Latyshko","doi":"10.15407/ubj94.03.026","DOIUrl":"https://doi.org/10.15407/ubj94.03.026","url":null,"abstract":"Bronchial asthma is developed as an immune response to allergen challenges accompanied by inflammation and fibrosis implicated in airway remodeling. To reveal the causative implication of Cu-containing amino oxidases semicarbazide-sensitive amine oxidase (SSAO), DAO and lysyl oxidase (LOX) in BA development we used their irreversible inhibitor semicarbazide and guinea pig model of BA induced by ovalbumin. Semicarbazide was introduced to asthmatic animals via drink or inhalation. At the 16th week after disease induction, the increase in the activity of pro-inflammatory SSAO and DAO in plasma (1.6 and 2 times, respectively) was observed. The introduction of semicarbazide to asthmatic animals via drink or inhalation significantly decreased activities of these enzymes compared to the untreated asthmatic animals. A considerable increase in IL-13 content and LOX activity in the lung tissue of asthmatic animals were observed that evidenced airway inflammation and pulmonary fibrosis development. The uptake of semicarbazide by guinea pigs with bronchial asthma led to normalization of LOX activity. Histological studies confirmed that semicarbazide attenuated morphopathological changes in the lungs of asthmatic animals. Thus, the data obtained indicate the direct participation of the studied enzymes in the progression of pathological processes in atopic bronchial asthma as well as the potential use of semicarbazide as a drug in complex anti-asthmatic therapy. This study aimed to investigate the effect of ferric oxide (Fe 2 o 3 ) nanoparticles administration on isoniazid-induced oxidative stress parameters in rat brain tissue. Forty adult male Wistar rats (200–250 g) were randomly divided into a group with no treatment as control and four experimental groups. Animals of experimental groups received intraperitoneally for 12 days daily saline, 50 mg/kg of isoniazid, 50 mg/kg of isoniazid and 0.2 or 0.4 mg/kg Fe 2 o 3 nanoparticles accordingly. The activity of catalase (CAT), superoxide dismutase (SOD), glutathione-S-transferase (GST), the level of glutathione (gSH), malondialdehyde (mda) and total protein were determined in brain tissue homogenates by spectrophotometric methods. It was shown that Cat and gSt activities, as well as gSH and mda levels in the brain tissue of animals in the isoniazid-treated group were increased compared with the control untreated group, while following the treatment with 0.2 or 0.4 mg/kg Fe 2 o 3 nanoparticles the studied oxidative stress parameters returned to the control level (P < 0.05). No changes in Sod activity in any of the treated groups were observed compared to the control. {"title":"Correlation between adiponectin level and obesity as a risk factor for allergy disease","authors":"M. Spasovska, T. The aim of this study was to determine the association of adiponectin with obesity, as a risk factor for the development of allergic condition in order to rationalize approach to its treatment. Research methods for inflammatory markers and biochemical parameters involve immunoassay technique. Statistical analysis was performed with Student’s t-test, Wilcoxon T-test and coefficient of correlation. The study included apparently healthy subjects and patients with allergy conditions with confirmed presence of specific IgE, classified into 2 groups according to their body mass index (BMI). The obtained data showed negative correlation (cor = – 0.6), between adiponectin levels and BMI values. Thus, decreased level of adiponectin is associated with increased BMI. The mean values of adiponectin in the studied population, with high statistical differences between the groups (19.1 ± 1.5; 17.7 ± 0.9), (18.8± 1.1; 16.6 ± 1.0) demonstrated the relationship between low adiponectin level and development of obesity, and what, in turn, increasd risk of developing allergic conditions. The assumption was made that adiponectin may be used as a sensitive biochemical marker for early diagnostics of allergic reactions. Pokhodylo","doi":"10.15407/ubj94.03.068","DOIUrl":"https://doi.org/10.15407/ubj94.03.068","url":null,"abstract":"Cage amides and imides bearing bicyclo[2.2.1]- and bicyclo[2.2.2]-subunits were synthesized and evaluated both for antimicrobial activity toward five key ESKAPE pathogenic bacteria: one Gram‐positive bacteria methicillin‐resistant Staphylococcus aureus (ATCC 43300), four Gram‐negative bacteria Escherichia coli (ATCC 25922), Klebsiella pneumoniae (ATCC 700603), Acinetobacter baumannii (ATCC 19606) and Pseudomonas aeruginosa (ATCC 27853) and for antifungal activity towards pathogenic fungal strains Candida albicans (ATCC 90028) and Cryptococcus neoformans var. Grubii (H99; ATCC 208821). Compound VP-4539 with bicyclo[2.2.2]octene motif demonstrated the highest cytotoxic activity towards C. neoformans, while human keratinocytes of HaCaT line, murine fibroblasts of Balb/c 3T3 line and mitogen-activated lymphocytes of peripheral human blood were found to be tolerant to its action. VP-4539 compound did not intercalate into salmon sperm DNA indicating that its cytotoxicity is not related to intercalation into nucleic acid. The viral activity was estimated with the use of inverted microscope PrimoVert (Germany) by destructive effect on BHK21 fibroblastic cell line. In vitro results demonstrated that decamethoxin (100 μg/ml) completely inactivated IBV coronavirus strain at exposure of 30 sec and more. At the lowest decamethoxin exposure of 10 sec the antiseptic virucidal activity was 33% and 36% of control at 24 and 48 h of cultivation respectively. Molecular docking analysis indicated the significant similarity of IBV and SARS-CoV-2 main protease (Mpro) structure. Docking studies of decamethoxin interaction with IBV Mpro and SARS-CoV-2 Mpro active centers demonstrated the ligand-protein complexes formation with the estimated binding energy of -8.6, -8.4 kcal/mol and key amino acid residues ASN26, GLY141, GLU187, GLU164, THR24, THR25, ASN142, GLY143, CYS145, HIS164 and GLU166. The aim of this work was to obtain solid lipid nanoparticles (SLN) from