This event causes the reaction of heart muscle (akinesis and dyskinesis of the ischemic parts of myocardial wall, arrhythmias, and haemodinamic disturbances) and severe systemic reaction (activation of neuroendocrine axis and inflammatory response). Haemostatis disturbances, which can be detected during the AMI and partly caused by the local coronary thrombosis, and partly by the mentioned heart and systemic reaction. A number of therapeutic procedures like venepuncture and almost all drugs commonly used, also influence the measurement of haemostatic parameters. A growing body of evidence has been collected to demonstrate the usefulness of these programs for reducing inter-laboratory variation, analytical errors and for improving the »state-of-the art«. The validity of EQA/PT programs is strongly affected by the quality of control materials, the design of the program, namely the ability to estimate analytical bias and imprecision, and the commitment of providers to assist in the education participant laboratories. The kinetics of metal assimilation and total accumulated quantity depended on the type of treatment. Following metal exposure, an increase in peroxidase activity in the seedlings treated with Cd and Ni/Cd, and in superoxide-dismutase activity during Ni/Cd treatment was observed. A parallel change of peroxidase isoenzyme pattern occurred. There was a good correlation between accumulated metals in needles and enzyme activities. The effect of Cd on the measured parameters was shown to be stronger comparing to the effect of Ni. Bošković","doi":"10.2298/JMH0201041P","DOIUrl":"https://doi.org/10.2298/JMH0201041P","url":null,"abstract":"Lately, in the diagnosis of acute myocardial infarction (MI), special attention is paid to the study of small molecular weight proteins, representing the strucrutal components of cardial muscle controctile proteins, such as cardial troponin T (cTnT). Therefore, the aim of this study was to determine and compare the specificity and sensitivity of cTnT myoglobine and the activity of standard tested enzymes such as: creatine kinase (CK), CK MB isoenzymes and lactic delytrogenase (LDH) in early diagnosis of acute MI. The study concerned the four groups of patients. The first group consisted of healty blood donors (n = 105), the second of patients with verified acute MI (n = 30), the third of a patients with polytrauma (n = 30), and the fourth of patients with cardiovascular diseases with no proved MI (n = 30). For the determination of cTnT level, the commercial Eleccys Troponin T STAT test was used (third generation), Boehringer Mannheim company, based on ECLIA (tehnique of electrochemical luminiscent analysis), designated for the work with automated analysis Elecsys 2010. The concentration of myoglobine (Mi) was determined by immunoturbiometric method, while the activity of other enzymes was determined by the standard IFCC methods with biochemical analyser HITACHI 911, Boehringer Manheim, and with original test reagents. The reference values of blood donor individuals found in the blood ranged from 0.01 to 0.028 ng/mL. In the group of patients with acute MI, biochemical markers (cTnT, CK, CK MB, Mi and LDH) were determined as soon as the material was received by the 24 h service and 4, 8 16, 24, 48, 72, 96, 120, 144, 168, 192 and 216 hours later. In patients with polytrauma the markers were tested 8 hours after surgery and in patients with other cardiovascular deseases with non documented acute MI immediattely after admission, and 4, 8, 16 and 24 hours later. The level of cTnT in patients with proved acute MI was increased at the first testing after admission. The highest values were found at hour 16, gradually declining thereafter, but maintaining above the higher control level in the next 216 hours after admission. In patients with cardiovascular diseases and non documented acute MI the level of cTnT was unchanged. Comparison of results of these two groups of patients showed statistically significant differences in cTnT levels in patients with acute MI during all tested time intervals. In the group of patients with polytrauma only the value of cTnT was within the normal level. Diagnostic precision of tested cardiac markers was also tested by ROC analysis. According to the data obtained, only cTnT exhibited statistically significiant diagnostic precision immediately after the admission of patients with the following calculated ROC AUC: 0.855 for cTnT, 0.716 for CK, 0.503 for CK MB and 0.552 for LDH, respectively. Pulmonary surfactant is synthesized in alveolar type-II granular pneumocytes and »packed« as lamellar bodies. Before delivery, the lamellar bodies diffuse through the bronchial tree into the amniotic fluid. Parameters which indicate the maturity of lungs, except the concentration of lamellar bodies, also are observed in the ratio against the quantity of surfactant with other components of the amniotic fluid (sphingomyelin and albumin). The study concerns the determination of the surfactant-to-albumin ratio and the lamellar body count in the amniotic fluid as predictors of fetal lung maturity. The research involved 90 pregnant women with gestational age from 