TRACP activity was estimated by the kinetic method, while OC and PINP concentrations were measured by the electrochemiluminescent procedure. Statistically significant differences between TRACP activity in patients affected by osteoporosis and both that in patients with osteopenia and the reference values were observed, while no changes in serum OC and PINP levels were recorded. Results suggest that determination of serum TRACP activity could improve the diagnostic potential of osteoporosis. For this purpose, D1-ICL2 and D1-ICL3 were cloned in pGEX-2T vector and expressed in E. coli BL21 as fusion proteins with glutathione-S-transferase (D1-ICL2-GST and D1-ICL3-GST). C-terminal part was cleaved into two fragments which were cloned in pGEX-2T and expressed in E. coli BL21 DE3 as fusion proteins with glutathione-S-transferase (D1-CTSF-GST and D1-CTLF-GST). The resulting soluble constructs were purified by affinity chromatography on glutathione-Sepharose. Ga subunits were expressed and purified as His-tagged proteins (Gao and Gai1 in E.coli BL21 DE3 and Gas in E. coli JM 109). For quantitative assay, varying concentrations of pure His-tagged Ga subunits were immobilized on His-Bind resin and titrated with fusion proteins and the interactions were estimated by a colorimetric assay for GST activity determination. Similar assay was employed to qualitatively demonstrate the interactions. For this purpose pure fusion proteins were immobilized on glutathione-Sepharose in known concentrations and treated with known concentrations of pure His-tagged Ga subunits. Thus created complexes were eluted from glutathione-Sepharose and analyzed by SDS-polyacrylamide gel electrophoresis (SDS-PAGE). It was shown that D1-CTSF interacts specifically with Gas subunit, and D1CTLF with Gao. No other interactions were observed. Standardizacija metoda za određivanje PSA komplikuje se pojavom PSA u serumu u obliku vise izoformi od kojih su najznacajnije forme slobodnog PSA, PSA vezanog za alfa-1-antihimotripsin, koje imaju antigene determinante za koje se vezuju razlicita antitela, kao i PSA vezan za alfa-2-makroglobulin, koji nema antigene determinante, tako da se ova izoforma PSA ne može odrediti pomocu imunoloskih postupaka. Postoji veliki broj imunoloskih metoda za određivanje PSA, koje se međusobno razlikuju po antitelima koja se koriste, po obeleživacu, standardima i po vrednostima PSA koje se dobijaju. Da bi se dizajnirao dobar imunoloski test neophodno je standardizovati kalibratore i odabrati odgovarajuca antitela, pri cemu se mora voditi racuna o ostalim karakteristikama testa kao npr. osobini cvrste faze, kinetici reakcije itd. Posto se nalazi u adenomatoznom, normalnom i kancerogenom tkivu prostate, dijagnosticki znacaj PSA je ogranicen usled preklapanja rezultata kod pacijenata sa karcinomom i benignom hiperplazijom prostate (BPH). Ovo je narocito izraženo za vrednosti PSA koje se nalaze u tzv. \"sivoj zoni\", i to između 4,0 i 10,0 ng/mL PSA. The requirements of the standard are separated in two main groups: management requirements and technical requirements. Management requirements can be implemented in the similar way in many testing laboratories, since they consider elements, common for all business systems. A laboratory should pay special attention to the fulfillment of technical requirements. In addition to the requirements of the standard, within each test area exist other applicable legislation. The protection involves first of all the recognition of the hazards and is implemented by reasonable approach, good behaviour of the personnel, adequate work process in all laboatories and continuing application of good laboratory practice and techniques in the laboatory work. The responsibility for the implementation of safety measures lies primanly on the head of the laboratory i.e. employer but also on the entire personnel. Each clinical-biochemical laboratory must have an official protection programme. Besides there is need of a secure transmission of the test requests from the clients to the central laboratory. We started a project together with other supra-regional laboratories in Germany in order to develop and establish a secure and robust data transmission system for the transmission of clinical data and laboratory information to the central laboratory. Based on common standards for laboratory data transmission and using a two dimensional barcode we developed an offline usable open source standard. Investigation of biochemical parameters such as protein, albumin, cholesterol, billirubin, AST, ALT, as well as haemostasis parameters, including PT, APTT, TT, F II, F V, F VII, AT, plasminogen, could help define the effect of decompression shunt and its possible deleterious effect on the liver function. Investigation included 29 patients with portal hypertension, 19 surgically treated and 10 medication treated patients. F II, F V, F VII, AT, plasminogen and albumin concentration and PT (%) value were statistically significantly lower in medication treated patients in relation with surgically treated patients following one month observation. FVII expressed the highest sensitivity in all investigated parameters. Albumin and billirubin concentrations were in correlation with: PT, F II, F V, F VII and APTT, while transaminases were only in correlation with F VII. ApoE genotypes were determined by restriction isotyping/MADGE analysis. Significant difference in apoE genotype frequencies between case and control group was observed (p<0.01). Patients affected by ischemic stroke had higher frequency of E4 allele and lower E2 allele than age-matched control subjects. Compared with persons without E4 allele, carriers of an E4 allele had 2.1 times higher risk of incident stroke. The aim of the study was to evaluate the importance frequency and treatment efficiency of hyperprolactinaemia in infertile patients. Eighty-seven infertile patients have been examined in this study. Matched samples of periph­eral blood were taken for hormonal analyses in the early follicular, periovulatory and midluteal phases of the cycle. Between the 4th and 7th day of the cycle ultrasonographic and haemodynamic examinations have been carried out at the ovarian and uterine levels. Hyperprolactinaemia was detected in 25 women, in which prolactin levels ranged 628.4-8000 mIU/L. We started dopamine agonists (bromocriptine/cabergoline) treatment individually dosaged. Menstrual cycle as well as prolactin levels restored in all patients 3 months after treatment initiation.