surface lipids (waxes) of Chelidonium majus L. (Papaveraceae) leaves and to estimate whether capsulation of dirhenium(III) as a component of the Re-Pt system into SLN will affect its antitumor activity and red blood cells (RBC) morphology in a rat model of Guerin’s carcinoma growth. Fourier-transform infrared spectroscopy, gas-liquid chromatography, microscopy, light scattering were used in the research. Solid lipid nanoparticles were obtained, characterized, loaded with cluster dirhenium(III) and being introduced together with cisplatin to rats with Guerin’s carcinoma resulted in RBC morphology preservation and a significant decrease in tumor weight. It was concluded that the lipid coating of the rhenium cluster compound did not reduce the antitumor effect of the Re-Pt system and protected RBC from toxic cisplatin influence. A new formulation of the Re-Pt system is proposed. {"title":"Dynamics of homocysteine level in patients with osteoporotic fracture","authors":"N. Hasanova","doi":"10.15407/ubj94.03.053","DOIUrl":"https://doi.org/10.15407/ubj94.03.053","url":null,"abstract":"The research was carried out in order to investigate the blood serum level of homocysteine (HCY) which is involved in bone metabolism and has prognostic significance in the monitoring of the regenerative processes in osteoporosis and osteoporotic fractures. The study was carried out on patients 45-83 years old divided into 3 groups: group I – 14 patients with osteoporosis confirmed by densitometry or X-ray examination, group II – 15 patients with non-osteoporosis fractures, group III – 25 patients with osteoporotic fractures. The control group consisted of practically healthy 14 people. In patients with various fractures osteosynthesis with Ilizarov apparatus or with metal plates was performed. After the operation, the patients were treated in an inpatient setting for a week, then sent for outpatient treatment and prescribed calcium and vitamin D supplements to accelerate the bone regeneration process. A blood sample was taken at 3 stages to monitor the dynamics of HCY level by Elisa test: on the 1st day before treatment, on the 10th day of treatment and 1 month after it. The results showed that on the 1st day before the treatment HCY concentration was statistically increased 2.7 times in group I, 5.6 times in group II, and 6.5 times in group III compared to the control group. In the month of recovery, a significant decrease in HCY level was observed in all treated groups but it still remained higher than in the control indicating the need to recommend additional therapeutic prescriptions. This 76 kDa protein was identified with mass-spectrometry as human serum albumin. Such molecular form of albumin was absent in blood serum of healthy human donors. The potential ways of generation of the unusual form of human serum albumin and its probable diagnostic value were discussed. Veliky","doi":"10.15407/ubj94.03.005","DOIUrl":"https://doi.org/10.15407/ubj94.03.005","url":null,"abstract":"Osteoporosis is a progressive systemic skeletal disease characterized by a decrease in bone density, impairment of its microarchitectonics, and an increased risk of fractures that occur under minimal or no mechanical stress. One of the main causes of osteoporosis is vitamin D deficiency, which leads to disruption of normal bone remodeling. The aim of our study was to analyze the features of the process of bone tissue remodeling by measuring the key biochemical markers of bone formation/resorption in primary and secondary osteoporosis, as well as to investigate the potential corrective effect of vitamin D3 supplementation. The work was conducted on rats with different osteoporosis models: alimentary, dysfunctional and secondary osteoporosis associated with diabetes mellitus. We used ELISA to measure 25(OH)D content in blood serum. Blood serum and bone tissue calcium, and alkaline phosphatase activity were determined with bioassay kits. The content of inorganic phosphate in blood serum and ash was assayed by the Dyce method. It was shown that all the studied pathological conditions were accompanied by vitamin D deficiency, which led to impaired absorption of calcium in the intestine and reabsorption of inorganic phosphates by the kidneys, reducing, as a result, their concentration in the blood serum. Hypocalcemia and hypophosphatemia contributed to the disruption of normal bone remodeling, excessive activation of alkaline phosphatase, and a decrease in the content of calcium and phosphate in bone tissue. Thus, sufficient vitamin D bioavailability was confirmed to be critical for effective bone remodeling in primary and secondary osteoporosis. {"title":"The level pro-inflammatory and anti-inflammatory biomarkers in patients with chronic mechanical low back pain under pulse radiofrequency therapy","authors":"I. Tekin, F. Conventional treatment methods are generally insufficient in the treatment of chronic low back pain. The aim of the study was to estimate the level of IFN, IL-1, IL-6 (proinflammatory), IL-10, IL-4 (anti-inflammatory) and VEGF proteins in the serum of patients with chronic mechanical low back pain under Pulse radiofrequency (PRF) therapy. The study was carried out on 40 patients 20-60 years old, diagnosed with chronic low back pain for at least 4 months, primary complaint on lumbosacral low back pain, pain intensity VAS (visual analog scale) score of 5 and above, not responding well to conservative treatment (analgesic drugs, physiotherapy, etc.). Therapeutic Radiofrequency applications were carried out with an RF generator (RFG 3C Plus, Radionics). Blood samples were taken 1 day before interventional treatment (control), then 1 day (group1) and 15 days (group 2) after. The serum level of IFN, IL-1, IL-6, IL-10, IL-4 and VEGF l was analyzed with ELISA test. It was shown that as a result of PRF treatment the level of IL-1 was decreased while the levels of IL-4 and IL-6 were increased. It was concluded that the increase in serum levels of proinflammatory cytokines may be correlated with the severity of pain and that the increase in the level of anti-inflammatory cytokines reduces pain by reducing inflammation.
