As a result of the study, the efficient sterilization protocol of alfalfa seeds was established with the use of sterilizing agents as 70% ethanol and 10% sodium hypochlorite which resulted in the production of 100% aseptically viable seedlings for the further in vitro studies. Callus initiation from cotyledons and hypocotyls was implemented on a Gamborg B5 nutrient medium containing 2,4-dichlorophenoacetic acid (2,4-D), kinetin in a concentration of 5,0 mg/l, and naphthaleneacetic acid (NAA) of 0,1 mg/l. Induction of morphogenic callus was achieved on a Gamborg B5 medium with 0,2 mg/l 6-benzylaminopurine (BAP). Further shoot initiation occurred on a hormone-free Gamborg B5 medium with the addition of Gamborg B5 vitamins, and on a half-strength medium with the addition of 0,2mg/l kinetin. It was found that established method of obtaining regenerated plants from the callus cultures of Medicago Varia proved itself effective.","PeriodicalId":248021,"journal":{"name":"BULLETIN of the L.N. Wheat is a grain which the most important source of food on earth. It contains 75-80% carbohydrates, 9-18% protein, fiber, many vitamins (especially B vitamins), calcium, iron, and many macro-and micro-nutrients. Fungal leaf diseases, such as leaf blotch caused by Septoria tritici and rust diseases caused by Puccinia graminis, Puccinia striiformis and Puccinia triticina are also a problem for spring wheat production. Despite the dry climate, the cultivation of susceptible varieties results in epidemics of leaf rust in one year out of four on average, affecting more than 1 million hectares, with estimated losses of up to 25-30 percent of yields. Salicylic acid and oxalic acids have the effects of promoting plant growth. We test whether they can positively impact wheat yield under disease conditions. Foliar seed application of Salicylic acid and oxalic acid on wheat cultivar leads to overall better performance of the plants and increases the yield significantly. Effect on wheat yield components of two substances (SA, and OA) in both ways such as seed treatment and foliar spray, believed to have growth-stimulating properties in plants. 0.2 mM OA and 0,5 mM SA+ 0,1 mM OA samples represented a good result in both seed treatment and foliar spray in all yield characteristics of Aray cultivars compaire to control. The results of this study will be useful to control fungal diseases of wheat.","PeriodicalId":248021,"journal":{"name":"BULLETIN of the L.N. Gumilyov Eurasian National University. BoNT has received particular use as a long-acting analgesic due to its ability to inhibit the exocytosis of pain neuropeptides, including CGRP, Substance P, and modulate the expression of pain-related receptors as TRPV1, NMDA, AMPA, P2X3. Botulinum toxin injections are effective in the treatment of many pain conditions: diabetic neuropathy, chronic migraine, spinal cord injury. This article reviews the possibilities of clinical applications of botulinum toxin based on the analysis of articles in the PubMed database for specific search terms. The keywords “botulinum toxin”, “treatment”, “clinical use”, “pain reduction” were used to obtain references. The possibilities for the clinical use of botulinum toxin are growing every year. Currently, at least 8 commercially available preparations of botulinum toxin types A and B are known, the most recognizable of which are Botox, Dysport, and MyoBloc/NeuroBloc. Future research should focus on improving the safety profile and increasing the efficacy of botulinum toxin-based drugs. New types of botulinum toxin obtained through molecular reengineering will significantly increase its therapeutic efficacy.","PeriodicalId":248021,"journal":{"name":"BULLETIN of the L.N. Molecular markers of papillary thyroid carcinoma (PTC) including the BRAFT1799A mutation, RET/PTC1,3 rearrangements, and differentially expressed SFTPB (up-regulated) and TFF3 (down-regulated) genes were prospectively assessed in FNAB (fine needle aspiration biopsy) material from thyroid nodules. We examined 73 cases, of them 59 PTC (papillary thyroid carcinoma), five nodular goiters, and nine follicular adenomas. Concordantly with cytology, molecular diagnosis of PTC by SFTPB and TFF3 expression levels was confirmed in 38/41 cases (92.7%); in them, 22/41 (53.6%) had BRAFT1799A mutation and 7/41 (17.1%) had RET/PTC rearrangements. For benign nodules, concordance was observed in 10/12 (83.3%) cases. Among 20 cases, including 19 suspicious for malignancy and one inadequate sample, 17 were positive for SFTPB/TFF3 test (85.0%). Of these, four were also positive for RET/PTC rearrangements, five were positive for BRAFT1799A and all resulted in PTC at histology. The remaining 3 cases were negative for three molecular markers. These nodules showed benign lesions in histology. Our results demonstrated that specific molecular tests improve the efficacy of preoperative diagnostics of PTC.","PeriodicalId":248021,"journal":{"name":"BULLETIN of the L.N. The authors studied the number of populations, sex, and age belonging of small mammals. Gero crushers and trap grooves with cones were used on the technogenic territory near the Pavlodar aluminum and Kazakhstan electrolysis plants. 173 animals were obtained: 104 in the background areas and 69 in technogenic territories. The animals belong to 15 species of two orders: Rodents and Insectivores. As a result of the research, a decrease in species richness (<0.695) and species diversity (<0.228) was noted, the dominance of Microtus gregalis (42) and Sicista subtilis (26.1) in areas subject to anthropogenic load was noted. The number of males participating in inbreeding in the territories near factories increases in the steppe mouse and narrow-crusted vole (67%), compared with the control (33%). At the control sites, there were fewer mature females (42%) than at the technogenic ones (58%). The fecundity of females (the number of embryos) is lower in the control area (±5.28) compared to the technogenic zone (±6.1).","PeriodicalId":248021,"journal":{"name":"BULLETIN of the L.N. The capsid protein (СР) P41 is required for virion formation and facilitates long distance movement of the virus. Along with RNAi suppression, P19 protein is involved in the development of severe disease symptoms in N. benthamiana and elicitation of Hypersensitive Response (HR) in tobacco. Our results show that wild type TBSV infection of Solanum lycopersicum (cv. Money maker) triggers resistance to the virus. Despite detectable accumulation levels of P19 protein in leaf and root tissues, the infection was not accompanied with obvious disease symptoms. Contrastingly, inoculation with TBSV mutant, lacking capsid protein P41 demonstrated susceptibility to TBSV. Moreover, Chl-FI analysis of plants infected with virus exhibited significant changes in metabolism. Our data suggests that in response to CP expression tomato plants have evolved defense mechanisms to resist viral infection.","PeriodicalId":248021,"journal":{"name":"BULLETIN of the L.N. In this case, the multiplicity of functions is not due to gene fusion or multiple proteolytic fragments. Nowadays, there have been identified several hundreds of proteins of this class, including proteins with a wide variety of functions. Some of them are involved in a cascade of biochemical reactions, using the active site for catalysis, and another part of the protein surface to interact with other proteins. Many of them are involved in the development of pathological conditions; therefore, data on the structural and functional organization and mechanisms of changes in the localization of such proteins may be in great demand in the field of prevention and treatment of various pathologies.","PeriodicalId":248021,"journal":{"name":"BULLETIN of the L.N.