32 to 40 weeks. The samples of the amniotic fluid were obtained by amniocentesis and they were filtrated through special filters. The surfactant-to-albumin ratio was measured by the Fetal Lung Maturity II (FLM) test, on a TDX instrument (ABBOTT). The concentration of lamellar body count was measured on a hematology cell counter by the platelet channel, on the Cell Dyn 3700 instrument (ABBOTT). The average value of S/A ratio was 55.11 mg/g (min=6.96 mg/g, max=112.00 mg/g). The surfactant-to-albumin ratio of less than 39.00 mg/g predicts respiratory distress syndrome with the probability of 92%. The average value of LB was 69.4 × 109/L (min=4.2 × 109/L, max=199.0 × 109/L). Lamellar body concentration of 34.0 × 109/L or less predicts respiratory distress syndrome with the probability of 93%. The surfactant to albumin ratio increased with gestation (r=0.373, p<0.005), as did lamellar body concentration (r=0.934, p<0.001). The two tests correlated with each other, r=0.341, p<0.005. In 90 patients delivered within 72 hours the surfactant-to-albumin ratio and concentration of the lamellar body correctly predicted six cases of the respiratory distress syndrome. Male Mill Hill hybrid hooded rats aged 15, 45 and 75 days were treated with L-thyroxine, T4 (40 mg/100 g body mass), s.c., one dose per day, 14 days (finally aged 30, 60 and 90 days, respectively). Effect of T4 on GSH-dependent antioxidant enzyme activities in the IBAT differs with respect to age. T4 treatment gradually decrease activities of all GSH-dependent antioxidant enzymes in 60 and 90 days old rats in comparison to young ones. GSH content in animals of 30 and 60 days old rats are lower in comparison with 90 days old rats, but the effects are oppposite. L-thyroxine treatment significantly increase GSH content in 30 days old rats (p<0.001) in respect with coresponding controls, while decrease in 60 and 90 days old animals were detected (p<0.01). Mean ejaculate volume was 3.3 mL (range 1-6 mL). Viscosity was increased moderately in six samples and greatly in four more. The number of motile sperm (0-165 million/mL; mean 49; median 39) was highly correlated with total number (P < 0.001). Concentrations of IGF-I and -II determined by displacement radioimmunoassay were positively correlated (P = 0.001). IGF-I ranged from 2.7 to 29.7 nmol/L (mean 9.6; median 6.5) and increased with viscosity. IGF-II ranged from 5.7 to 39.0 nmol/L (mean 16.0; median 13.5). IGFBP-2 and putative IGFBP-4 were detected by autoradiography of ligand blots after polyacrylamide gel electrophoresis. These binding proteins were stable in ejaculates stored at -18 °C with protease inhibitors and remained associated with higher molecular weight complexes during molecular size chromatography with 1 mol/L acetic acid. They eluted at least partially in the expected fractions with 1 mol/L sodium chloride but further attempts at purification were unsuccessful due to progressive degradation. The animals were sacrificed by decapitation 24 h after the end of the diet. Triglycerides, total cholesterol, and LDL cholesterol plasma levels, as well as LDL/HDL ratio, and vitamin E plasma level significantly raised (p 0,05). This ubiquitous mechanism enables functioning of general but also specific cell-tissue processes, involved in maintaining of cell homeostasis. Normal transduction processes disturbance represents the most frequent molecular basis in pathogenesis and complications of many diseases. Understanding of all sequences involved in signalling pathway cascades offers a possibility of different modulators application in therapeutic purposes. The activation of phagocyte NADPH oxidase is one of the best studied pathways of signal transduction. Meanwhile, reactive oxygen species generated in the process could modulate numerous cell transduction pathways. Diabetes mellitus may be associated with increase in oxidative stress, which may contribute to a long-term tissue damage. In addition to damage, oxidative stress can initiate cell signalling cascades that modulate cell function and finally could lead to the cell death. Selakovic","doi":"10.2298/JMH0203269M","DOIUrl":"https://doi.org/10.2298/JMH0203269M","url":null,"abstract":"In the present study we employed Nw-nitro-L-arginine methyl ester, non-specific potent nitric oxide synthase inhibitor and a selective inhibitor of neuronal nitric oxide synthase, 7-nitroindazole, reportedly to investigate the possible involvement of nitric oxide in quinolinic acid-induced striatal toxicity in the rat. Quinolinic acid was administered unilaterally into striatum of adult Wistar rats in the single dose of 150 nmol/L. The other two group of animals were pretreated with Nw-nitro-L-arginine methyl ester and 7-nitroindazole respectively. Control groups of animals were treated with 0,154 mmol/L saline solution likewise. Nitrite levels was decreased in the ipsi- and contralateral striatum and forebrain cortex in the group treated with nitric oxide synthase inhibitors and neurotoxin compared to quinolinic acid-treated animals. In the same structures, activity of glucose-6-phosphate dehydrogenase was also decreased, compared to quinolinic acid-treated animals.
