S. burahol leaves contain phenolics, flavonoids, and terpenoids which can interfere with DNA isolation. The use of direct PCR kits can detect genes without DNA extraction.The objective of study was to determinethe method of gene detection ofStelechocarpus buraholusing directPCRkit.In each location, one tree was taken as a source of leaf samples from Garut, Purwodadi Botanical Gardens, Kyai Langgeng Gardens, Yogyakarta Palace, Turi Sleman, Wanagama, Karanganyar, and South Kalimantan, except Bogor Botanical Gardens, two trees were taken. The primers used for the trials were ITS 1F primers and 4R primers. In the sequencing stage, PCR product samples of 40 -50 μl that showed positive results were detected by electrophoresis. The PCR product was measuredat ± 750 bp from ten samples. Direct PCR kits can be used for S. buraholgene detection, time and energy efficient, only requires a small amount of tissue, and reduces contamination due to DNA extraction. Result of population selection level in Java Island have been used to build Provenan Seed Stand (PSS) nyamplung in KHDTK Wonogiri (Central Java) as an improved seed source with high oil yield and fruit productivity. Seed become nyamplung part that is used as oil raw material which is strongly influenced by flowering and fruiting period. Information on flowering fruiting ability and period is needed to support the availability of raw materials in a sustainable supply for the nyamplung oil industry. Information on flowering fruiting ability and period is needed to support the availability of raw materials in a sustainable supply for the nyamplung oil industry. Observation of flowering and fruiting period focused on 465 individual trees in nyamplung PSS at KHDTK Wonogiri through monthly monitoring by calculated flowering fruiting individual tree during 5 years (2014-2018). The results showed that the flowering period of nyamplung in TBP Wonogiri varied during 2014-2018. Flowering and fruiting occurs almost throughout the year. However, the peak of flowering and fruiting occurs mostly in March-August (end of the rainy season to the middle of dry season). In general, the trend of flowering and fertilization increased in 2014-2016, then decreased in 2017-2018, which is thought to be influenced by the higher canopy density. The environmental factor that most influences flowering is number of rainy days, while fertilization of TBP was strongly influenced by temperature. In order to maintain the superiority of this characteristics, a proper vegetative propagation techniques is required to multiply selected trees.The purpose of this study is to determine the effect  of cutting materials from sprouts on seedlings and mother trees. The study was arranged in a completely randomized design with factorial pattern. The factors tested were cutting materials (namely sprouts on seedlings and on the mother tree branches) and application of growth hormone types (control, pasta and  powder). The results showed that the survival rates and the growth of cuttings from seedlings and mother trees  were relatively similar. However, the rooting percentages and the growth of cuttings were significantly influenced by the interaction between the two factors. The survival rates of shoot cuttings varied from 51.92 to 82.30% and rooting percentages range from 32 to 70%. Ex situ conservation need to be carried out to protect them from extinction. Preparation genetic materials in nursery (seedlings) in term of seedlings was very important to support success of establishment of ex situ conservation plot. This research has objective to know the role of genetic factor to growth caharacteritics of kayu kuku seedlings from South East Sulawesi provenance. This research was arranged in Completely Randomized Complete Bock Design with 40 families as a treatment and 3 replications. The result showed that genetic diversity was observed affecting the growth characteristics such as height, diameter and sturdiness index among families. Estimation of family heritability (h 2 f) for height, diameter and sturdiness index were 0,94; 0,91 dan 0,91 respectively. Genetic correlation between height and diameter was found high positive namely 0,95. Forster) merupakan salah satu jenis tanaman MPTS yang memiliki potensi untuk dikembangkan dalam program rehabilatsi hutan dan lahan di daerah Sulawesi Utara. Terbatasnya informasi mengenai teknik budidaya jenis matoa sering menjadi kendala, utamanya tentang teknik penyimpanan benih yang sesuai jika benih tidak langsung disemai. Penelitian ini bertujuan untuk mengetahui pengaruh waktu dan media simpan terhadap viabiltas benih matoa. Rancangan yang digunakan dalam penelitian ini adalah acak lengkap yang disusun dengan pola faktorial, terdiri dari 2 (dua) faktor, yaitu: 1) waktu simpan dan 2) media simpan. Waktu simpan terdiri dari 3 faktor, yaitu 5 hari, 10 hari dan 15 hari,  sedangkan media simpan terdiri dari 4 faktor yaitu alumuniu foil, kain blacu, cocopeat dan tanpa media simpan. This test involved  identification of the fungal species that inhabited the experimental plant after inoculation of BCA and  pathogen. Identification of inhabitant fungi in the BCA control activity test can be conducted by culturing the fungi on artificial media. Fungal species can be identified based on morphological characters or genetic  characters of fungal culture. This study was conducted to determine the potential of PCR ITS-RFLP molecular  markers in the initial selection process of fungal isolates before identification based on DNA sequences and to  select the enzyme that produce polymorphic PCR ITS RFLP pattern. Only a few know that kayu merah has two subspecies (forma) that are differentiated based on the surface structure of the central seed case. This study aims to know the differences on morphological and growth characters of two populations/subspecies kayu merah i.e. Pulau Seram, Mollucas (smooth pods, forma indicus) and Pulau Flores, East Nusa Tenggara (prickly pods, forma echinatus). The genetic relationship of these two populations/subspecies was analyzed using PBSTAT program, while their growth parameter analyzed using SAS program. The study found differences on morphological and growth characters of kayu merah from  Pulau Seram and Pulau Flores. Kayu merah from Pulau Seram has many smooth spots and little grooves on the surface of its stem and has tapered leaf tip shape, while kayu merah from Pulau Flores has many grooves and little smooth spots, and has splitting leaf tip form. The two populations/subspecies were clearly clustered into their own group. Kayu merah from Pulau Seram has better growth than that of Pulau Flores. Leaf samples were collected for DNA templates and 6 SSR primers were used. Genetic data of 280 trees were analyzed by FSTAT software , while PCoA was analyzed by GeneAlex. Results showed that number of allele (N A ) per SSR primer between 3 and 4, the allelic richness (A R ) ranged 2.805 to 4.000, the gene diversity (H E ) ranged 0.018 to 0.631 , coefficient inbreeding (F IS ) of overall SSR primers were insignificant deviate from Hardy-Weinberg Equilibrium, excepted NY3. The mean H E values in the TBP was in low (mean H E = 0.272). However, the mean coefficient inbreeding value s was not significant. PCoA analysis showed that the seed stand consisted genetically related trees; the 280 trees originated from only 5-6 mother trees. This is because the stand was originated from a conservation stand with narrow genetic di fferentiation . Shorea tengkawang produces tengkawang oil which usually used as a raw material and supporting material for the manufacture of food, pharmaceuticals and cosmetics. This study determines the genetic relationship of S. gysbertsiana to another three Shorea species uses RAPD (Random Amplified Polymorphism DNA) specific loci. There were 13 specific alleles obtained from 6 RAPD primers. Among these specific alleles, there were 8 loci shared between S. gysbertsiana and S. macrophylla, and 2 loci shared with S. stenoptera.  There were no specific loci shared between S. gysbertsiana and S. pinanga. This study found one specific locus for S. gysbertsiana and one specific locus between S. stenoptera and S. pinanga. Based on global industrial development, the forest plantations industry will increase in the following years. Eucalyptus pellita has become main species in Indonesia forest plantations because it has a short cycle and wood products are suitable to forest industry. The average productivity of E. pellita plantations in Indonesia is still low and high variation. In an effort to increase the productivity, the first step is a better understanding of genetic control on growth and basic density. This study aims to determine the genetic parameters for growth and basic density of E. pellita clones on two different sites. The clonal trials was conducted at two different sites. Both clonal trial are designed using RCBD. The number of clones tested in both trial was 30, 5 blocks and 25 tree/plot. The result of the study showed that the effects of clones vary greatly according the enviromental conditions. The clones- environemntal interaction of growth trait is higher than the basic density. This is in line with genetic parameters of growth trait that are less stable than the basic density. The expected genetic gain of growth trait is higher than the basic density and at the same time there was a weak genetic correlation (there is even a negative) between growth trait and basic density.
